Abstract

Staphylococcus aureus is a commensal bacterial  of the body that can become pathogen at any time. Along with the increase of antibacterial resistant cases, it is necessary to explore the new potential metabolites as antibacterial. Endophytic bacteria that live on turmeric (Curcuma longa L.) are thought to be able to produce secondary metabolites that are similar and even more varied than their host plants. To determine the antibacterial effect of secondary metabolite compounds of turmeric (Curcuma longa L.) endophytic bacteria on Staphylococcus aureus. Endophytic bacterial isolates were subcultured on NA media with a streak plate method. Antibacterial activity test useds disk diffusion method. Potential endophytic bacterial isolates were characterized based on colony morphology, cell morphology and biochemical properties and secondary metabolite compounds produced are identified with a ciulei method. A total of 17 endophytic bacterial isolates were successfully subcultured. All endophytic bacterial isolates had antibacterial activity range from 7.4-13.76 mm. One of the most potential isolates was M8 isolate which had similarity with a genus Pseudomonas. The secondary metabolites that was produced bt M8 isolates were alkaloids, terpenoids and saponins. The most potential endophytic bacteria as antibacterial belong to the genus Pseudomonas and had alkaloids, terpenoids and saponins.

Abstract

The 96% ethanol extract of jamblang fruit peel has anthocyanin which has the potential to be used as a coloring agent for lipstick products. Jamblang fruit peel was extracted with 96% ethanol by kinetic maceration and tested for standard extract quality test, phytochemical screening, total anthocyanin values measured at 510nm and 700nm and antioxidant IC50 by DPPH method. The lipstick is made in 3 pH variations (Acid, Neutral and Alkali). Melting point, hardness, stability test with cycle test method, changes in shape, color and odor, smear test, pH test and skin irritation of male albino rabbits were also carried out in this study. Results showed that Jamblang fruit extract had antioxidant activity IC50 of 196.30 g/mL (average activity), total Anthocyanin value 0.2642%. Free from heavy metal content. The anthocyanin color in the extract of Syzigium cuminii L also changed according to the change in pH. The lipstick preparation is easy to apply, stable in three test cycles and does not irritate the skin of the shaved male albino rabbit. The conclusion, Jamblang fruit peel extract can be used as a natural dye in lipstick preparations.

Abstract

Diabetes Mellitus (DM) is a degenerative disease that can cause various complications such as cardiovascular, stroke, and hypertension. Antidiabetic drugs that are commonly circulated in the market, have side effects that can harm other organs such as liver, kidneys and other organs. Bay leaves have properties as inhibitors of the α-glucosidase enzyme in vitro and in vivo. The aims of this study was determine the activity of ethanolic extract of bay leaves by ultrasonic extraction method in inhibiting the α-glucosidase enzyme in vitro and in silico. Bay leaves were extracted using 70% and 96% ethanol by ultrasonic method for 15 minutes, and filtered and concentrated using a rotary vacuum evaporator 4 times. The extract obtained was tested for phytochemical content and the activity of α-glucosidase inhibitory assay. The extract with the best IC50 value was then separated using column chromatography (SiO2, dichloromethane, methanol) and produced 7 fractions and tested for α-glucosidase enzyme inhibition. The best fraction (fraction 6) was analyzed by Liquid Chromatography Mas Spectra (LCMS-MS). The results showed that bay leaf extract containing alkaloids, flavonoids, tannins, steroids, phenols, and fraction-6 had the best IC50 of 30.82 ppm. The results of LCMS-MS analysis of chemical compounds as inhibitors of the α-glucosidase enzyme were delphinidin (m/z 303) and had a rerank score of -75.59. In the in silico test using PLANTS docking software.

Abstract

Almost all diseases in the body cause pain. Currently the most widely used analgetic has not regardless from side effects. The development of new and effective pain medications that are natural is expected to have the benefits of treatment. 75% ethanol extract of dandang gendis leaf (Clinacanthus nutans (Burn F) Lindau) dosage 279,3 mg/kg BW and 96% ethanol fraction of bakung leaf (Crinum asiaticum) dosage 250 mg/kg BW were shown to have antinociceptive activity. The purpose of this study was to determine the antinociceptive effectiveness of a combination of 75% ethanol extract from dandang gendis leaves and 96% ethanol fraction of bakung leaf. In this study was used chemical method induction (Sigmund's method) in 25 male mice which were divided into 5 treatment groups, namely treatment group I as negative control was given CMC 0.5%, treatment group II as positive control was given acetosal 100 mg / kg BW, treatment groups III, IV and V were given combinationsextract. Each group was given treatment orally, thirty minutes then induced with 0.6% acetic acid intraperitonially, after which it was observed and counted for the amount of stretching for sixty minutes. Data were analyzed by one-way ANOVA to see differences in antinociceptive effects in each treatment group. The results of this study showed that all treatment groups had a significant difference (p <0.05), the combination of 75% ethanol extract dandang gendis leaf and 96% ethanol fraction of bakung leaf with a dose of 100 mg / kg BW had good effectiveness with a percent effectiveness value of 92.50%. The results of this study indicate that the combination of 75% ethanol extract from dandang gendis leaf (Clinacanthus nutans (Burn F) Lindau) and 96% ethanol fraction from bakung leaf (Crinum asiaticum) with the same concentration ratio, have relatively higher antinociceptiveof the activity of each extract, in other words both of them work together.
