Abstract

AbstractThe aim of this research are to learn and find out the katG gene mutation of Mycobacterium tuberculosis as the primary resistence properties to the first-line oral agents isoniazid ( INH ) in patients with AFB ( Acid Fast Bacilli ) ( + ) pulmonary tuberculosis. This research applies descriptive observasional mode due to the detection of the presence of katG gene mutation of Mycobacterium tuberculosis as the primary resitance properties to the first-line oral agents isoniazid( INH ) in patients with AFB ( Acid Fast Bacilli ) ( + ) pulmonary tuberculosis. Fifty samples are obtained from 16 community health center by using accidental sampling technique. Nested PCR is the method to analyze the katG gene of Mycobacterium tuberculosis as the target in this research to collect data. The result of this research shows that a mutation presence  in the primer complimenter region of  katG gene of Mycobacterium tuberculosis as the target are found in 11 samples ( 22% ) wich are expressed as resistance and 39 ( 78% ) are expressed as sensitive or have no mutation. All the eleven mutation detected samples are caused by nucleotide mismatch. [JAMBS,2014;1(1):1-15]Keywords:  Primary Resistance,  katG gene,  Mycobacterium tuberculosis.
