Abstract

Cocoa peels and coffee peels as food industry have low economic value but have great potential because of the high levels of pectin in them. This research used microwave assistant extraction (MAE) method from kako peel and coffee peel waste and has assessed their potential as a source of raw material for capsule shells. This research included observing the optimization of pectin extraction techniques, standardizing pectin, and observing the quality of the resulting capsule shells. The pectin used comes from cocoa peel and coffee peel waste in West Java. The test results showed that the quality of the pectin obtained from cocoa shells and coffee husks complied with IPPA (International Pectin Produce Association) standards so that these pectins could be used for industrial purposes. While the capsule shells obtained have met the organoleptic standards and the disintegration time of the capsule shells according to the Indonesian Pharmacopoeia. Some of the parameters that meet these standards were the size of the capsule shell, both the uniformity of the size of the capsule cap, the uniformity of the size of the capsule body and the minimum disintegration time. Meanwhile, the weight uniformity standards have not met the requirements. The coffee peel capsule shells also meet the Indonesian Pharmacopoeia standards in terms of uniformity of capsule cap size, uniformity of capsule body size, minimum disintegration time, and uniformity of weight. In addition, capsule shells made from cocoa peel and coffee peel meet the requirements for the characteristics of good capsule shell materials where the results of the elongation strength and tensile strength tests meet the quality requirements. So it can be seen that the pectin from cocoa and coffee peels waste have good character to be developed as a source of pectin for pharmaceutical raw materials with the addition of other excipients, such as CMC-Na and carrageenan

Abstract

Black garlic is the result of processing garlic through a heating process at a temperature of 70oC with a relative humidity of 70-80% for 21 days without additional treatment so that the water content decreases. Black garlic has antibacterial activity due to the presence of bioactive compounds from the phenolic and flavonoid groups. This study aims to determine the secondary metabolite content and antibacterial activity of the black garlic extract fraction against Propionibacterium acnes bacteria. Extraction of Black garlic by maceration method used 96% ethanol as solvent and to obtain extract fraction of Black garlic by multilevel fractionation method using n-hexane, ethyl acetate, and ethanol as solvents. To determine the content of secondary metabolites, phytochemical screening was carried out in each fraction. Antibacterial activity test of Propionibacerium acnes used disc diffusion method with various concentrations of n-hexane, ethyl acetate, ethanol fractions 2%, 4%, 6%, 8%, 10%, positive control (clindamycin 1%), negative control and media control. Data analysis was carried out with qualitative, quantitative, and statistical analysis. The results obtained were the extract fraction of black garlic contains secondary metabolites of flavonoids, alkaloids, saponins, and tannins. The results of the antibacterial activity test showed that the garlic extract fraction had antibacterial activity seen from the presence of an inhibition zone. The largest inhibition zone in the n-hexane fraction of 10% concentration of 8.50 ± 0.10 mm was included in the medium category. The positive control was 12.02 ± 1.41 mm and the negative control gave no inhibition zone.

Abstract

Chitosan has many functions in the health sector, one of which can be used as an anti cholesterol supplement. Co-processing is a technique carried out to obtain new additives by combining two or more existing additives, where the combinations of these materials will complement each other, so that new additives with better (superior) properties are obtained. The purpose of this study was to determine the formulation and evaluation of mud crab shell (Scylla serrata) chitosan tablets from co-processed excipients with PEG 4000 as a meltable binder with a ratio of 10% (F1) and 20% (F2). Co-processed excipients are made using the melt granulation method with additional ingredients, namely lactose as a filler and primogel as a disintegrant. Tablets were printed using the direct compression method with the addition of Mg Stearate as a lubricant. The results of the co-processed excipient evaluation showed that each granule met the requirements for the flow rate test, angle of repose test, compressibility test and Hausner's ratio. From the results of statistical tests using the Mann-whitney test method, there is a significant difference between co-processed excipient PEG 4000 10% and PEG 4000 20% as a meltable binder for the parameters of water content with F2 moisture content (1.67%) lower than in F1 (3.33%). In the evaluation of chitosan tablets, they produced tablets that met the test requirements for weight uniformity, tablet friability (F1), tablet hardness, disintegration time (F2) and did not meet the requirements for the tablet size uniformity test. The selected formula for this study was F2 with a PEG 4000 meltable binder of 20% because it produced tablets that met disintegration time with low water content.
