Abstract

Sambiloto (Andrographis paniculata) has been known empirically as a plant that has benefits as a traditional antidiabetic Javanese medicine to treat diabetes mellitus (DM). The main compound contained in sambiloto is andrographolide which has a hypoglycemia effect. Andrographolide has low solubility in water and poor bioavailability due to biotransformation by P-glycoprotein in the intestinal tract which causes andrographolide to metabolize more quickly in the duodenum and jejunum. To overcome this extract ethanol of sambiloto (EES) formulated to patch transdermal. Enhancers can affect the characteristics of transdermal patch preparations which affect the pharmacological activity of transdermal patch preparations. The critical point of transdermal patch preparation is the selection of enhancers which is one of the compounds to increase the penetration of active substances in the skin by disrupting the permeability of the stratum corneum. The aim of this research is to determine the effect of variations in the combination of oleic acid and tween 80 as an enhancer on the physical characteristics of EES patches.The methods of extraction sambiloto use soxhetation with ethanol 96% as solvent. The formulation of EES patch transdermal using solvent casting technique with various concentration enhancer tween-80 and oleic acid. This research was conducted by making 3 formulas, where F1  (1:1), F2 (1:3) and F3 (1:3). The evaluations include weight uniformity, thickness, water content, pH and folding endurance of the patch. The result of visual appearance the patch EES has a dark green with aroma specific of EES. The characteristics of EES transdermal patches were affected by the ratio of tween-80 and oleic acid as the enhancers. The combination of oleic acid and tween 80 as a permeation enhancer has an influence on physical characteristics (weight and thickness) of EES patches.

Abstract

Kalangkala merupakan tumbuhan khas Kalimantan yang termasuk dalam genus litsea pada bagian biji tanaman kalangkala (Litsea angulata) memiliki  aktivitas antioksidan senyawa yang berperan sebagai antioksidan pada biji Kalangkala (Litsea angulata) adalah kuersetin. Kuersetin memiliki  aktivitas antioksidan melalui penghambatan jalur stres oksidatif yang berhubungan erat dengan pembentukan tirosinase yang menyebabkan hiperpigmentasi pada kulit. Tujuan dari penelitian ini adalah untuk mengetahui potensi Kalangkala (Litsea angulata) sebagai agen inhibitor tirosinase baru dari bahan alam secara pendekatan molekuler docking. Pada tahap penelitian ini persiapan yang dilakukan meliputi persiapan reseptor dan native ligand, validasi metode docking, preparasi senyawa kuersetin, dan molekuler docking senyawa kuersetin terhadap protein uji enzim tirosinase. Hasil yang diperoleh dari docking molekuler antara Kuersetin biji Kalangkala (Litsea angulata) dan  protein protein tirosinase memiliki nilai afinitas binding (ΔG) dan konstanta inhibisi (IC) sebesar -5.20 kcal/mol dan 154.02 µM. Interaksi ikatan hidrogen yang terbentuk pada senyawa kuersetin menunjukkan bahwa senyawa tersebut afinitas yang nilainya lebih rendah dibandingkan native ligand sehingga berpotensi sebagai agen inhibitor tirosinase baru.

Abstract

Kanker merupakan pertumbuhan sel yang tidak normal yang mana sel tersebut bisa tumbuh dan menyebar ke bagian tubuh lainnya bahkan menyebabkan kematian. Benzoxazole termasuk dalam kerangka senyawa yang sangat aktif secara biologis. Berbagai turunan benzoxazole yang diteliti secara ekstensif dapat memberikan aktivitas anti bakteri, anti jamur dan aktivitas anti kanker. Penelitian ini bertujuan untuk mengetahui bagaimana hubungan kuantitatif struktur aktivitas (HKSA) dari senyawa turunan benzoxazole serta membangun suatu persamaan berdasarkan HKSA. Sebanyak 14 struktur senyawa turunan benzoxazole yang memiliki aktivitas sebagai anti kanker yang akan digunakan pada penelitian ini. Pemilihan beberapa prediktor yang digunakan didasarkan atas seberapa besar pengaruh nilai dari beberapa prediktor yang mewakili parameter hidrofobik, elektronik, dan sterik yang berfungsi sebagai variabel bebas terhadap aktivitas senyawa berupa logIC50 yang berfungsi sebagai variable terikat yang sebelumnya sudah dilakukan optimasi geometri dan perhitungan energi total. Untuk pengembangan model HKSA, 14 senyawa tersebut dibagi menjadi training set sebanyak 11 senyawa dan test set sebanyak 3 senyawa. Model persamaan HKSA terbaik ditentukan dari hasil analisis HKSA dengan parameter hidrofobik, elektronik dan sterik. Model persamaan yang dapat memprediksi nilai aktivitas biologis (log IC50) yang paling mendekati nilai aktivitas biologis (log IC50) eksperimen merupakan model persamaan HKSA yang terbaik. Dari analisis hasil SPSS regresi linear dengan metode enter diperoleh suatu persamaan HKSA Log IC50 = -9.424 + (-0.963*HOMO) + (0.256*ClogP) + (0.487*logS) + (0.200*Dipole) + (0.009*Mw) dengan nilai R = 0.984, R2 = 0.969, Fhitung = 31.275, Ftabel = 5.050, Fh/Ft = 6.192 > 1. Persamaan HKSA yang diperoleh dapat dikatakan sudah cukup baik dan valid baik secara internal maupun eksternal yang kemudian persamaan tersebut kemudian bisa digunakan untuk mendapatkan senyawa baru turunan benzoxazole dengan aktivitas sitotoksik yang lebih baik dan lebih potensial.

Abstract

Purpose: Archive management capacity is important for the performance of local government agencies, so adequate human resources are needed. This activity aims to improve the performance of archive managers so as to achieve a more effective and efficient archiving quality. Methodology: This activity is carried out with an active mentoring method involving several students implementing the activity. Participation observation activities are implemented through assistance during routine office activities. Evaluation of activities is carried out qualitatively through observation and quantitatively through pre-test and post-test questionnaires. Results: It is known if there have been changes in behavior and skills in some performance indicators. Likewise, quantitatively it is known if there is a change in all indicators of attitudes and skills of the target group. Limitations: Performance performed only on manual archiving activities, not involving technology or other tool support. Contribution: The results of this study will be useful for archiving activities, lecturers and archival students who need insight into archiving activities at the DPRD Office. As a reference for the program to strengthen the capacity of human resources in the field of archives.
