Some of the people in Jetis Sukoharjo Village use dug wells as the main source for washing dishes, bathing and so on. Use of water for daily needs such as brushing teeth and washing food to be processed into ready-to-eat food. Escherichia coli is a group of coliform bacteria. The presence of coliform bacteria in water indicates the presence of microbes that are enteropathogenic and toxigenic which are dangerous to health. The higher the E. coli contamination in the water can cause digestive disorders, diarrhea and even food poisoning (KLB). This research, apart from informing about the presence of E. coli contamination from well water samples in Jetis Sukoharjo village, also tested butterfly pea flower kombucha, which has the potential to be an antibacterial source. This study aims to identify E. coli bacteria from well water samples from Jetis village, Sukoharjo subdistrict and test the average diameter of the inhibition zone in butterfly pea flower kombucha on E. coli bacteria from well water samples from Jetis village, Sukoharjo. This research method was carried out descriptively and quantitatively (ANOVA). The results of the research have shown that the examination carried out in Jetis Sukoharjo Village found contaminants in the form of Escherichia coli based on the results of isolates on MCA (Mac Conkey Agar) media with colonies characterized by round shape, flat and dry surface, convex surface, small size, and reddish color. Furthermore, with biochemical tests, the results were Indol (+), MR (Methyl Red) (+), VP (Voges Proskauer) (-), and Citrate (-). From the results of the identification test, it can be stated that the bacterial isolates resulting from the isolation of dug well water truly contain pure Escherichia coli. Telang flower kombucha at all sugar concentrations has potential as an antibacterial for E. coli originating from well water samples from Jetis Sukoharjo Village.
