Abstract

Avocado plants (Persea americana Mill.) are plants that can be widely used as medicinal plants. Avocado seeds are often used empirically as traditional medicine, namely anti-inflammatory, overcoming constipation, maintaining endurance and treating diabetes mellitus. The use of avocado seeds as traditional medicine is because they have secondary metabolic compounds, one of which is flavonoids. This study aims to determine the total levels of flavonoid compounds in avocado seed ethanol extract using the UV-Vis spectrophotometry method. The study was conducted using qualitative and quantitative testing. The results of the study on avocado seed ethanol extract using qualitative testing with the Wilstater Cyanidin method and the  reagent method showed positive flavonoid content. While qualitative testing using the UV-Vis spectrophotometry method, it was obtained that the avocado seed ethanol extract had flavonoid content with an average level of 101.70 mgQE/g extract.   ABSTRAK Tanaman alpukat (Persea americana Mill.) merupakan tanaman yang secara luas dapat digunakan sebagai tanaman obat. Biji alpukat secara empiris sering digunakan sebagai obat tradisional, yaitu anti radang, mengatasi sembelit, menjaga daya tahan tubuh dan mengobati diabetes mellitus. Pemanfaatan biji alpukat sebagai obat tradisional karena memiliki senyawa metabolik sekunder, salah satunya yaitu flavonoid. Penelitian ini bertujuan untuk mengetahui total kadar senyawa flavonoid pada ekstrak etanol biji alpukat menggunakan metode spektrofotometri UV-Vis. Penelitian dilakukan dengan menggunakan pengujian kualitatif dan kuantitatif. Hasil penelitian pada ekstrak etanol biji alpukat menggunakan pengujian kualitatif dengan metode Wilstater Cyanidin dan metode pereaksi  menunjukkan positif memiliki kandungan flavonoid. Sedangkan pengujian kualitatif menggunakan metode spektrofotometri UV-Vis, diperoleh bahwa ekstrak etanol biji alpukat memiliki kandungan flavonoid dengan rata-rata kadar yaitu sebesar 101,70 mgQE/g ekstrak.

Abstract

Escherichia coli is a bacteria that causes diarrhea transmitted through water or food contamination and is treated with antibiotics. Excessive antibiotics have the potential to cause resistance. Natural ingredients such as clove leaves are used as an alternative to overcome resistance because they have antibacterial activity. This study aims to determine the antibacterial activity of clove leaf extract (Syzygium aromaticum (L.) Merr. & Perry) against Escherichia coli bacteria and its minimum inhibitory level. Clove leaves were macerated using 96% ethanol. Identification of eugenol content was carried out using thin layer chromatography. The antibacterial activity test in this study used the well diffusion method with concentrations of 1%, 2%, 3%, 4%, 5%, 6%, 7%, 8%, 9%, 10%. In this study, the results of clove leaf extract extraction were obtained with a yield of 15.7975%. The results of identification using thin layer chromatography showed that clove leaf extract contains eugenol. Ethanol extract of clove leaves has antibacterial activity against E. coli bacteria at concentrations of 5%, 6%, 7%, 8%, 9% and 10%, while at concentrations of 1% to 4% it has no antibacterial activity.
