Effect of Staurogyne elongata Leaf Extract on Macrophages, Fibroblast Proliferation, Collagen Deposition, and Excisional Wound Closure in BALB/c Mice

Abstract

A wound is a disruption of tissue integrity caused by physical, chemical, thermal, microbiological, or immunological injury, resulting in structural and functional alterations of the skin. Wound healing is a complex process involving inflammatory, proliferative, and remodeling phases. Macrophages regulate inflammation and phagocytosis, whereas fibroblasts contribute to tissue repair through collagen synthesis. Reundeu (Staurogyne elongata) leaf extract possesses antibacterial and antioxidant properties that may enhance wound healing. This study aims to evaluate the effects of Staurogyne elongata leaf extract on macrophages, fibroblast proliferation, collagen deposition, and excision wound healing in BALB/c mice. This study was conducted at the Universitas Jenderal Achmad Yani laboratories in Cimahi from October 17, 2025, to January 27, 2026. It evaluated the effects of S. elongata leaf extract on wound healing rate, macrophage and fibroblast counts, and collagen density in mouse excision wounds. A post-test-only control group design was used. Forty mice were assigned to five groups: a negative control, a positive control (0.5% chlorhexidine), and treatment groups receiving extracts at 6.25%, 12.5%, and 25%. Tissue samples were collected on days 0, 1, 3, and 7. Wound healing was assessed by wound area reduction. Data were analyzed using Kruskal-Wallis and Mann-Whitney tests (p≤0.05). The 6.25% extract group showed the highest wound healing rate and wound closure on day 7. The highest macrophage count was observed in the 25% extract group on day 3, whereas the highest fibroblast count and collagen density were found in the 12.5% and 25% extract groups on days 3 and 7. In conclusion, these findings suggest that S. elongata leaf extract promotes wound healing by enhancing macrophage activity, fibroblast proliferation, collagen deposition, and tissue regeneration in mice.
