This study aimed to describe the antibiotic susceptibility patterns of Staphylococcus aureus isolated from diabetic ulcer patients with varying degrees of ulcer severity at Prof. Dr. Margono Soekarjo Hospital, Purwokerto. This study employed a descriptive observational design with a cross-sectional approach. A total of 20 diabetic ulcer patients were enrolled using an accidental sampling technique from October to November 2025. Bacterial identification was performed through microbiological examination, while antibiotic susceptibility testing was conducted using the Kirby–Bauer disk diffusion method. Ulcer severity was classified according to the Wagner classification system. The results showed that 70 bacterial isolates were obtained from the 20 diabetic ulcer patients, of which 46 isolates were identified as S. aureus. The susceptibility testing revealed that the susceptibility rates of S. aureus to linezolid, cefoxitin, and ciprofloxacin were 43.5%, 15.2%, and 26.1%, respectively, with variations observed across different Wagner grades. In contrast, most S. aureus isolates exhibited resistance to the tested antibiotics, including linezolid (54.3%), cefoxitin (80.4%), and ciprofloxacin (63.0%). The antibiotic susceptibility patterns of Staphylococcus aureus isolated from diabetic ulcers varied according to ulcer severity. Therefore, continuous monitoring of antibiotic susceptibility patterns and the availability of local bacterial resistance data are essential to support rational and effective antibiotic selection.
