AbstrakKasus pemalsuan daging sapi dengan daging babi masih tetap ditemukan dan memiliki perhatian serius terkait dengan masalah halal terutama di Indonesia yang merupakan negara mayoritas muslim. Analisis spektrum FTIR memungkinkan untuk dapat menganalisis perbedaan kandungan protein pada daging babi dan sapi. Penelitian ini bertujuan untuk menganalisis daging sapi dan daging babi dengan FTIR dengan reaksi enzimatis menggunakan enzim protease. Sampel dibuat dalam beberapa kelompok yaitu sampel daging tanpa penambahan enzim, sampel dengan enzim protease, sampel yang tidak dikeringkan, serta sampel yang dikeringkan. Hasil penelitian menunjukkan FTIR dapat membedakan daging sapi dan daging babi. Spektrum FTIR yang dihasilkan memiliki keterulangan yang baik dan serapan khas dari daging sapi maupun daging babi. Spektrum derivatif pertama FTIR daging babi menghasilkan perbedaan pita-pita serapan pada bilangan gelombang 1318 cm-1, 1019-1066 cm-1, 705 cm-1, 681 cm-1 yang tidak ditemukan pada daging sapi.Kata kunci : daging sapi, daging babi, reaksi enzimatis, spektrofotometri FTIR, spektrumderivatif pertamaAbstractAdulteration case of beef with pork still remain found and had serious attention due to halal issue especially in Indonesia as a moslem majority country. FTIR spectrum analysis makes it possible to analyze differences in protein content in pork and beef. This study aims to analyze the beef and pork with FTIR with enzymatic reaction using protease enzymes. Samples were made in several groups, namely samples without enzyme addition, samples with protease enzymes, non-dried samples, and dried samples. The result of this research indicate that FTIR method successfully differentiate beef to pork. The analysis result showed the measurument using FTIR spectrum were relative repeatable and produced some unique bands both pork or beef. The first derivative spectrum FTIR of pork produced different absorption bands at wave numbers 1318 cm-1, 1019-1066 cm-1, 705 cm-1, 681 cm-1 which were not found in beef.Keywords : beef, pork, denaturation of enzyme, spektrofotometri FTIR, first derivative spectrum
