This study aimed to determine the secondary metabolite content and antibacterial activity of ethanolic extract of purple ruellia leaves (Ruellia tuberosa L.) against the growth of Staphylococcus aureus. The study used a laboratory experimental method with extract concentrations of 5%, 10%, 15%, and 20%. Phytochemical screening revealed the presence of alkaloids, flavonoids, tannins, saponins, and steroids. Antibacterial activity was evaluated using dilution and well diffusion methods. The results showed that all extract concentrations inhibited the growth of Staphylococcus aureus. The average inhibition zone diameters at concentrations of 5%, 10%, 15%, and 20% were 4.33 mm, 7.00 mm, 10.67 mm, and 14.67 mm, respectively. The 20% concentration exhibited the highest antibacterial activity with a strong category. One-Way ANOVA analysis showed a significance value of 0.005 (p<0.05), indicating significant differences among the tested concentrations. It can be concluded that the ethanolic extract of Ruellia tuberosa L. leaves possesses antibacterial activity against Staphylococcus aureus and has potential as a natural antibacterial agent.
