Background: The pleura is a serous membrane that folds on the surface of the lungs to form a two-layer membranous structure. The gold standard in establishing the diagnosis of pleural effusion is cytological examination. An important stage in cytological examination is the staining process, where the staining technique uses eosin. Eosin as a dye component is carcinogenic and poses a risk to health and the environment if used in the long term. Beetroot containing red pigment, which is a betacyanin compound, was developed as a natural alternative to eosin. Research Objective: This study aims to determine the use of Beetroot (Beta vulgaris L.) as a substitute for eosin in Papanicolaou staining. Research Method: This is an experimental study with an Intact Group Comparison design consisting of two treatment groups, namely staining using eosin and beetroot. Data analysis was carried out using descriptive statistics. This study was conducted at the Barokah Palembang anatomical pathology laboratory on March 16-17, 2025. The population taken was pleural effusion fluid preparations located at the Barokah Palembang anatomical pathology laboratory, the samples used in this study amounted to 32 samples calculated using the Federer formula. Results: : The results of staining using eosin showed that the quality of the preparation was 100% very good, while for staining using beetroot, the quality of the preparation was not good at 18.7%, less good at 50.0%, and good at 31.2%. Conclusion: Alternative staining beetroot can be made into a coloring alternative to the eosin substitute in Papanicolaou coloring.
