Abstract

One of the plants used as medicine by the community is the Papuan batik leaf (Graptophyllum pictum L. Griff). Empirically, this leaf is used to treat anemia, treat wounds, liver swelling, gallstones, and treat coughs. This study aims to determine the results of the extract yield based on differences in cold extraction methods (maceration) and hot methods (soxhletation and reflux) and to identify functional groups contained in plants using the FTIR spectroscopy method and to see differences in total alkaloid and flavonoid compound levels based on differences in extraction methods using the UV-Vis spectrophotometry method. The results of the research that has been carried out indicate that there is the most effective extraction method used, namely the soxhletation method with an extract weight of 14 g and a yield percentage of 28%. Phytochemical screening test of Papua batik leaf ethanol extract using maceration, soxhletation, and reflux methods proved to contain alkaloid, flavonoid, tannin, terpenoid, and steroid compounds, also proven by the results of FTIR test with the presence of N-H, C=C, C-H, C-N, and C-O functional groups in the extract. The highest flavonoid content in the maceration method was 322.414 mg/g extract and the highest alkaloid content in the maceration method was 144.6 mg/g extract.
