Abstract

Abstract. Coriandrum sativum L. (coriander) seeds are widely recognized as a medicinal herb with various pharmacological activities and considerable potential for pharmaceutical development. However, information regarding their safety, particularly their effects on embryonic development, remains limited, necessitating an acute toxicity evaluation. This study aimed to assess the acute toxicity of the ethanolic extract of Coriandrum sativum L. seeds on zebrafish (Danio rerio) embryos and to determine the median lethal concentration (LC₅₀) as an indicator of toxicity. The study employed a true experimental design using the Fish Embryo Toxicity Test (FET) in accordance with the OECD Guideline 236. Zebrafish embryos were exposed to extract concentrations of 0.1, 1, 10, 100, and 1000 µg/mL for 96 hours. Embryo mortality and developmental abnormalities were analyzed using probit regression to determine the LC₅₀ value. The results demonstrated that the ethanolic extract induced embryonic abnormalities, including coagulation, delayed hatching, absence of somite formation, and lack of heartbeat. The LC₅₀ value was 18.7393 µg/mL, indicating that the extract is classified as slightly toxic. These findings suggest that the ethanolic extract of Coriandrum sativum L. seeds exhibits acute toxicity toward zebrafish embryos; therefore, its safety should be carefully considered before further development as a medicinal agent. Abstrak. Biji ketumbar (Coriandrum sativum L.) merupakan tanaman herbal yang memiliki berbagai aktivitas farmakologis dan berpotensi dikembangkan sebagai bahan obat. Namun, informasi mengenai keamanan penggunaannya, khususnya terhadap perkembangan embrio, masih terbatas sehingga diperlukan uji toksisitas akut. Penelitian ini bertujuan untuk mengevaluasi toksisitas akut ekstrak etanol biji ketumbar (Coriandrum sativum L.) terhadap embrio ikan zebra (Danio rerio) serta menentukan nilai LC₅₀ sebagai indikator tingkat toksisitas. Penelitian menggunakan metode eksperimental (true experimental design) dengan pendekatan Fish Embryo Toxicity Test (FET) mengacu pada pedoman OECD 236. Embrio ikan zebra dipaparkan pada ekstrak dengan konsentrasi 0,1; 1; 10; 100; dan 1000 µg/mL selama 96 jam. Data mortalitas dan abnormalitas embrio dianalisis menggunakan regresi probit untuk menentukan nilai LC₅₀. Hasil penelitian menunjukkan bahwa ekstrak etanol biji ketumbar menyebabkan abnormalitas berupa koagulasi embrio, keterlambatan penetasan, tidak terbentuknya somit, dan tidak adanya denyut jantung. Nilai LC₅₀ yang diperoleh sebesar 18,7393 µg/mL, yang termasuk dalam kategori slightly toxic. Dengan demikian, ekstrak etanol biji ketumbar memiliki toksisitas akut terhadap embrio ikan zebra sehingga aspek keamanannya perlu diperhatikan sebelum dikembangkan lebih lanjut sebagai bahan obat.
