Abstract

Background: Cervical cancer is the fourth most common cancer worldwide and is caused by infection with high-risk Human Papillomavirus (HPV). Sirtuin 1 (SIRT1) plays a role in cell cycle regulation, apoptosis, and the response to oxidative stress, contributing to the progression of cervical cancer cells. Oxidative stress induced by H2O2 can influence the expression of genes involved in cancer cell adaptation. Purpose: To analyze the effect of H2O2 administration on SIRT1 gene expression in the HeLa cell line. Method: This is an in vitro experimental study using the HeLa cervical cancer cell line. Cells were cultured and divided into three groups: an untreated control group, a 250 µM H2O2 treatment group, and a 1000 µM treatment group. SIRT1 gene expression was analyzed using quantitative real-time polymerase chain reaction with GAPDH as the reference gene. Data analysis was performed using the Kruskal-Wallis test with a significance level of 0.05. Results: Median (min-max) SIRT1 gene expression increased in the 250 µM and 1000 µM treatment groups compared to the control group. However, statistical analysis showed that the difference in SIRT1 gene expression between groups was not statistically significant. Conclusion: Administration of 250 µM and 1000 µM H2O2 did not affect or decrease SIRT1 gene expression in HeLa cell lines.   Keywords: Cervical Cancer; HeLa Cell Line; HPV-18; Oxidative Stress; Sirtuin 1.   Pendahuluan: Kanker serviks merupakan kanker dengan kejadian tertinggi ke-4 di dunia yang disebabkan oleh infeksi Human Papillomavirus tipe high-risk. Sirtuin 1 (SIRT1) berperan dalam regulasi siklus sel, apoptosis, serta respons terhadap stres oksidatif dan berperan dalam progresivitas sel kanker serviks. Stres oksidatif yang di induksi H2O2 dapat memengaruhi ekspresi gen yang berperan dalam adaptasi sel kanker. Tujuan: Untuk menganalisis pengaruh pemberian H2O2 terhadap ekspresi gen SIRT1 pada lini sel HeLa. Metode: Penelitian ini merupakan studi eksperimental in vitro menggunakan HeLa cervical cancer cell line. Sel dikultur dan dibagi menjadi tiga kelompok, yaitu kelompok kontrol tanpa perlakuan, kelompok perlakuan H2O2 dosis 250 µM, dan 1000 µM. Ekspresi gen SIRT1 di analisis menggunakan quantitative real time polymerase chain reaction dengan GAPDH sebagai gen pembanding. Analisis data dilakukan menggunakan uji Kruskal-Wallis dengan tingkat kemaknaan 0.05. Hasil: Median (min-max) hasil ekspresi gen SIRT1 meningkat pada kelompok perlakuan dosis 250 µM dan 1000 µM dibandingkan kelompok kontrol. Namun, hasil analisis statistik menunjukkan bahwa perbedaan ekspresi gen SIRT1 antar kelompok tidak bermakna secara statistik.   Simpulan:pemberian H2O2 dosis 250 µM dan 1000 µM tidak memengaruhi atau menurunkan ekspresi gen SIRT1 pada lini sel HeLa.   Kata Kunci: Kanker Serviks; Lini Sel Hela; Sirtuin 1; Stres Oksidatif; HPV-18.
