Abstract

Abstract. In Indonesia, helminth infection prevalence ranges from 2.5% to 62%, driven by tropical climate, high population density, and low hygiene awareness. This study aimed to evaluate the anthelmintic activity of Jatropha curcas L. leaf extracts and fractions against adult pig roundworms (Ascaris suum), and to determine the PC50 and LC50 values. An experimental laboratory study used negative controls (0.5% CMC-Na), positive controls (pyrantel pamoate and piperazine citrate), and test groups treated with ethanol leaf extract (5%,10%,15%,20% and 25% w/v), an ethanol:water fraction (1%, 2%,4% and 8% w/v), and n-hexane and ethyl acetate fractions (1% and 2% w/v). Results showed the ethanol extract possessed anthelmintic activity, producing spastic-type paralysis. The 10% extract induced 50% paralysis, while 20% and 25% caused 60% mortality; the 8% ethanol:water fraction induced 40% paralysis. PC50 values for male and female worms were 9.0240% and 7.4542%; LC50 values were 27.3401% and 21.3944%. The fraction gave PC50 values of 43.2713% male and 14.1840% female. Abstrak. Di Indonesia, prevalensi infeksi cacing berkisar antara 2,5% hingga 62%, yang dipengaruhi oleh iklim tropis, kepadatan penduduk yang tinggi, dan rendahnya kesadaran akan kebersihan. Penelitian ini bertujuan untuk mengevaluasi aktivitas antelmintik ekstrak serta fraksi daun Jatropha curcas L. terhadap cacing Ascaris suum, serta menentukan nilai PC50 dan LC50. Penelitian eksperimental laboratorium ini menggunakan kontrol negatif (CMC-Na 0,5%), kontrol positif (pirantel pamoat dan piperazin sitrat), serta kelompok uji yang diberi perlakuan berupa ekstrak etanol daun (5%,10%,15%,20% dan 25% b/v), fraksi etanol-air (1%, 2%,4% and 8% b/v), serta fraksi n-heksana dan etil asetat (1% dan 2% b/v). Hasil menunjukkan ekstrak etanol memiliki aktivitas antelmintik dengan paralisis tipe spastik. Konsentrasi 10% menyebabkan paralisis 50%, sedangkan 20% dan 25% menyebabkan mortalitas 60%; fraksi etanol-air 8% menyebabkan paralisis 40%. Nilai PC50 cacing jantan dan betina masing-masing 9,0240% dan 7,4542%; nilai LC50 masing-masing 27,3401% dan 21,3944%. Fraksi etanol-air menghasilkan PC50 sebesar 43,2713% (cacing jantan) dan 14,1840% (cacing betina).
