Abstract

Control and eradication of a number of infectious diseases are primarily attributed to effective vaccination programs. A concerted effort is still imperative to develop novel vaccines and improve the immunogenicity of existing ones with regards to efficacy, immunogenicity and safety. Rational design of vaccines using subunit vaccines is a potentially safer alternative to conventional vaccines, yet they are poorly immunogenic without additional adjuvant. Using antigen carriers to enhance their immunogenicity in the forms of adsorption or encapsulation with a delivery system has been widely investigated as an alternative to currently available adjuvants. This review aims to elaborate on the existing nanotechnology being used to develop more immunogenic subunit vaccines, with focus on particulate delivery systems for development of prophylactic vaccine candidates.

Abstract

Encapsulation is a method for embryogenic tissue coating system by Na-alginate which has synthetic seed preservation purpose. To produce sugarcane synthetic seed need Coleoptillar embryogenic callus and apical bud that it is encapsulated by encapsulation media (MS0 + 4% Na-alginat+ 100 mM CaCl2). In this study, PEG (0%, 1%, 2%, 3%) and PBZ (0 ppm, 1 ppm, 2 ppm, 3 ppm) add to media encapsulation which are give desiccation effect (PEG) and Giberellin synthesis blockade system. Both of PEG or PBZ is used to sugarcane synthetic seed growth retardation that it is implied as storage period. The result are show that PEG 2% is the most effective concentration which is added in encapsulation media. Dessiccation effect of PEG 2% can make synthetic seed storage period until 3 weeks, synthetic seed regeneration produce 89,32 planlets and planlets clorophill concentration is 26,03 ????g/mL. PBZ 1 ppm is the most effective concentration to make storage period until 5 weeks, synthetic seed regeneration produce 39 planlets and planlets clorophill concentration is 35,81 ????g/mL.

Abstract

Asparagine synthetase (AS) is one of the enzyme involves  in ammonium metabolism in plant, it catalyzes the transfer of the amino group of glutamine to aspartate giving asparagine. Asparagine serves as a major nitrogen transport and storage molecule in many higher plants. This research used legume plant Coronilla rostrata callus culture for primary and secondary metabolism study. On primary metabolism, the culture were used to study the phenomenon of asparagine accumulation. Because of the responsiveness of the culture, we  investigated the use of this culture for model system to conduct molecular biology study of the nitrogen metabolism. The aim of this study was to isolate...cDNAs enconding  Asparagine synthetase  from  C. rostrata because of the asparagine accumulate in this culture.  Several PCR based approaches  were conducted such as RT-PCR, LM-PCR and RACE. Using these methods, two AS cDNAs was  isolated from C. rostrata. AS1 (GenBank no AY081945) which a complete cDNA sequence and AS2 which a partial cDNA sequence.  (GenBank no AF488726). Abstract

Human interferon alpha 2a (hIFNα2a) is a cytokine regulating immune system that has been used in hepatitis and cancer treatments. It has wide biological potency covering antiviral, antiproliferative and immunomodulative activities. This mini review discusses Roferon-A as a prominent commercial product of recombinant hIFNα2a which is produced in bacterial system, Escherichia coli, as therapeutic protein for several diseases, such as chronic viral Hepatitis B, Hepatitis C, melanoma, hairy cell leukemia and renal cell carcinoma. The discussion focuses on the development process with regard to its manufacturing, preclinical and clinical studies, as well as therapeutic efficacy. In addition, we also discuss biosimilar development of hIFNα2a and its potential future developments in the context of enhancing pharmacokinetic profiles

Abstract

The hapten-binding site of the antibody Fab-fragment K411 B specific towards the herbicide atrazine (2-cllloro-4-(ethylamino)-6-(isopropyJamjno)- 1,3,5-triazine) was modified by means of structural modeling and site-directed mutagenesis. A triple mutant (GlnL89GlulValH3711e/GluL3Val) of the Fab-fragment showed an increased affinity towards the hapten HlCVC6 (4-arnino-6-chloro-l ,3,5-triazine-2-(6-aminohexanecarboxylic acid» compared to the affinity ofthe wild-type Fab-fragment lowards the same hapten. However, the mutant exhibited substantially lower affinity towards the hapten H/CVC6 Ihan towards atrazine and the hapten iPr/ CVC6 (4-chloro-6-(isopropylamino)-1,3,5-triazioe-2-(6-aminohexanecarboxylic acid», which is usual ly used in the synthesis of enzyme tracers in ELISA for atrazine. Advantage was taken of the low cross-reactivity and increased affinity of the mutant Fab fragment towards H/CVC6 to improve the sensitivity of a direct-competitive ELISA tor atrazine. HlCIlC6 was covalently conjugated with horseradish peroxidase (HRP), and the conjugate H/CI/C6-HRPwas used as enzyme tracer in the ELISA for atrazine. An eight-fold improvement in sensitivity ofa direct-competitive ELISA for atrazine could be achieved using the tracer H/CVC6-HRP compared to the sensitivity of ELISA using the tracer iPr/CVC6-HRP. The detection limit for atrazine was as low as 0.01 i g/l.
