Abstrak. 4-(methyl nitrosamino )-1-(3-pyridyl)- l-butanone (NNN) dan NNK merupakan metabolit dari Tooacco Specific Nitrosamine (TSNA) yang dihasilkan dari proses merokok dan kedua metabolit ini bersifal karsinogenik. Dalam metabolismenya derifat TSNA ini melalui beberapa jalur mekanisme yang berbeda, NNK melalui mekanisme initial carbonyl reduction, pyridine oxydation, alpha-hydroxylation. NNN melalui denitrosation, pyridine-Nsoxydation dan pyrolidine oxydation. Hasil metabolisme NNN dan NNK membentuk berbagai metabolit seperti HPB, 7-methylguanine, 0-6 roethylguanine dan 0-4 methyl thymine yang berperan pada gangguan pada penyandian DNA.
