Abstract

Fibrosis is a progressive pathological process characterized by excessive accumulation of extracellular matrix and represents a major cause of organ failure, including in the kidney, liver, and lung. Currently available antifibrotic therapies have limited efficacy and are associated with potential adverse effects, highlighting the urgent need for new drug candidates derived from natural sources. The seagrass Enhalus acoroides, which is abundant in Indonesian waters, has been reported to contain flavonoids, polyphenols, sterols, and bioactive fatty acids that may modulate oxidative stress and inflammation, two key mechanisms involved in fibrogenesis. This review aims to evaluate the biochemical and pharmacological potential of E. acoroides as a source of antifibrotic compounds for national drug development. The methodology involved a literature review of scientific databases focusing on phytochemical profiles, pharmacological activities, and molecular mechanisms related to fibrosis. The findings indicate that E. acoroides exhibits significant antioxidant and antibacterial activities, contains major secondary metabolites (notably flavonoids and phenolics), and shows mechanistic relevance to the TGF-β/Smad signaling pathway, a central regulator of fibrogenesis. In conclusion, E. acoroides has promising potential as a candidate antifibrotic phytopharmaceutical; however, further validation through in vitro, in vivo, and advanced pharmaceutical formulation studies is required

Abstract

Gastritis, commonly known as stomach ulcers, is a widespread condition caused by poor eating habits, certain foods, infections, and NSAID use. In Indonesia, WHO reports a 40.8% prevalence, making it a major health issue. This study aimed to evaluate the effects of ethanol-extracted shallot (Allium sp.) skin and bulb on malondialdehyde (MDA) levels and gastric histopathology in rats with indomethacin-induced gastritis. Gastritis was induced in Rattus norvegicus, followed by treatment with shallot extract. MDA levels and tissue damage were then analyzed using Hematoxylin-Eosin staining. Results showed a significant reduction in MDA levels, especially with 200 mg/kg body weight of shallot skin extract, which achieved a 92.18% decrease. Histopathological analysis revealed notable improvements in gastric tissue, with reduced ulceration and inflammation. The findings support the potential of shallot extract in lowering oxidative stress and repairing gastric damage, suggesting its promise as an alternative therapy for gastritis.

Abstract

Traditional herbal medicines are widely used due to their perceived safety and natural origin; however, illegal addition of pharmaceutical substances such as paracetamol and sodium diclofenac poses adverse health effects. The objective of this study was to determine whether paracetamol and sodium diclofenac were present in jamu pegal linu samples via Thin Layer Chromatography (TLC) analysis. The study obtained all samples from local herbal stores in Pajarakan, Probolinggo. TLC analysis on silica gel GF254 plates was performed with compound-specific solvent systems: a 4:1 (v/v) ethyl acetate-chloroform mixture for paracetamol, and a 3:2 (v/v) ethyl acetate-n-hexane mixture for sodium diclofenac. Results revealed paracetamol was detected in three out of four samples with a retardation factor (Rf) of 0.61, while sodium diclofenac was found in three samples with Rf values around 0.90. One sample showed no presence of both compounds. These findings indicate illegal adulteration of herbal medicine, which may cause liver toxicity, kidney impairment, and cardiovascular risks. Therefore, strict supervision and consumer education are essential to prevent adverse health impacts.

Abstract

Kata kunci: temulawak; natrium diklofenak; estradiol; malondialdehid; tikus putih   Abstract Oxidative stress results from an imbalance between the production of reactive oxygen species (ROS) and the body's antioxidant capacity, which triggers cell damage through lipid peroxidation. Malondialdehyde (MDA) levels are widely recognized as biomarkers of this condition. Estradiol administration may elevate ROS and MDA levels. Curcuma xanthorrhiza Roxb.(Temulawak) contains curcuminoids and xanthorrhizole which exhibit antioxidant and anti-inflammatory properties. This study aimed to evaluate the effect of diclofenac sodium and temulawak ethanol extract on the serum MDA levels of estradiol-induced female rats, as well as determine the best dose. Using a post-test only control group design involving 25 Wistar mice divided into five groups: negative control, positive control (sodium diclofenac 10 mg/kgBB), and cucumin extract treatment at doses of 250, 500, and 1000 mg/kgBB. MDA levels were measured using the TBARS method using UV-Vis spectrophotometry, and then analyzed with Kruskal–Wallis and Mann–Whitney. Results showed significant differences among groups (p=0.000). Sodium diclofenac and curcuma extract lowered MDA compared to negative controls. A dose of 1000 mg/kgBB provided the greatest reduction, equivalent to diclofenac (p>0.05). In conclusion, temulawak ethanol extract effectively reduced estradiol-induced serum MDA levels in rats, with 1000 mg/kgBW identified as the most effective dose.
