Ekologia adalah jurnal ilmiah yang diterbitkan untuk mengakomodasi tulisan hasil penelitian bagi sivitas akademika Universitas Pakuan khususnya dan instansi lain di luar Universitas Pakuan pada umumnya. Jurnal ini memuat artikel primer yang bersumber langsung dari hasil penelitian Ilmu Dasar dan Lingkungan Hidup. Ekologia diterbitkan dua kali dalam setahun yaitu pada bulan April dan Oktober oleh Fakultas Matematika dan Ilmu Pengetahuan Alam-Universitas Pakuan. Abstract

Aquaculture of nilem fish needs parent and larvae. The good parent is available from research station with good facilities and management, so it can distribute to farmer by transportation. The important factors that should be notice in transportation process is usage of anaesthetic ingredients to exhilarate fish during transportation process. This research was used nutmeg bean oil contained of miristisin that able to exhilarate. The result of main research showed that the more nutmeg bean oil concentration the more fast nilem fish will be insensible, the lowest concentration (75 mg/l) sensible time of 97,11 minutes while the highest concentration (150 mg/l) showed insensible time of 10,13 minutes. The result of conscious time observation showed that 75 mg/l  resulted conscious time of 17,5 second while 150 mg/l resulted concious time of 740 second. 150 mg/l concentration is the best concentration because it has the fastest insensible time of 10,13 minutes, the longest concious time of 740 second, has hatch value degree and the longest larvae-life and  effective for exhilarate nilem fish. Nutmeg bean oil has influence towards sperm quality, but all of the treatments have the same effect (not significant) towards sperm quality, egg-impregnation degree, egg hatch and nilem fish larvae-life.  Keywords:  nutmeg bean oil,  anaesthetic ingredients in delivery process,  spermatozoa quality for spawning, nilem fish

Abstract

The aim of this study is to compare the potential of ethanolic and water extracted of teki grasstuber on   white  male  rat strain Sprague  Dawley  at  two  months of age and ± 200 g of body weight. The rats were divided among four groups. Each group consists of six rats. The  groupswere ethanolic and water extracted  of  teki  grass tuber, positive control group (Paracetamol) and a negative control group (distilled water). Analgesic potency was tested by inducing pain stimuli previously to the animals, in the form of  heat stimuli  by putting it ona hot plate with  the temperature of 55°C. The responses of rats observed were licking and moving or lifting of their legs. Observations  were made for 1 minute, before  and  after  administration  of  pain stimulus at 15, 30, 45  and 60 minute. The results showed that ethanol and  water  extract  at an effective dose of 49 mg / 200 g of BW, gave relatively the same effect as an analgesic or   positive control group. Keywords : analgesic, ethanol and water extract of teki grass tuber, pain

Abstract

Menopause is a condition in which menstrual cycles stalled due to gland gonadotropin no longer carry out their duties in work affecting the ovaries to produce estrogen. Will halt production of estrogen can lead to osteoporosis (bone loss), kolesterolemia and heart disease. Basil herbs contain chemicals suspected of having estrogenic activity. This study aims to determine the optimum dose estrogenic activity and n-hexane extractof herbs basil (Ocimum americanum L.). animal testing is used 25 female rats were divided into 5 treatment groups. The result showed that all three doses of n-hexane extract of basil herbs have estrogenic activity. The average increase in estrous cycle in groups III, IV and V was 143 hours, 144,6 hours, and 148,6 hours, while the average of scoring against ovarian and uterine vascularitation in group III, IV and V are 2,2; 2 and 2,4. In weighing the ovaries and uterus weights on average in groups III, IV and V are 1,36 g, 1,72 g and 1,7 g. From the above result it can be oncluded that n-hexane extracts of basil herb can cause estrogenic activity with an effective dose of 0,2 g/200g BW. Key words: Basil (Ocimum americanum L.), estrogen wung, n-Heksan

Abstract

Peripheral blood sel mononuclear cell (PBMC) is a white blood cell that can be isolated using the gradient centrifugation method. Ficoll is a reactant which can be used in this method because it produce a high viability i.e. 97.1% ± 1.0%. This research aims to isolate PBMC from healthy human blood. PBMC’s result is used for analysis of dengue virus immune response.Dengue viruses infect PBMC, so going on the secretion of cytokine which is one marker of severity of dengue disease. Expression of cytokines can be measured using quantitative real-time reverse transcription – polymerase chain reaction (qRT-PCR).Venous blood from healthy human is takenaseptically using anticoagulants heparin, then PBMC is isolated with ficoll gradient centrifugation method to produce buffy coat that contain PBMC. Viability of PBMC is calculated using hemositometer with the help of a microscope.Based on the results obtained by PBMC isolation per mL of venous blood that is 2, 08x106 up to 2, 61x106 with an average of 2, i.e. 31x106 withviabilityof 95-99%. These results indicate that this method is quite effective in isolating PBMC from healthy human blood.Keyword : blood, ficoll, centrifugasigradien, PBMCs

Abstract

Inventory of mangroves on the island of Buton, Southeast Sulawesi was conducted in June 2010. The method used in this research is exploratory. Based on the results recorded 21 species of mangroves spread Kakenauwe region. Twelve species of them are included in the category of rare plants refers to the IUCN 2014 which Sonneratia ovata (Near threatened), Acanthus ilicifolius (Least Concern), Acrostichum aureum (Least Concern), Aegiceras corniculatum (Least Concern), Allophylus cobbe (Least Concern, ) Avicennia marina (Least Concern), Ceriops tagal (Least Concern), Cordiasebestena (Least Concern), Nypa fruticans (Least Concern), Pongamia pinnata (Least Concern), Rhizophora mucronata (Least Concern), and Xylocarpus moluccensis (Least Concern) . Ceriops tagal, Rhizophora mucronata, Bruguiera gymnorrhiza and Sonneratia ovata is a species that is often found at that location. Most of the mangrove plant species have the potential, ie as an ornamental plant, traditional medicine, foodstuff, as well as producing high-quality wood.

Abstract

Cystelis® tablet is an expectorant medicine which contain 30 mg/tablet of Ambroxol Hydrochloride. To ensure the safety and effectiveness of the medicine must be well controlled especially the analytical methode which used to determined the actived ingredient in the medicine. This analytical methode must resulting the amount of Ambroxol Hydrochloride which can be trusted. The purpose of this research are to test and proof the result of determination Ambroxol Hydrochloride in Cystelis® tablet could be trusted in daily test in laboratorium. Analytical methode  validation is a effort to get and documented proof that the result of test methode can be trusted with some specification. The Parameters of analytical methode validation are precision,  linearity, accuracy, range, selectivity, and stability test (robustness). Analytical methode that used in pharmacy specially High Performance Liquid Chromatography (HPLC), using mobile phase metanol : buffer potassium dyhidrogen phosphate 0,01 M (70:30) pH 6,0 and stationary phase C18 Symmetry (150 mm x 4,6 mm) with particle size 5mm, flow rate 1,0 mL/minute, injection volume 10,0 mL and measured at wavelength 247 nm. Analytical results showed that the placebo did not gived any analytical respons in selectivity test. Accuracy which know as  % recovery showed the average of % recovery 99,31% (98,70 – 100,44%), range between 80%-120%. Linearity at consentration range 70% - 130% with coefficient correlacy (r)  0,99924. The result of precision which is repeatability with average of % RSD 0,25%.  Based on t value and F value on intermediate precision showed that there is no differently result gived by the analytical methode with different analyst and time. Stability test solution and mobile phase composition with % bias £ ± 2% and %RSD £ 2%. Based on the analytical methode validation result the determination of Ambroxol Hydrochloride in Cystelis® tablet with high performace liquid chromatography we can conclude that this analytical methode is valid to implemented in daily inspection at Quality Control laboratory PT. Abstract

Fish eggs semah in cultivating on average only hatch of 67%, so still far stated well and need to do a proper egg incubation technique to increase the degree of hatching. Efforts are being made at other hatcheries as Catfish is using the technique of egg incubation method of stagnant water and MacDonald jars, but this system still has flaws, that can lead to poor water quality (stagnant water) and the presence of abnormal larvae (Mac Donald jars). In this study, the technique of egg incubation is carried out using a multilevel system tray modification. The results of the use of multilevel system tray in Aquarium shows very different real power against egg hatch (HR) fish semah, whereas against an increase in the degree of abnormality of fish larvae semah influence is relatively the same.The use of 3-floor tray (A3), can enhance degrees most high 89.19% hatching, larval fish and the degree of abnormality 1.63%.                                                                                                      Key words : System tray multilevel, tetas power eggs (HR), the degree of abnormality, Tor douronensis

Abstract

Inventarisasi tumbuhan di kawasan mangrove Desa Wakai dan Desa Taningkola, Kecamatan Una Una, Kabupaten Tojo Una Una, Sulawesi Tengah telah dilakukan. Hasil penelitian mencatat setidaknya 33 jenis tumbuhan ditemukan di kawasan mangrove kedua desa tersebut, dimana 4 jenis diantaranya sudah termasuk langka menurut IUCN tahun 2012, yaitu Intsia bijuga (Colebr.) Kuntze, Acrostichum aureum L, Dolichandrone spathacea (L.f.) Seem. dan Lumnitzera littorea (Jack) Voigt. Bruguiera gymnorhiza (L.) Lam. dan Lumnitzera littorea (Jack) Voigt adalah jenis yang sering ditemukan di kedua lokasi. Abstract

Buah semangka merah (Citrullus vulgaris rubrum) dan buah semangka kuning (Citrullus vulgaris flavum) merupakan tanaman obat yang berkhasiat sebagai peluruh air seni dan digunakan untuk melarutkan batu ginjal. Telah dilakukan pengujian potensi sari buah semangka merah dan sari buah semangka kuning sebagai peluruh batu ginjal kalsium oksalat secara in vitro. Pengujian dilakukan dengan merendam batu ginjal kalsium oksalat dan menganalisis kadar kalium serta kalsium oksalat yang larut secara Spektrofotometer Serapan Atom. Hasil penelitian menunjukkan bahwa sari buah semangka merah dan sari buah semangka kuning dapat melarutkan batu ginjal kalsium oksalat. Kelarutan batu ginjal kalsium oksalat tertinggi dalam sari buah semangka kuning pada konsentrasi 100% sebesar 16,25 x 10-5 mol/L sedangkan pada sari buah semangka merah pada konsentrasi 100% sebesar 10,83 x 10-5 mol/L. Kadar kalium diduga menjadi faktor yang mempengaruhi kelarutan batu ginjal kalsium oksalat. Kandungan kalium semangka kuning sebesar 0,02454% lebih tinggi dibandingkan dalam semangka merah sebesar 0,02093%. Kata kunci  : Semangka merah (Citrullus vulgaris rubrum), Semangka kuning (Citrullus vulgaris flavum), Kalsium oksalat, Batu ginjal

Abstract

PT. Aneka Tambang has reclaimed land due to gold production activities.  Reclamation land is a process whereby the damaged land is repaired to its natural state.  Indicators of succession can be showed in the presence of plants and animals.  Birds are a good indicator for assessing biodiversity in a region, because can occupy a large habitat of  near the top of the food chain.  The spread of plant seeds can be done by birds and mammals through its feces. Identification and observation of bird and mammal diversity can be used to improve the conservation of birds and mammals in the area.  The purpose of this research was  to study the diversity of birds and mammals.  The method used were Indices Ponctuels d’Abondance(IPA) method for birdwatching and transect lines for mammalian observation.  Result showed  33 speciesof birds and 9 species of mammals were found in this study. The bird species was dominated by the Cucak Kutilang type and the mammal was dominated by Squirrel Kekes.Key words : PT. Aneka Tambang, diversity ,bird ,mammal
