Abstract

The present study describes a procedure for indirect regeneration of Moringa oleifera using leaves and stems as explants derived from in vitro germinated seedlings grown on Murashige and Skoog (MS) medium supplemented with different concentrations of auxin and cytokinins. The best callus induction was observed on MS medium containing 1.5 mg.L-1Indole-3-butyric acid (IBA) with 1.5 mg.L-16-benzylaminopurin (BAP) on stem explants .However, when callus was transferred onto a maintenance  medium supplemented with different concentrations of IBA (0.0, 1.0, 1.5, 2.0 or 2.5) mg.L-1and BAP (0.0, 1.0, 1.5, 2.0 or 2.5) mg.L-1, for increasing  callus mass, the combination of  1.5 mg.L-1  IBA with 1.0 mg.L-1 BAP resulted in best response in increasing callus fresh weight. A high shoot proliferation percentage was achieved at the combination 1.0 mg.L-1Kinetin (Kin) with 0.4 mg.L-1Naphthalene acetic acid (NAA) recorded (90%).The rooting percentage was 100% for shoots regenerated from callus cultures when MS medium was supplemented with 2.0 mg.L-1 IBA .After acclimatization, 80% of plants were able to survive.Keywords: Moringa oleifera, Indirect organogenesis, Explants, In vitro, Callus.

Abstract

Zingiber purpureum Roxb. was a plant that has the potential to be developed as a chemotherapy agent. Bioassay guided fractination aims to examine the anticancer activity of n-hexane, ethyl acetate, and methanol fractions from ethanolic extract of bangle plant rhizome to colon cancer cell type WiDr and Vero cells. The apoptotic test and modulating cell cycle were observed by flow cytometry method. Isolation process of the most active fraction. The pure isolates obtained were identified by structures with structural elucidation using ultraviolet, 1D-NMR, 2D-NMR and liquid chromatography-mass spectrometer. Obtained 2,3-dihydro-5-hydroxy-7-methoxy-2-phenylchromen-4-one compound of ethyl acetate fraction number 21. With a value of CC50 to a WiDr cell of 11.24 ± 3.44 μg/mL and 342.22 ± 4.98 μg/mL to Vero cells.

Abstract

The bioactive properties and therapeutic importance for kefir grains encourage us to study the optimum culture conditions for maximum propagation and production. The kefir grains were cultured at different change conditions included (type of culture media, temperature, incubation time, and agitation speed). Additionally, the antagonistic activity for kefir was evaluated  against four pathogenic bacteria (Staphylococcus aureus , Staphylococcus saprophyticus, Esherichia coli O157 and  Klebsila penumonia) in two different methods ;First, the antibacterial activity was estimated by using kefir grains and  measuring the inhibition zone of growth .Second , the antibacterial activity was estimated by using the kefir whey  (fermented milk supernatant )after clarification process . The results showed The optimum culture condition  were achieved  by using the skim milk as culture media , incubation at 37 ◦C , for 24 h at static condition , these conditions lead to increase the growth rate to about  55.6% (70.1 gm/L), while the using of culture media supplemented with 5% glucose ,1% lactose and 5% yeast extract , the growth rates were (45.8 %, 41.3 % and 2. 2 %) respectively ,with biomass about (65.6 , 63.4 and 46 gm/L) respectively , in general ,these supplement lead to reduce the  rate of kefir grain biomass. The result of antibacterial activity of kefir grain revealed the highest inhibition zone was 4.05 cm for S.aureus, 3.55 cm for both S. saprophyticus and K. penumonia while the least inhibition zone was 2.9 cm for E. coli O157 strain, whereas the antagonistic activity of kefir whey     showed the inhibition rates were 86.6 %  for S. aureus, 90.7% for both S. saprophyticus and K. penumonia and 89.6 % for E. coli O157 strain . We concluded from this study the optimization of growth environment is very important for achieving the maximum production of kefir grains biomass and the kefir grain evaluated had high antagonistic activity toward the pathogens .This indicate potential using these grains as functional food.Keywords: Kefir grain; Culture conditions; Biomass; Antagonism; whey; Pathogenic bacteria.

Abstract

Objective: Cataract is the leading cause of blindness in the world with increasing trend both in prevalence and incidence. Surgery is the only definitive treatment for cataract but often unaffordable for society in developing countries. The incidence of cataract could be prevented by understanding its risk factors and formulate health programs according to them. The purpose of this study was to assess the prevalence and risk factors of senile cataract among 50 years old or older Balinese population. Method: The population-based cross-sectional study was conducted from September 2015 until July 2016 in Bali province. The variable’s data were obtained through direct examination and interview. Poisson regression analysis was used to assess the definitive correlation between risk factors and cataract. A p-value <0.05 or value which falls within 95% confidence interval were considered significant. Result: 550 subjects were enrolled during the study. The number of cataracts found in this study was 231 (42%). According to the analyses, cataract was significantly associated with older age (PR: 3.05; p=0.001), low educational level (PR: 2.18; p=0.04), diabetes (PR: 2.96; p=0.001), and the rate of ultraviolet exposure (PR: 1.92; p=0,002). Conclusion: The prevalence of cataract among 50 years old or older in Balinese population was 42%. Older age, low education, diabetes mellitus, and ultraviolet exposure were found to be significant and independent risk factors for cataract in 50 years old or older Balinese population.Keywords: Cataract, Prevalence, Risk factor.

Abstract

Waste polymers are recently getting higher research interest and one of the solutions is synthesizing synthetic biodegradable polymers where the focus is to modify polymer chemical structure without altering its intended application. Biodegradation of polystyrene is one of the major research concern and is extremely difficult to accommodate the best physical, chemical and optical properties of the desired polystyrene end products. Doping polystyrene with copolymer such as N-benzyl-4-vinyl pyridinium chloride, PSt-co-BVP(Cl), has been found able to be bio-degradable when treated with activated sludge in the soil. The biodegradation of polystyrene copolymer that contained 46.1-mol % BVP(C1) is found to follow first-order kinetic and with a half-life of 5.5 days when about 0.5 or 1.0 g/kg of polystyrene is utilized as basic materials. Polymeric base materials have gained a wide theoretical interest and practical application in sensor technology which motivated this research in finding ways to modify the PSt-co-BVP(Cl) and investigate its chemical structure modification by low dose laser irradiation. The laser photon modifies PSt-co-BVP(Cl) of its physical, chemical and surface morphology where the strong oncoming of BVP(Cl) with microbial cells and viruses can be tuned as bio-materials sensor.

Abstract

This paper aims to estimate the effects ofthree operating parameters such asinitial pH, temperature and oxidant agents (like H2O2 and Fe2+) on the stability of the food azocarmoisine dye under UV-A/ carmoisine/ ZnO system. The performed experiments in this work proved that the optimum initial pH is at zero point charge of ZnO and equal to 9.14 and with efficiency % equal to 90.936 at 8 min. The study of temperature was enhanced this photo reaction with activation energy equal to7.562 kJ mol-1.The addition of oxidant agents to an aqueous solution of carmoisine dye was declined the activity of decolorization, that referred to the addition of these compound will increase the resistance of carmoisine dye to light. The quantum yield ( ) for decolorization of carmoisine dye with the using ZnO without and with the using oxidant agent (H2O2 and Fe2+) was following to the sequences:  without the oxidant agent >  with the using oxidant agent (H2O2) > with the using oxidant agent (Fe2+) and equal to 0.485, 0.279 and 0.141 respectively.The physical deactivated process mostly was dominated on this photoreaction.From the other word, the addition of oxidant agents is useful to decrease the degradation of this dye when used it in food's industries.Keywords: Carmoisine dye, Food red 3, acid red 14 dye, Azorubine, Oxidant reagents, food's dye.

Abstract

Background: Diabetes a devastating disease throughout the world. It is related with numerous mechanisms, the first one is oxidative stress. Oxidative stress shows an main role in the pathogenesis and the complications of disease. Objective: The aim of the current study was to measure five important factors in patients with type 2 diabetic patients (the amount of 8-OHDG, IL-6, IL-7, Blood Pressure, and Pulses). These factors were surveyed in type 2 diabetic patients to compare with healthy controls and study the association among 8-OHDG in diabetic patients with other parameters in the study. Materials & Methods: Population consists of 51 patients diagnosed with T2DM and a control group of 31 healthy people. 8-OHDG, interleukin-6, interleukin-7 and blood pressure levels were assessed in all subjects. Results: Levels of five markers in serum (8-OHDG, IL-6, IL-7, Blood Pressure, and Pulses) were significantly greater inT2DM group compared to control group. The receiver operator curve (ROC) analysis of the forthcoming variations revealed the descending order of serum IL-7 (0.975), IL-6 (0.972), and 8-OHDG (0.899), Conclusion: the study recommends that 8-OHDG level in type 2 diabetic patients may be a useful marker of DNA damage. So we should monitor this variable level in blood continuously to maintain their health.Keywords: ● T2DM ● Interleukin-6 ● Interleukin-7 ●8-OHDG.

Abstract

AbstractObjective The innate immune system has evolved as the first line of defense against microorganisms, which involves specific pathogen recognition receptors such as toll-like receptors (TLR). The aim of the present study was to investigate whether the TLR2 gene polymorphism could influence the susceptibility to pulmonary tuberculosis (TB) and its expression level as well as its concentration in patients’ sera.Methods A total of 120 subjects, including 80 patients with TB including 40 multiple drug resistance (MDR), 20 recently diagnosed (RD) and 20 old cases (OC) of TB patients, in addition to 40 healthy control, were examined for TLR2 polymorphism over locus -100 microsatellite guanine-thymine repeats, 24969 (T>C) and 5305 (C>T). TLR2 gene expression was assayed using a real time PCR technique; furthermore, sera from 68 TB patients and 20 healthy control were assayed for TLR2 level using ELISA technique.Results We found that the TC genotypes showed a significantly increased frequency in MDR patients than control (35% vs.12.5%, respectively; **P = 0.034), and the associated OR value 3.77 with EF value of 0.25. That may regard TLR-2 rs3804100 TC as TB susceptibility genotype in the investigated Iraqi patients. Whereas the TLR-2 serum levels varied significantly (*p<0.05) between studied groups in regard to TT genotypes. For TLR-2 rs7696323, all TB patients and control groups have CC genotypes (Wild type). In respect to (GT)n microsatellite repeat; GT repeat alleles had insignificant differences (p > 0.05) between MDR and OC than control groups, while RD group was significantly higher [**P = 0.006] than control group in GT(24) repeat allele, with OR value of 5.2 and EF value of 0.33. Furthermore, this GT repeat allele associated with susceptibility to TB. For TLR-2 gene expression, the results revealed a significant decrease (*p< 0.05) in TB patients in comparison to control groups. A novel TLR-2 rs3804100 SNPs were detected in Iraqi Arabs at g.24870 in 15%, g.24864 in 1.25% and g.25212 in 32.5% of patients. Nevertheless, in control the novel SNPs were g.24887 in 2.5%, g.25212 in 7.5% and g. 25216 in 7.5%. On the other hand, a novel TLR-2 rs7696323 SNPs were determined in Iraq Arabs patients and control at g.23934 in 10%, g.23564 in 1.25%, g.23460 in 6.25%, g.23445 in 11.25%, g. 23452 in 10% and g. 23477 in 10% of TB patients. While in control group, g.23957 in 7.5%, g.23438 in 7.5%, g.23445 in 7.5%, g.23452 in 32.5% and g.23443 in 2.5% of control.ConclusionTLR2 genetic polymorphisms influence susceptibility to pulmonary TB. TLR2 variants play a role in the development of TB disease, probably by affecting TLR2 expression level. Moreover, the present study showed that during anti-tuberculosis treatment, which were seems likely responsible for controlling infection and excess inflammation

Abstract

The reaction of diethylcarbamic chloride (one mole equivalent), ammonium thiocyanate (two mole equivalents) and thiosemicarbazide (one mole equivalent) in an acetone medium resulted in the formation of the unexpected4,4-dimethyl-2,6-dithioxo-1,3,5-triazinan-1-yl)-3-(diethylaminocarbonyl)- thiourea3in 83% yield. The chemical structure of the title compound was confirmed by IR, NMR and elemental analysis. Further, a suggested mechanism has been proposed to illustrate the route for the formation of the title compound. In addition, theoretical evaluations of the stable geometries of compound3were carried out using semi-empirical molecular quantum calculations with the PM3 method.Keywords: Thiosemicarbazone, Cyclization reaction, Spectroscopic analysis, Computational calculations.

Abstract

A  cross sectional biochemical and molecular genetics study was achieved from December 2016 to September 2017, enrolled seventy patients (36 males, and 34 females) with somatotrophic pituitary adenoma continuously attended to the National diabetic center in Baghdad. Their ages were between 20 to 70 years. This research aimed to observe the frequency in 665bp fragment of exonic 10 of aryl hydrocarbon receptor (AHR) genes mutations. Polymerase chain reaction (PCR) was used for the detection ofAHRmutations as a cause of acromegaly, by using specific primers for amplification of exon 10 ofAHRgene on chromosome no.7. Sequencing was applied forPCRproducts ofAHRgene, ten different alterations inAHRgene were diagnosed .Four mutations inAHRwere recognized in 20/70 (29%) patients which their codons A474T with substitution adenine by thymine (A/T), 176 and 180 with substitution thymine by adenine (T/A), and codon 411 with substitution of (C/T) for 665bp.We concluded that ten novelties SNP were detected in Iraqi acromegalic cases. Genetic variants potentially inducing functional abnormalities of the aryl hydrocarbon receptor (AHR) pathway are associated with a more severe acromegaly, increased pituitary tumor size, and somatostatin analog resistance in patients living in HR areas.
