Abstract

Abstract. Acne vulgaris is a chronic inflammatory disorder of the pilosebaceous unit associated with the proliferation of Cutibacterium acnes. Tea tree oil (TTO) derived from Melaleuca alternifolia exhibits antibacterial and anti-inflammatory properties; however, its application is limited due to volatility and poor stability. This study aimed to develop a chitosan-based TTO nanoemulsion patch and evaluate its physicochemical properties, stability, and antibacterial activity against C. acnes. The research was conducted experimentally through TTO characterization using GC-MS, nanoemulsion preparation with Tween 80 and propylene glycol, optimization using a 2² factorial design, and patch preparation using the solvent casting method. The optimized nanoemulsion was obtained using 25% Tween 80 and 25% propylene glycol, resulting in a transmittance value of 99.629%. The nanoemulsion showed favorable characteristics, including a particle size of 11.6 ± 0.265 nm, PDI of 0.048 ± 0.039, pH of 6.44 ± 0.0794, viscosity of 79.167 ± 0.971 cPs, pseudoplastic flow behavior, and good thermodynamic stability. The best patch formulation was Formula 3 containing 10% gelatin, based on physical evaluation and stability assessment. However, the TTO nanoemulsion patch did not exhibit antibacterial activity against C. acnes, indicating that the active compound release from the patch matrix was not yet optimal. Further studies on drug release profiles are required to improve the effectiveness of the topical delivery system. Abstrak. Jerawat (Acne vulgaris) merupakan penyakit inflamasi kronis pada unit pilosebasea yang melibatkan proliferasi Cutibacterium acnes. Tea tree oil (TTO) dari Melaleuca alternifolia memiliki potensi sebagai antibakteri dan antiinflamasi, tetapi penggunaannya terbatas karena sifat volatil dan stabilitas yang rendah. Penelitian ini bertujuan untuk mengembangkan patch nanoemulsi TTO berbasis kitosan serta mengevaluasi karakteristik fisik, stabilitas, dan aktivitas antibakterinya terhadap C. acnes. Penelitian dilakukan secara eksperimental melalui karakterisasi TTO menggunakan GC-MS, formulasi nanoemulsi dengan Tween 80 dan propilen glikol, optimasi menggunakan desain faktorial 2², serta pembuatan patch dengan metode solvent casting. Hasil penelitian menunjukkan formula optimum nanoemulsi diperoleh pada konsentrasi Tween 80 25% dan propilen glikol 25% dengan nilai transmitan 99,629%. Nanoemulsi memiliki karakteristik fisik yang baik dengan ukuran partikel 11,6 ± 0,265 nm, PDI 0,048 ± 0,039, pH 6,44 ± 0,0794, viskositas 79,167 ± 0,971 cPs, sifat alir pseudoplastis, serta stabil secara termodinamika. Formula terbaik patch diperoleh pada Formula 3 dengan kandungan gelatin 10% berdasarkan evaluasi mutu fisik dan stabilitas. Namun, patch nanoemulsi TTO belum menunjukkan aktivitas antibakteri terhadap C. acnes karena belum terjadi pelepasan zat aktif yang optimal dari matriks patch. Penelitian lanjutan mengenai profil pelepasan zat aktif diperlukan untuk meningkatkan efektivitas sistem penghantaran.
