This study aimed to develop and validate a guided inquiry-based Cell Biology practicum guide integrated with Maluku local wisdom and to examine students’ responses to the developed product. The study employed a Research and Development (R&D) approach using the 4D model, limited to the Define, Design, and Develop stages. The research involved six expert validators and 20 second-semester Biology Education students in a limited trial. Data were collected using expert validation sheets and a student response questionnaire and analyzed using Aiken’s V, feasibility percentages, Cronbach’s Alpha, and descriptive statistics. The results showed an average Aiken’s V score of 0.86, indicating high validity, while the overall feasibility percentage reached 89.05%, categorized as highly feasible. The student response questionnaire demonstrated high reliability with a Cronbach’s Alpha value of 0.819. Student responses reached 83.25%, categorized as good. These findings indicate that the developed practicum guide is valid, feasible, and suitable for limited implementation in Cell Biology learning.
