Radicle emergence as a rapid indicator of sorghum seed vigor to predict mean germination time, vigor index, and field emergence. This study aimed to develop a standardized radicle emergence test for sorghum (Sorghum bicolor) seeds by determining the optimal single-count timing and temperature for the simultaneous assessment of seed vigor and viability. This test was conducted by calculating the percentage of seeds whose radicles had emerged by at least 2 mm in length during a single observation period at the beginning of germination. Seed lots with low radicle emergence rates showed slow germination, reflecting an early physiological indicator of seed deterioration and a key factor contributing to reduced vigor. The experiment was arranged in a randomized complete block design with one factor: 12 sorghum genotypes, and four replications (blocks). Radicle length was measured at 2-h intervals from 40 to 48 h, with a minimum length of 2 mm. Results of radicle emergence (production of 2 mm radicle) at 25°C and alternating temperatures 20⇔30°C for 44 h (the optimal time), mean germination time, germination percentage, germination speed, vigor index, maximum growth potential, tetrazolium test, and field emergence were compared using 12 genotypes of sorghum seeds with standard germination above 83%. The single counts of radicle emergence performed at 25°C and 20⇔30°C were highly correlated with key vigor and viability indicators. Correlations with vigor-related traits, such as mean germination time (R²=0.87-0.81), vigor index (R²=0.83-0.89), germination speed (R²=0.73-0.71), and field emergence (R²=0.77-0.70), were strong, as were those with viability parameters, including standard germination (R²=0.66-0.59) and tetrazolium test results (R²=0.58-0.52). This study concludes that a single count of radicle emergence at 44 h, conducted at either 25°C or 20⇔30°C, is a method for simultaneous assessment of seed vigor (mean germination time, vigor index, germination speed, and field emergence) and viability in sorghum, offering a significant improvement over more time-consuming tests, such as germination test that requires 10 days.

Biodiversitas 23: 2570-2579. Sorghum is an alternative source as a substitute for rice. Leaf rust (Puccinia purpurea) is a constraint in sorghum cultivation and resistance to rust disease is a condition for releasing sorghum varieties. This study aimed to screen sorghum accessions for resistance to leaf rust disease. The screening was carried out on 48 sorghum accessions and seven check varieties, namely Numbu, Kawali, Bioguma 1, Pahat, Soraya 1, Soraya 2, and Soraya 3. Evaluation of leaf rust resistance under field conditions was carried out in IPB University experimental field Cikarawang-Bogor (275 asl) from February 2021 to June 2021. The experiment was conducted in an augmented design with four replications for check varieties. Identification of leaf rust resistance was also carried out using molecular markers designed from the Lr34 and Rp1D genes. The results revealed that 5 genotypes, namely genotypes 31 (Demak 2), 24 (IS 18551), 43 (Lokal bima 3), 17 (ICSV 93036), and 33 (Demak 5) were resistant to leaf rust with high yield potential. The results of identification using molecular markers showed that all accessions had Lr34 and Rp1D gene fragments.

Biodiversitas 26: 628-635. Sorghum is a nutrient-rich food crop, an alternative commodity to substitute rice and other cereal crops, and can be grown on dry land; thus, it is important for food diversification to support food security and sustainable agriculture development in Indonesia. Utilization of high-quality seeds of superior varieties will improve sorghum productivity and quality. This study aimed to characterize the performance of 12 sorghum genotypes and identify potential lines for developing superior varieties. The planting materials used were nine BRIN sorghum lines (GHP-2, GHP-16, CTY-43, GH-1, GH-7, GH-9, G-5, G-7, G-8) and three national varieties (Bioguma 1, Pahat, Samurai 2). This research was conducted from June to October 2024 at the Institut Pertanian Bogor Leuwikopo Experimental Farm, Bogor, and Seed Quality Testing Development Center for Food Crops and Horticulture, Depok, West Java, Indonesia. The experimental design was a single-factor (genotypes) randomized complete block design with four replications. Seeds were planted in plots of 1x2.8 m with a spacing of 70x20 cm. Observations were made on agronomic characters and seed quality. The characters of plant height, 1000-seed weight, flowering age, and seed weight per plot significantly differed among genotypes. GH-9 line showed the highest productivity (2467.61 g seeds per plot) and 1000-seed weight (33.7 g). Of the 12 genotypes tested, the flowering ages ranged from 57 to 65 days after planting, with the fastest flowering age (57 days) on the CTY-43 line. Plant height measurements showed the lowest plant height (123.4 cm) on GHP-16, while the highest was GH-7 (228.9 cm).
