Abstract

Ciprofloxacin, a widely used fluoroquinolone for the treatment of bacterial infections, has been associated with adverse effects on male reproductive function. This study investigated the protective effects of ascorbic acid on semen quality and testicular histomorphology in male Wistar rats treated with ciprofloxacin. Twenty healthy adults male Wistar rats (100-150 g) were randomly assigned to four groups (n = 5 per group): Group A received distilled water, Group B received ciprofloxacin (135 mg/kg), Group C received ciprofloxacin (135 mg/kg) plus vitamin C (200 mg/kg), and Group D received vitamin C alone (200 mg/kg). After acclimatization, the test substances were administered orally for 14 days. The animals were euthanized 24 hours after the final treatment to evaluate sperm motility, viability, and morphology, as well as testicular histomorphology. Administration of ciprofloxacin alone caused a significant decrease (p <0.05) in sperm motility and viability, accompanied by a marked increase in sperm morphological abnormalities. Testicular histoarchitecture also revealed severe damage, characterized by interstitial oedema, thinning and disorganization of the germinal epithelium, vacuolation of spermatogenic cells, and the absence of mature spermatozoa within the seminiferous tubules. Co-administration of ascorbic acid significantly ameliorated these toxic effects, resulting in reduced sperm abnormalities, improved sperm parameters, and restoration of normal testicular histoarchitecture with active spermatogenesis. In addition, vitamin C alone significantly improved normal sperm morphology compared with the distilled water-treated control group. This study suggests that ascorbic acid supplementation may mitigate ciprofloxacin-induced reproductive dysfunction.

Abstract

The offspring sex ratio in mammals is generally considered to occur by chance, with many reports indicating an approximately 1:1 (male:female) ratio. The ability to influence this natural biological ratio could provide significant socioeconomic benefits in livestock production. Although sex-sorted semen offers a means of predetermining offspring sex, its high cost and reduced fertility following processing limit its accessibility to many livestock farmers in developing countries. Therefore, identifying a simpler and more affordable alternative would be highly beneficial. A total number of twenty-one goats were used for this experiment, comprising one Boer buck and 20 West African Dwarf (WAD) goat does weigh between 25 and 35 kg.  Estrus was synchronized using two intramuscular injections of 250 µg Synchromate® administered 11 days apart. Semen was collected from the buck using an electroejaculator (EE) after restraint, evaluated, and extended with a standard egg yolk sodium citrate extender. Each doe was artificially inseminated twice. Before each insemination vaginal pH was measured using a digital pH meter and recorded. Based on the vaginal pH at insemination, seven does were inseminated under acidic conditions, four under neutral conditions, and nine under alkaline conditions. All twenty (20) inseminated does gave birth to a total of 33 kids, comprising 15 females and 18 males. Further studies are needed to investigate the mechanisms responsible for changes in vaginal pH during estrus and to determine whether manipulation of vaginal pH could provide a practical approach to influencing offspring sex ratio in goats.

Abstract

The semen quality of exotic boar breeds introduced into Cameroon has been poorly assessed, and the characteristics of their chilled spermatozoa remain largely unknown. In artificial insemination (AI) programs, regular monitoring of semen quality is essential to maintain high reproductive performance. This study aimed to evaluate the response of semen from Landrace, Large White, and Duroc boars to chilled storage. Adult boars weighing 145-194.3 kg were housed individually in standard finishing pens (12.96 m²) equipped with a feeder and drinker. Semen was collected, chilled, and evaluated at 0, 24, 48, and 72 hours after collection. Analysis of fresh semen showed that sperm concentration was significantly higher (p < 0.05) in Duroc (218.6 ± 18.54 × 10⁶spz/mL) and Large White (200.6 ± 19.07 × 10⁶spz/mL) than in Landrace (149.2 ± 26.5 × 10⁶spz/mL), whereas semen volume showed the opposite trend. After 72 hours of storage, only Duroc semen exhibited significant reductions (p < 0.05) in the percentage of normal sperm cells (NSC), sperm viability, sperm membrane integrity, and seminal pH. In contrast, sperm kinematic parameters (TM, PM, VAP, VSL, VCL, and LIN) declined significantly (p < 0.05) with storage time in Landrace and Large White semen. In conclusion, semen from Landrace and Large White boars showed greater tolerance to chilled storage than semen from Duroc boars.

Abstract

Understanding the factors that influence dairy farmers’ awareness of infertility and reproductive performance is critical for improving productivity. This cross-sectional survey sampled farmers across six Tanzanian regions (Njombe, Mbeya, Tanga, Kilimanjaro, Arusha, and Morogoro) and used multivariable analyses to examine associations between 13 predictors and four outcomes: recognition of infertility signs, recognition that failing to produce a calf annually indicates infertility, pregnancy status, and calving interval. Key findings showed that smaller herd sizes were associated with poorer recognition of infertility signs and lower pregnancy rates, while less experienced farmers surprisingly demonstrated better recognition of infertility signs and higher pregnancy rates. Farmers who regarded repeat breeding as a major or moderate constraint had shorter calving intervals than those who considered it a negligible constraint. Farmers relying exclusively on artificial insemination (AI) were more likely to identify failure to produce a calf annually as an indicator of infertility than those using both AI and natural service. Aside from region, other predictors showed no clear or consistent associations in either the univariable or multivariable models. This lack of detectable association does not imply the absence of an effect but rather indicates that the data could not distinguish between positive, negative, or null associations. Region emerged as the most consistent and influential predictor, showing significant associations with at least one category of each outcome variable. Overall, the results highlight the need for context-specific, regionally tailored interventions to improve reproductive performance in Tanzania's smallholder dairy systems.

Abstract

This study evaluated the reproductive performance of Dorper sheep in an intensive system in West Java, Indonesia. Data included ewe parity (P1 and P2), lambing interval (LI), litter size at birth (LSB), weaning litter size (WLS), age at first lambing (AFL), annual reproductive rate (ARR), and offspring sex ratio. Data analysis was performed using the Kruskal–Wallis and Mann–Whitney tests. The strength of the correlation was interpreted based on the correlation coefficient (r), where values ​​close to 0 indicate a weak relationship, while values ​​close to ±1 indicate a strong relationship. Statistical significance was set at P < 0.05. The results showed a stable ARR value of 1.64 across genders, confirming the success of the accelerated lambing system (3 lambing in 2 years) with an average lambing interval of 259–265 days. There was an increase in the number of male lambs, reaching 60% by 2025, which correlated with the improved pre-conception nutritional status of the ewe. Parity did not affect the number of offspring per litter but demonstrated good stability in the trophic environment. Spearman’s correlation analysis showed that the relationships between reproductive parameters tended to be weak and insignificant, indicating that reproductive traits were relatively independent. In conclusion, an intensive rearing system can overcome tropical climate challenges and maximize the genetic potential of Dorper sheep. An ARR of 1.64 indicates good reproductive performance under the evaluated production conditions. Although a higher proportion of male offspring may be advantageous in meat-oriented production systems, further studies evaluating both the biological determinants of offspring sex ratio and its economic impact are needed before management recommendations can be made.

Abstract

Heat stress severely affects the redox balance of poultry semen, as the limited supply of natural antioxidants is unable to neutralize the excessive production of free radicals. This oxidative stress can reduce sperm quality and reproductive productivity in Japanese quails during summer. Dietary supplementation may provide a cost-effective approach to maintaining semen quality under such conditions. The present study was conducted to assess the effects of dietary vitamin C on semen volume, antioxidant activity, sperm concentration, motility, viability, plasma membrane integrity, and acrosomal and DNA integrity over four weeks. Thirty male Japanese quails were maintained at an ambient temperature of 35-40 °C (May-June), divided into five groups, and supplemented with 40, 80, 150, or 170 mg vitamin C/kg diet, with a control group, for four weeks. Semen was collected weekly by manual massage, and sperm parameters were evaluated using three replicates per group per week. Vitamin C supplementation did not significantly affect semen volume (p> 0.05). However, semen quality parameters and antioxidant activity increased significantly (p< 0.05) up to the third week, with no further increase in the fourth week. The 150 mg/kg treatment showed the highest sperm concentration, motility, viability, plasma membrane integrity, acrosomal integrity, DNA integrity, and antioxidant activity at week three, with these values maintained in the fourth week. Thus, dietary vitamin C at 150 mg/kg may help maintain semen quality and antioxidant activity in Japanese quails during summer heat stress.

Abstract

Parity affects metabolic status and reproductive performance in female livestock. Since metabolic status can influence ovarian function and fertility, assessing serum biochemical profiles during estrus synchronization may help predict reproductive performance. This study assessed parity-related changes in serum biochemistry, estrus response, and conception rate following double prostaglandin F2α (PGF2α) estrus synchronization in Ettawa grade goats. A total of 24 goats were divided into three parity groups: low parity (0-1), medium parity (2-3), and high parity (4-5). All goats received two PGF2α injections at an 11-day interval. Serum samples were collected one day before the first injection (Day 0) and one day after the second injection (Day 12) to measure glucose, triglyceride, and cholesterol levels. The goats that exhibited estrus underwent double artificial insemination at a 12-hour interval, and conception was confirmed by ultrasonography 45 days after insemination. Following synchronization, the medium- and high-parity groups showed increased serum glucose levels (p< 0.05), while serum cholesterol levels were also increased in the medium- and high-parity groups (p< 0.05). After synchronization, serum triglyceride levels decreased in all groups (p< 0.05). The estrus response in the low-parity group was 12.5% lower than that in the medium- and high-parity groups; however, this difference was not statistically significant. The conception rate was over 80% in all groups. In conclusion, double PGF2α estrus synchronization resulted in parity-related changes in serum biochemistry, characterized by increased glucose and cholesterol levels and decreased triglyceride levels, while no significant differences in estrus response or conception rate were detected among the parity groups.
