Galam plant (Melaleuca cajuputi) has active compounds of galam plant that have anticancer, anti-inflammatory, antibacterial, hepatoprotective, anthelmintic activities. This research aims to find out the thin layer chromatography profile of flavonoid compounds in the ethanol extract of galam leaves. This research is a quantitative research with a laboratory experimental research design by analyzing the thin layer chromatography (TLC) profile of flavonoid compounds with various types of eluents, namely: Eluent n-Hexane: Ethyl acetate (8:2), Eluent n-Hexane: Dichloromethane (8:2) and Ethyl acetate: Dichloromethane (9:1) with quercetin as a comparator. The best eluent is expressed from the Rf value of the compound that is close to the Rf value of quercetin. The result of KLT of flavonoid compound with ethyl acetate:n-Hexane (8:2) eluent, quercetin showed an Rf value of 0.46 and 1 spot that is close to the Rf value of quecetin, namely A2 (0.47). In eluent n-Hexane: Dichloromethane (8:2) shows Rf value of quercetin 0.36 and Rf A1 value: 0.32 and in eluent Ethyl acetate: Dichloromethane (9:1) Rf value of quercetin is 0.76 and there are no spots close to the Rf value of Quercetin. The conclusion of the best eluent in the thin layer chromatography profile of the flavonoid compound of galam leaf extract is ethyl acetate:n-Hexane (8:2).
