Abstract

The fixation process is the first stage in the manufacture of histopathological preparations which aims to preserve the tissue and harden the tissue, so that the tissue to be observed does not change in shape or size. BNF fixation solution (Neutral Buffer Formalin) 10% has been used as a routine fixative and It has been be the gold standard in histology laboratories for decades. The material used in this study was a 10% BNF solution because it is easier to use and it can be used to preserve tissue for a long period of time. This study aims to determine the differences in the microscopic results of the liver tissue of mice (Mus musculus) fixed with 10% BNF for 6 hours and 16 hours. This type of research is descriptive analytic. The research was carried out at the Barokah Laboratory/ dr.Mezfi Unita on 04-05 March 2022 with a total sample of 20 samples of mice liver tissue (Mus musculus). The results of the study on 10% BNF fixation for 6 hours and 16 hours showed that the average results were not good. The conclusion from the results of the study that there was no difference in the microscopic results of liver tissue that were fixed with 10% BNF for 6 hours and 16 hours using Hematorylin-Eosin staining. 10% BNF fixation with a time of 6 hours and 16 hours can be used as an alternative in histopathological examination in the short term and long term.

Abstract

Kandidiasis merupakan infeksi yang dapat bersifat primer maupun sekunder, disebabkan oleh Candida albicans, salah satu penyebab umum keputihan (fluor albus). Indonesia dengan iklim tropis memiliki keanekaragaman tanaman yang berpotensi sebagai obat alami, termasuk rumput teki (Cyperus rotundus Linn.) yang sering dianggap gulma. Umbi rumput teki mengandung senyawa kimia yang berpotensi sebagai antifungi alami. Penelitian ini bertujuan menguji potensi ekstrak umbi rumput teki terhadap C. albicans dengan berbagai konsentrasi. Penelitian bersifat deskriptif eksperimental menggunakan umbi rumput teki sebagai populasi. Ekstraksi dilakukan dengan metode maserasi menggunakan pelarut etanol 96% pada empat konsentrasi: 25%, 50%, 75%, dan 100%. Flukonazol digunakan sebagai kontrol positif dan CMC 1% sebagai kontrol negatif. Uji daya antifungi dilakukan menggunakan metode difusi cakram dan dilusi untuk menentukan Konsentrasi Hambat Minimum (KHM), dengan media Sabouraud Dextrose Agar (SDA) dan Sabouraud Dextrose Broth (SDB). Hasil penelitian menunjukkan diameter rata-rata zona hambat: konsentrasi 25% sebesar 6,75 mm; 50% sebesar 7 mm; 75% sebesar 12,75 mm; 100% sebesar 16 mm. Zona hambat kontrol positif (flukonazol) sebesar 22 mm, sedangkan kontrol negatif (CMC 1%) sebesar 0 mm. Standar deviasi rata-rata zona hambat adalah 7,79. KHM ekstrak umbi rumput teki terhadap C. albicans diperoleh pada konsentrasi 100%. Kesimpulannya, ekstrak umbi rumput teki memiliki aktivitas antifungi terhadap Candida albicans, dengan efektivitas meningkat seiring kenaikan konsentrasi, meskipun masih di bawah efektivitas flukonazol.
