Abstract

Strain of BL5 was reported as a mannanase producer. The purposes of this study are identification, purification and characterization of mannanase from BL5. The Internal Transcribed Spacer (ITS) regions analysis showed that BL5 strain have 93% similarity with Aspergillus ustus isolate UOA/HCPF 9236. An extracellular mannanase from the culture supernatant of a fungus A. ustus BL5 was purified. SDS-PAGE of the purified enzyme showed a single protein band of molecular mass 50 to 51 kDa. The mannanase exhibited optimum catalytic activity at pH 7.0 and 55C. The metal ions Ca2+, Cu2+ and SDS inhibited complete enzyme activity. The metal ion Mg2+ and EDTA increased complete enzyme activity. The value of Vmax = 5,88 ?mol mannose/min/ml and Kmax = 0.64 mg/ml. Mannanase of A. ustus BL5 be able to hydrolyzed porang mannan.

Abstract

Sago solid waste as by product result of sago palm processing potentially have great value for some bio processing such as source of carbon on microbes substrate. Utilization of sago solid waste as substrate for filamentous cyanobacteria cultivation is a rareresearch activity in Indonesia as one of the three leading world companies producer of sago starch. Therefore, we focused on sago waste treatment using cyanobacteria agent in order to rid the waste and gain the value of nutrition rich starch from that waste. Several concentrations of growth media contain sago solid waste were tested into cyanobacteria culture and solid waste hydrolysis as pretreatment more conducted in the research. Result shows that addition of sago solid waste (50, 500 and 5000 ppm) into media can increase the algae biomass production which is rich of carbohydrate and protein.The proximate value of biomasses with sago solid waste treatment was exhibited significantly higher than control. During cultivation exposure time, the solid waste is gradually consumed by the algae. The study implies that sago solid waste can be used as substrate for the biomass production of filamentous cyanobacteria which is known as source of active compounds, potential resource for valuable bioproduct.

Abstract

Manganese (Mn) has a very important role in various industrial fields such as iron, steel, food, agriculture and chemical industry. High grade Mn ore(>40%) are generally processed through the pyrometallurgy process, whereas for low grade Mn ore (<40%) is processed through the hydrometallurgical treatment process. This study aims to characterize the Mnore in Tanggamus and Way Kanan Regency Lampung Province and prospective process. Experiment begins with a sampling of the area in Tanggamus and Way Kanan mine. Sample preparation is then performed by reducing the particle size of the ore through the stages of crushing, grinding and particle size distribution. Mn ore sample which has been reduced then performed characterization using X-Ray Fluorescence (XRF), X-Ray Diffraction (XRD) and Scanning Electron Microscope (SEM). The results showed that the characteristics of Mn ore in Tanggamus and Way Kanan mine was different. Mn ore Tanggamus have the highest contentof 33.84% Mn associated with Fe2O3 minerals, while Mn ore Way Kanan had content 18.03% associated with SiO2 minerals. Both Mn ore are included in low grade Mn ore (<40). Prospective processing of manganese ore is initiated by benefitiation and proceed by leaching method.

Abstract

LIPI Press official website, penerbit.lipi.go.id, provides hundreds of data catalogs of scientific publications. When referring to the site as a provider of publications catalog, the website has weakness and do not follow thedevelopment of web technology. In this paper, analysis of the website was conducted by using eight (8) web analyzer namely WooRank, Seoptimer, Lipperhey, iWebChk, SEO Workers, Sailor SEO, SEO Site Checkup, and SEOAnalyzer. The results showed that the six web analyzers provide low valuation on the site and only 2 other web analyzers provide medium valuation.

Abstract

Research of the influences of temperature and length of storage on the viability of gayam (Inocarpus edible) seeds was carried out at Macropropagation Laboratory, Botany Devision, Research Centre for Biology, Cibinong Science Center. The research was using Randomized Block Design with 3 factors and 3 replications, in which each replication had 5 samples. The first factor was length of storage with 4 levels i.e. 0 (before storage), 1, 2 and 4 weeks. The second factor was temperature of storage with 2 levels i.e. 20C and 26C. The third factor was treatment of seeds with 2 levels of unpeeled (TKPS) and peeled (KPS). The result showed that during the storage, moisture content decreased, value of ion leakage increased and gayam seeds viability decreased. The storage at a temperature of 20C was better than 26C, in which the values of ion leakage were 25599.04 and 34662.85 ?Scm-1, with the seed viability of 83.33 and 80.41%, respectively. Meanwhile the viability of unpeeled seed was better then the peeled one, the viability of unpeeled and peeled seeds were 87.08a, and 76.66b %, respectively. Gayam seeds could be stored up to 4 weeks, at 20C unpeeled seed with a high value of viability (86.66ab %). The research on the water sorption of gayam seeds is necessary to determine the appropriate moisture content of gayam seed storage.
