Abstract

Background: Typhoid fever caused by Salmonella Typhi remains as an infectious disease that still becomes a health problem, especially in developing countries due to its fairly high morbidity. Vaccine development efforts, particularly about proteins that play a role in enhancing immunity, are adhesin protein (Protein AdhO36) and the addition of immunoadjuvant Beta Glucan that can improve the cellular immune system.Objective: This study was aimed to predict the ligand-receptor bonds (Dectin1-Beta Glucan; TLR2-Beta Glucan, TLR4-Beta Glucan) and the ligand-receptor bonds (Dectin1-AdhO36; TLR2-AdhO36, TLR4-AdhO36).Method: To determine the binding affinity of the bond between Beta-Glucan and Dectin1, TLR2, and TLR4, a molecular approach was specifically used using PyRx 0.8. Whereas to find out the interaction between Adh036 and target proteins Dectin1, TLR2, and TLR4, Patchdock software was used. The results of the interaction would be visualized using PyMol software. Amino acids involved in docking interactions were identified using KFC protein hotspots.Results and Conclusion: The result indicated  the binding affinity between receptor Dectin1-ligand Beta Glucan and Beta Glucan TLR2  was (-6.5 kcal/mol), and TLR4-Adh036 shown the highest interaction strength with global energy -1462.6.  This results shown the potential effect of AdhO36 and Beta Glucan combinations  enhanced cellular immunity against Salmonella Typhi infection.Keywords: AdhO36, Beta Glucan, TLR2, TLR 4, Dectin-1. Abstract

Background: Fetuin-A (FetA) is a 63 kD phosphorylated glycoprotein that relates to insulin resistance inhibits autophosphorylation and the activity of insulin receptor tyrosine kinase (IR-TK) at the receptor levels. FetA positively correlates with insulin levels, and homeostasis model of insulin resistance (HOMA-IR). However, the phenomenon is obtained from several studies; a decrease did not always follow the increase of FetA in glucose uptake in the tissue. Aim: To determine the impact of Fetuin-A and Palmitate on TLR4 mediated Glucose Transporter-4 translocation inhibition in normal glucose-tolerance human skeletal muscle cell (h-SkMC). Method: Normal h-SkMC culture were randomly divided into five arms and treated with human- insulin, and deoxy-glucose (GI arm), or human-insulin, deoxy-glucose, and FetuinA (GIF arm), or human-insulin, deoxy-glucose and Palmitate (GIP arm), or human-insulin, deoxyglucose, FetuinA and Palmitate (GIFP arm). The control arm was without any treatment. The effects of insulin, glucose, FetuinA, and Palmitate on the expression of TLR4, Akt, GLUT-4 were evaluated by immunofluorescence microscopy. The expression of TNF- and IL-6 were measured by ELISA assay, and glucose uptake was measured by spectrophotometry assay. The results were obtained and subject to comparative test and pathway analysis. Results: The treatment with glucose+insulin (GI), FetA (GIF), Palmitate (GIP), and Palmitate+FetA (GIFP) increased TLR4 expression significantly compared to control arm (p<0.05) and only treatment with GI and GIF significantly increased the expression of GLUT-4 compared to control arm (p<0.05). All treatment arms and control arms were not significantly different in altering glucose transport. The treatment with Palmitate significantly decreased GLUT-4 expression and independence of TLR4 expression. Conclusion: The treatment with FetA alone or in combination with Palmitate in normal h-SkMC significantly increases the expression of TLR4 but does not alter GLUT-4 translocation and glucose transport. These results provide novel evidence indicating that acute exposure of FetuinA and Palmitate on normal glucose-tolerance h-SkMC does not induce insulin resistance.Keywords: FetA, Palmitate, TLR4, GLUT-4, TNFα, IL-6, Glucose transport.

Abstract

Perilla is an edible plant and has medicinal properties. The leaves have a very pleasant sweet taste and are used as a spice. Perilla seed and oil are rich sources of phenolic compounds, antioxidants, polyunsaturated fatty acids (α-linolenic acid, linoleic acid, oleic acid, and palmitic acid, omega-3 and omega-6 polyunsaturated fatty acids) and minerals. Perilla is commonly used to flavor the foods. Objective of the present study was focused on the effect of blanching, drying and storage of herbal tea produced from Perilla. Optimal results found that herbal tea would have stable antioxidant capacity while blanching at 95oC in 4 seconds with the presence of 4% CaCl2 submersed in blaching solution; drying at 40oC with 2.0 cm in dimenstion of Perilla; storage under vaccum in PET/AL/PE bag within 6 months without any deterioration.Keywords: Perilla, Herbal tea, Blanching, Drying, Storage, vacuum.

Abstract

The present study deals with the investigation of serum TNF-α, IL-6, brain derived neurotrophic factor (BDNF), monocyte chemo-attractant and protein1 (MCP-1), fasting glucose and serum insulin levels of apparently healthy Egyptian premenopausal obese  women and compared with healthy non obese control women. This study aimed to determine assoction between inflammatory markers, BDNF, MCP-1, obesity measures and insulin resistance (IR) risk. The study included 120 apparently healthy Egyptian women, 60 obese and 60 non- obese. IR has been estimated by the Homeostasis Model Insulin Resistance (HOMA-IR). Results showed marked elevation in inflammatory markers and MCP-1 in obese women than controls. Significant reduction of BDNF levels was observed in obese cases with significant negative correlations with obesity measures and HOMA-IR and positive relations with TNF-α and MCP-1. In conclusion our data demonstrated increase of inflammatory markers and MCP-1 as well as decrease of BDNF levels in obese women, suggesting that these biomarkers might be positional candidate linking obesity markers that play important role for development of IR in apparently healthy Egyptian obese premenopausal women.

Abstract

Introduction: Community Health Center as health service unit that was the forefront of basic health services was expected to provide quality health services in accordance with community needs. Widodaren Community Health Center was one of Community Health Center in Ngawi Regency area which since 2015 has a full “BLUD” (Regional Public Service Agency) status, and all villages were still in pra advisory villages status. To saw how much quality of health services, it was necessary a Community Satisfaction Survey (CSS). The purpose of implementing CSS was to measure level of community satisfaction with basic health services by the Widodaren Community Health Center, Ngawi Regency in 2019. Community Health Center as health service unit that was the forefront of basic health services was expected to provide quality health services in accordance with community needs. Methods: This research was survey. Samples were part of community who use health services at Widodaren Community Health Center who were selected by quota sampling technique. Variables studied were the level satisfaction of health services provided community health center. Collecting data with questionnaire. Data analysis with calculating the Community Satisfaction Index. Presentation of data in the form of tables and spider web diagrams. Results: The results showed that there was an influence of the physical activity model (Gambo Rasa) on blood glucose levels of type-2 diabetes mellitus. Results of this research are the service quality in all service units was above the target of 85, and an average of 88.187. Six service units were in excellent category and three service units in good category. Service quality based on service dimensions was above the target of 3,395 and an average of 3,530. Discussion and Conclusion: Four dimensions in excellent category and five dimensions in good category. The service quality of Widodaren Community Health Center in all service units and all dimensions are in good and excellent categories. As an effort to improve quality services, it is necesarry survey of needs or expectations of the latest, and satisfaction survey in 2020 as an evaluation of quality improvement efforts that have been made based on the results of CSS at 2019. Abstract

Background and Objective: Using fused to metal porcelain as crowns is the most common methods of replacing missing teeth.IL-1-beta plays a key role in immunity and inflammation. Gingival crevicular fluid (GCF) reflecting the periodontal inflammation was sampled in the same area. This study aimed to compare the amount of IL-1β in the GCF of porcelain -fused to metal crowns with that of healthy teeth. Method: This case-control study was performed on 16 people with porcelain- fused to metal crowns with clinically acceptable standards. The history of non-systemic disease, not smoking and taking any regular medications up to 6 past months was taken. Healthy teeth corresponding to veneered teeth in the opposite side of the jaw were selected as control group. After the initial sampling the teeth received a phase of a non-surgical dental treatment, and the sampling was repeated at 1 and 3 months after the initial sampling. After collecting data with SPSS23 statistical software the data was entered into computer.To compare the concentrations of IL-1β in the two groups Paired t-test was used. Results: The concentration of IL-1β at the all three intervals of testing in teeth with porcelain fused to metal crowns was higher than of it in the control group (P< 0.001). Furthermore, the concentration of IL-1β was decreased in both groups during the whole time. (P<0.001 & P<0.003). Conclusion: The concentration of IL-1β in GCF of teeth with porcelain- fused to metal crowns with clinically acceptable standards is higher than of that in healthy teeth. Porcelain- fused to metal crowns are involved in the production of IL-1β.Keywords: IL-1β, porcelain- fused to metal crowns, GCF, PFM.

Abstract

One of the concepts which has received a great deal of attention during the recent years from the organizational-industrial psychologists is the fatigue, exhaustion, lassitude, torpidity, languor and indolence in the job operators which is known as the so-called job burnout. The objective of the present study is to identify and prioritize the effectors which influence the job burnout in the staffs working in Amir Al-momenin hospital. The present study is an applied research in terms of the methodology it adopts because the indices extracted from the study literature are identified and rated according to the study conceptual model and the organizations can take advantage of the results presented herein to device the best strategy and choose the most optimum indices and put them into practical use. In the present study, the study theoretical principles and the study background have been drawn out of the library searches and the study of university articles and journals and also it has to be mentioned that the field study methods and thorough analysis of the study population have been applied as a source of collecting statistical information for testing the study hypotheses. The study population in the present study is basically consisted of the entire managers working in Amir Al-momenin hospital the total number of whom reaches to 150 individuals and subsequently a number of 108 individuals were selected as the study sample volume by making use of Morgan’s table. To analyze the data, AHP method has been employed along with making use of Expert Choice software. The results of the study indicated that environmental, organizational, individual and vocational factors are found of a great effect on the job burnout in the staffs.Keywords: Job burnout, Environmental factors, Organizational factors, Individual factors, Vocational factors.
In Vitro Effect of MRSAc in Bacteriocins Produced from MRSA on Propionibacterium Acnes Comparing with Antibiotics Mais E. Ahmed

Abstract

Propionibacterium acnes most isolation higher in female than male (14% and 4%) respectively all strain resistance antibiotic and using purification Bacteriocins from MRSA isolation from wound to control acne vulgaris. In this study, Futurity one skin swabs were obtained from patients suffering from acne vulgaris from consultation unit in Baghdad teaching Hospital, from both sexes. Ten isolates (18%) of anaerobic gram positive Probionibacterium acnes were identified and diagnosis by culture, Api 20A system, Resistance to antibiotics and Vitek 2 ANC System. The local isolation Methicillin-resistant Staphylococcus aureus MRSA strain from wound specimen diagnosis by cultures, Biochemical test and Vitek 2 ANC System. It was found that P. acne isolates were higher in female than male (14% and 4%) respectively, and more prevalence in age (16-21) years patients and lower in older age. The effect of some antibiotics on Probionibacterium acnes was investigated, and the results showed that all isolates were resistance to antibiotics (Azithromycin, Erythromycin, Metronidazole and Clindamycin) and have been sensitive to (Levofloxacin). There is a need potently replacements to antibiotics to axing the impedance to treat acne infections. Purified bacteriocin (MRSAcin) produce from using against resistance P. acnes their wide inhibition zone by the well diffusion assay (WDA) method and Paper disc method reached (15 and 11) mm respectively at concentration 62.5 µg/ml compared with sensitive antibiotic disc of Levofloxacin that diameter reached 9mm. The current in vitro study proved that P. acne was resistant to most antibiotics except levofloxacin. Bacteriocins (MRSAcin) had a good antibacterial activity and its effects exceed the effects of levofloxacin antibiotic.Keywords: Bacteriocin, MRSA, P. acnes

Abstract

The atypical antidepressant Nefazodone  used in different concentrations (2.5, 5, 10 µg/ml ) to study its genotoxic effects on HepG2 cell line by using single cell gel electrophoresis assay (Comet) and spectral karyotyping test (SKY).The results showed that Nefazodone causes a significant increment in the  three important parameters (tail length, percent of DNA in tail and tail moment) of comet assay for the three used concentrations in a dose-dependent manner when compared with negative control (p˂0.01). SKY analysis was performed at the concentration of (10 µg/ml). Both the treated and negative control cells showed abnormal chromosomal number (44-54 XY) per cell, common chromosomal rearrangements and other abnormalities in the majority of cells analyzed. As a result Nefazodone has no chromosomal aberration at 10µg/ml and the comet assay results conceder as false positive.Keywords: Nefazodone, Genotoxicity, HepG2 cell line.

Abstract

Thalassemia syndrome is the commonest, well-known monogenetic inherited hematologic disorder that caused by a decrease or an absence of globin production. The study aimed to assess the differences in the socio-emographical characters, clinical finding and adherence to the treatment stratigeis between thalassemia major and other types of thalassemia. An analytical, cross sectional-retrospective- hospital based  study,  including the past 5 years; from 2012  till the end of 2017   in thalassemia center-Al-Habboby hospital-Thiqar health directorate-Iraq.    All the registered cases of thalassemia were included in this study   .The study include 103 patient with mean age of (10.47± 4,936), 52.4% were female, thalassemia major was representing the main proportion (68%) of the studied population, while only(32%) were with other types of thalassemia syndromes such as  sickle thalassemia and thalassemia intermedia, Early age of thalassemia major presentation was the main finding in this study, Al-Nasiriya was the main site of inhabitation. Age of presentation, address, occupation, and educational level, Osteal lesion, growth retardation and carrying of some known complication were of significant association with the different types of thalassemia. Second degree relativeness was the main form of consanguinity among the parents thalassemia major, There was no significant statistical association in adherence to the treatment strategies among the transfusion and non-transfusion dependent thalassemia in their compliance to their applicable regimen. Socio-demography and clinical character are differ among different types of thalassemia, Early age of thalassemia major presentation was the main finding in this study,   osteal lesion, growth retardation, cholelithiasis and infection with viral hepatitis had significant difference, while the consanguinity, blood group and  adherence to the treatment had no significant association.Keywords: Thalassemia , Socio-demography , Thi-Qar.
