Abstract

Studies on genet ic position of several iso lates liv ing symbiot i a lly on ParaseriCllltli esfalcalaria have been carried out using amplification and se uenc ing tec hni ques of 16S rDNA and specific gene fo r nodulation (/lode) . Abstract

The embryogenesis (SE) has important role for genetic engineering of cassava (Manihot esculenta Crantz). However, the success of SE induction depend on plant growth regulator s (PGR)s and treatment enriched in induction media.  This experiment tried to induce callus formation of cassava from several in vitro explants: immature leaf, apical bud, and internode; and to develop somatic embryogenesis of cassava in several media enriched with tyrosine and copper sulphate (CuSO4) added into media enrich with picloram as treatment.  Different response of explants source to callus induction treatment from those three varieties in callus induction as well as friable callus formation were found in this experiment. The best medium to induce varied with variety; MS media supplemented 12 mg/L picloram + 0.5 mg/L CuSO4 was the best for “Adira 4” and  half MS and half GD media supplemented 12 mg/L picloram + 100 mg/L tyrosine for “Malang 6”.  All treatments resulted somatic embryo which developed indirectly and in morphologically normal somatic embryos

Abstract

One sesquiterpene compound has been isolated and identified out of the ethylacetate extract of bawang hutan (Scorodocarpus borneensis Becc.) barks. The barks were macerated with methanol, and then partitioned with mixture of ethylacetate-water (1:1).  Fractionation of the ethylacetate phase by column chromatography gave pure compound. Based on data interpretation from of ultra-violet (UV) spectra, Fourier Transform Infra Red (FT-IR), NMR 1D (1H and13C-NMR); NMR 2D (HMQC, COSY, HMBC), and comparison with literature, the pure isolated compound was determined as sesquiterpene   compound, cadalene-β-carboxylic acid, which exhibited LC50 of 42.32 ppm. Keywords: bawang hutan, Scorodocarpus borneensis, Olaceaeae, brine shrimp lethality test.

Abstract

BTM 11 is unknown species of microalgae, but has active compounds that can inhibit viruses. One of proteins produced by microalgae is a lectin. Lectin is a carbohydrate-binding protein found in various microalgae that show antiviral and antibacterial activity. The purpose of this study was to perform identification of the species of microalgae BTM 11, isolation, characterization, and assay of lectin inhibitory activity. Microalgae BTM 11 was identified to have homology with Cyanobacterium 99% and Geitlerinema sp 98%. Lectin of microalgae BTM 11 was isolated by ammonium sulfate precipitation of 75% with a molecular weight of 17 kDa. Lectin protein activity of microalgae BTM 11 was able to inhibit the enzyme activity of RNA helicase hepatitis C by 57.90% and 27.55%. In addition, the protein was able to suppress the activity of Staphylococcus aureus ATCC 6538, E. coli EPEC K.1.1. and Salmonella typhii ATCC 25241. Activitiy of lectin was stable at 30 °C and was unaffected by the action of the enzyme. These results indicate that lectin of microalgae BTM 11 could be a alternative to antiviral and antibacterial proteins.

Abstract

Dandelion (Taraxacum officinale Weber ex F.H. Wigg) is a medicinal plant species, reported to have some active compounds useful as anti-cancer, anti-inflammatory, antioxidants, and diuretics. Study on in vitro secondary metabolic production and tissue culture of these plants has been reported. This research was aimed to investigate plant regeneration from leaf blade, petiole and root as explants grown in selected media. Three different type of explants were cultured in MS solid medium supplemented with BAP (0, 0.5, and 1.0 mg/l) combined with NAA (0, 0.5, 1.0, and 2.0 mg/l) to select the best medium for spontaneous regeneration. Explants were grown for 6 weeks, the viability of explants, and formation of callus, shoots as well as roots were recorded. The results showed BAP at 1 mg/l combined with at 0.5 mg/l of NAA was suitable for shoot formation, whereas NAA alone was suitable for root formation from leaf blade and petiole. Root was the best explant for shoot regeneration, callus was grown at the first two weeks, and multiple shoots were grown after 3 weeks. No roots were found from root explant. The best medium for multiple shoot regeneration was MS containing 1 mg/l BAP without or with addition of 0.5 mg/l NAA.Keywords: Taraxacum officinale Weber ex F.H.
