Abstract

Uncaria gambir Roxb., commonly known as Bajakah Kalalawit, is a plant endemic to Kalimantan, Indonesia, with potential medicinal properties. While previous studies have investigated the phytochemical composition of U. gambir leaves, limited information exists regarding the constituents of its stem extract. This study aimed to comprehensively characterize the chemical composition of U. gambir stem extract using Liquid Chromatography-High Resolution Mass Spectrometry (LC-HRMS). Ethanol (96%) was employed as the solvent for maceration extraction (1 : 5, plant material : solvent ratio) for three days. Subsequently, the dried extract was subjected to LC-HRMS analysis. Compounds with an Area Under the Curve (AUC) value greater than or equal to 1% were considered as major constituents. The results revealed the presence of 18 distinct chemical compounds in the U. gambir stem extract, providing valuable insights into its phytochemical profile and laying the foundation for further investigations into its potential pharmacological activities.

Abstract

Bajakah Kalalawit (Uncaria gambir Roxb.) is a typical plant of the Dayak tribe of Kalimantan, Indonesia. This study aims to evaluate the antidiabetic potential of Bajakah Kalalawit stem extract through in silico and in vitro approaches. In silico analysis was conducted to identify the interaction of bioactive compounds in 96% ethanol extract of Bajakah Kalalawit stems with the alpha-glucosidase enzyme (PDB ID : 5NN8), which is the main target in the treatment of diabetes. The results of molecular simulations showed that 2 bioactive compounds of the flavonoid group, namely Bikisocoumarin (-8.25 kcal/mol) and Pinostrobin (-5.81 kcal/mol) contained in 96% ethanol extract of Bajakah Kalalawit stems have significant inhibitory potential against the enzyme. Furthermore, in vitro tests were conducted to assess the inhibitory activity against the alpha-glucosidase enzyme and its effect on reducing blood glucose levels. The experimental results showed that 96% ethanol extract of Bajakah Kalalawit stems was able to significantly (ρ < 0.05) inhibit alpha-glucosidase activity (IC50 = 0.296 ± 0.004 μg/mL). This finding indicates that Bajakah Kalalawit has the potential as a source of active antidiabetic ingredients that can be further developed as an alternative therapy for diabetes management.

Abstract

Abstrak Gangguan Kecemasan adalah jenis gangguan jiwa terbanyak kedua di Indonesia. Upaya untuk mengurangi kecemasan pada remaja di Indonesia saat ini menghadapi berbagai kendala, salah satunya adalah minimnya literasi kesehatan jiwa dan minimnya edukasi terkait keterampilan manajemen stress. Semakin meningkatnya kejadian kecemasan, semakin penting dalam mengatasi masalah tersebut dengan cara yang tepat. Salah satunya dengan menggunakan herbal lavender dan chamomile. Oleh karena itu, tujuan penelitian ini adalah untuk membandingkan tingkat pengetahuan remaja terkait manfaat konsumsi teh lavender dan chamomile dalam mengurangi tingkat kecemasan pada remaja khususnya pada saat menghadapi ujian sekolah di Mts dan MA Asshodiqiyah Kota Semarang. Metode penelitian ini yaitu pemberian edukasi, pretest dan posttest. Metode yang digunakan adalah cross-sectional dengan cara survey. Data dikumpulkan melalui kuesioner terstruktur yang mencakup indikator pengetahuan dasar terkait kecemasan, manfaat lavender dan chamomile. Responden penelitian sebanyak 45 murid MTS dan MA Asshodiqiyah Semarang. Peningkatan persentase jawaban dari pre-test (70,9%) ke post-test (93,1%) dengan adanya perbedaan signifikan (p=0,002) pada pengetahuan siswa menunjukkan bahwa informasi diterima dengan baik. Edukasi berkelanjutan terkait gangguan kecemasan secara menyeluruh direkomendasikan untuk meningkatkan pemahaman siswa atau remaja di Kota Semarang. Kata kunci: kecemasan; tehe lavender; teh chamomile; ujian sekolah; remaja.Abstract Anxiety disorders are the second most prevalent type of mental disorder in Indonesia. Efforts to reduce anxiety among adolescents in Indonesia currently face various challenges, including limited mental health literacy and insufficient education regarding stress management skills. As the prevalence of anxiety continues to increase, it becomes increasingly important to address this issue through appropriate interventions. One potential approach is the use of herbal remedies such as lavender and chamomile. Therefore, the objective of this study was to compare adolescents’ levels of knowledge regarding the benefits of consuming lavender and chamomile tea in reducing anxiety levels, particularly during school examinations, among students at MTS and MA Asshodiqiyah in Semarang city. The research methodology involved the provision of educational interventions followed by pre-test and post-test assessments. The analytical method employed was a cross-sectional study with an survey. Data were collected using structured questionnaires that included indicators of basic knowledge related to anxiety as well as the benefits of lavender and chamomile. The study respondents consisted of 45 students from MTS and MA Asshodiqiyah Semarang. An increase in the percentage of correct responses from the pre-test (70,9%) to the post-test (93,1%), with a statistically significant difference (p = 0.002), indicates that the information was well received by the students. Continuous and comprehensive education on anxiety disorders is recommended to enhance students’ and adolescents’ understanding in the city of Semarang. Keywords: anxiety; students; lavender tea; chamomile tea; exam.
