Abstract

Abstract. Leunca leaf  is one of the medicinal plants used in traditional medicine because it contains bioactive compounds that have the potential to provide pharmacological effects, such as anti-inflammatory, antibacterial, antioxidant, and anticancer. However, data on the safety of its use for embryonic development are still limited, so it is necessary to carry out acute toxicity testing. This study aims to evaluate the effect of acute toxicity of ethanol extract of leunca leaf on zebrafish embryos and determine the LC₅₀ value produced. The research method used is true experimental design with the FET method referring to OECD guidelines No. 236 of 2013. A total of 120 zebrafish embryos were divided into negative control group and treatment groups, each group consisting of 20 embryos with an extract concentration of 0,1; 1; 10; 100; and 1000 ppm every 24 hours for 96 hours. Embryonic mortality and abnormality data were analyzed using probit regression to obtain LC₅₀ values. The results showed that exposure to ethanol extract of leunca leaves caused death and abnormalities in embryonic development, including embryonic coagulation, disturbance of somite formation, and non-formation of heart rate. The LC₅₀ value obtained for 96 hours of 0.2329 ppm is classified as Highly Toxic. Abstrak. Daun leunca merupakan salah satu tanaman obat yang dimanfaatkan dalam pengobatan tradisional karena mengandung senyawa bioaktif yang berpotensi memberikan efek farmakologis, seperti antiinflamasi, antibakteri, antioksidan, dan antikanker. Namun, data mengenai keamanan penggunaannya terhadap perkembangan embrio masih terbatas sehingga perlu dilakukan pengujian toksisitas akut. Penelitian ini bertujuan untuk mengevaluasi efek toksisitas akut ekstrak etanol daun leunca terhadap embrio ikan zebra serta menentukan nilai LC₅₀ yang dihasilkan. Metode penelitian yang digunakan adalah true experimental design dengan metode FET mengacu pada pedoman OECD No. 236 Tahun 2013. Sebanyak 120 embrio ikan zebra dibagi menjadi kelompok kontrol negatif dan kelompok perlakuan yang masing-masing kelompok terdiri dari 20 embrio dengan konsentrasi ekstrak 0,1; 1; 10; 100; dan 1000 ppm setiap 24 jam selama 96 jam. Data mortalitas dan abnormalitas embrio dianalisis menggunakan regresi probit untuk memperoleh nilai LC₅₀. Hasil penelitian menunjukkan bahwa paparan ekstrak etanol daun leunca menyebabkan kematian dan kelainan perkembangan embrio, antara lain koagulasi embrio, gangguan pembentukan somit, dan tidak terbentuknya detak jantung. Nilai LC₅₀ yang diperoleh selama 96 jam sebesar 0,2329 ppm termasuk dalam kategori Highly Toxic (sangat toksik).
