Abstract

Background: Fibrosis involves excessive extracellular matrix (ECM) deposition, with matrix metalloproteinase-9 (MMP-9) playing a critical role. Natural compounds targeting MMP-9 may offer therapeutic potential. Objective: This study evaluated the antifibrotic effect of Allium sativum extract on fibroblast cells through in vitro and in silico approaches targeting MMP-9. Methods: Human dermal fibroblasts were treated with A. sativum extract (25, 50, and 100 µg/mL) for 24 and 48 hours. Cell viability was assessed using MTT assay. MMP-9 expression was analyzed via quantitative real-time PCR (qRT-PCR) and immunocytochemistry (ICC). Major garlic compounds were docked to MMP-9 using AutoDock Vina. Results: The 50 µg/mL concentration maintained >85% cell viability and significantly reduced MMP-9 gene and protein expression. S-allyl cysteine exhibited the strongest binding affinity (−6.8 kcal/mol) and formed hydrogen bonds with key residues (Glu402, Ala189, His405) of MMP-9. Conclusion: Allium sativum extract demonstrates antifibrotic activity by downregulating MMP-9 expression and inhibiting its function, supporting its potential as a natural MMP-9 inhibitor for fibrosis-related disorders.

Abstract

Adolescence is one of the periods of human development. This is a time of change or transition from childhood to adulthood that includes biological, psychological, and social changes. Adolescence usually begins at the age of 10 – 13 years and ends at the age of 18 – 22 years. Meanwhile, according to WHO, adolescents are individuals who are experiencing a transition period that gradually reaches sexual maturity, experiences a change in the psyche from a child's soul to an adult, and experiences a change in economic circumstances from dependence to relative independence. There are two main aspects of change in adolescents, namely physical or biological changes and psychological changes. Many problems will arise as a result of neglecting reproductive health. Problems that arise due to lack of knowledge about reproductive health are Unwanted Pregnancy (KTD), abortion, early marriage and marriage, STIs or STDs and HIV/AIDS (Marmi, 2013). One of the efforts that can be made to reduce these numbers is by conducting health education about how to care for reproductive organs, education about adolescent development during puberty, health education about the impact of pornography, health education about unwanted pregnancies (KTD) and abortion, health education about HIV/AIDS and sexually transmitted infections, and health education about maturing the age of marriage with involving the role of the government, parents, and also peer groups. By carrying out these activities, it is hoped that it will be able to increase the knowledge of adolescents, so that it can increase adolescents' awareness of the importance of reproductive health problems. And reduce the incidence of adolescent reproductive health cases.

Abstract

Mental health problems have become unresolved health problems in society, both at the global and national levels. Especially during the COVID-19 pandemic, mental health problems will be more difficult to solve. The increasingly concerning condition of people's mental health will affect national productivity. This could hinder Indonesia from transitioning into a developed country by 2045. Mental health services are crucial and require the same attention as physical health. According to the Indonesia-National Adolescent Mental Health Survey 2022, 15.5 million (34.9 percent) adolescents experience mental problems and 2.45 million (5.5 percent) adolescents experience mental disorders. Of that number, only 2.6 percent accessed counseling services, both emotional and behavioral. The government has been thinking about mental health, but it has not been a priority. Access to mental health services is expensive and not inclusive. Meanwhile, attention to mental and physical health has not been equal in national health insurance. In Indonesia, we know many cases of adolescents aged 15-17 who experience excessive anxiety resulting in depression and suicide.
