Abstract

Cloning gene involves the construction of a recombinant plasmid that inserted in a competent cell. On the other hand, genetic engineering requires bioinformatic analysis to be converted into tabulation and data interpretation. The study, titled "cloning block 2 MSP1 gene of Plasmodium falciparum isolate Jayapura city and bioinformatics analysis" is aimed to improve the technique of cloning the MSP1 gene of P. falciparum, initiated the creation of DH5α competent cells, ligations and transformations, plasmid isolation, confirmation the recombinant plasmid and able to perform bioinformatics analysis and construct phylogenetic tree. This study began with the manufacture of E. coli DH5α competent cells, MSP1 gene ligation in pJET1.2/blunt vector and transformation by using the heat shock transformation method, plasmid isolation of alkali lysis method, then plasmid confirmed by PCR and sequencing method, further sequence analysis and phylogenetic tree construction. The results showed that confirmation of MSP1 gene presence in pJET1.2/blunt with PCR was successful. From a total of 4 positive colonies grown in liquid culture, then isolated plasmid and confirmed with PCR obtained electroferogram bands with a size about 1049 bp indicates the presence of MSP1 gene in plasmid. Based on the results, cloning of MSP1 gene using pJET1.2/blunt cloning vector and competent cell E. coli DH5α has been successfully performed. Bioinformatics analysis of sequencing result and phylogenetic tree were constructed successfully with 2 clusters isolate of malaria patients from Jayapura city. Key words: Bioinformatics, cloning gene, heat shock transformation, MSP1, P. falciparum.

Abstract

This article is essential in a further analysis that related with potential feasibility function of Xanthostemon novoguineensis in phytotechnology from one of an endemic plant in Papua, particularly in the remediation of polluted environment and the phytostructure of open green space area. This review aims to analyse the potential of this plant based on literature review and compare each research results  towards some species from Myrtaceae family. X. novoguineensis may be cultivated on an open green space area in Papua due to complying suitable requirements. Those requirements are namely ecology function as a source of insect diets, social-cultural functions in represent culture, the the relationship among Papuan Ethnic and their nature, and aesthetic functions due to its beautiful flower. In addition, the plant has a potential in phytoremediation of polluted environment due to its adaptation ability in a dry soil condition. As the comparison, the phytostructure of open green space area context, X. chrysanthus has been cultivated on a public and private open green space area in Malaysia and Singapore. It has an important role as ecology function due to provide the source of food to many birds species undirectly. Owing to make the city landscape greater, the flower of X. chrysanthus, which has conspicuously and magnificently yellow colour, is a suitable plant to be cultivated. Furthermore, in the remediation assessment of polluted environment, X. chrysanthus has the abilities in adaptation on the dry environment. The range levels acid (pH) of soil are 2-2.5. Interestingly, X. melanoxylon, which is an endemic species from Solomon Island, is growth naturally in an ultrafamic soils, consist of a high concentration of chromium, nickel, and magnesium. It also has a low degree of plants biodiversity. Therefore, according to X. chrysanthus and X. melanoxylon, X. novoguneensis has a tremendous potential as a candidate in Phytostructure and Phytoremediation. However, the further investigation is required to reveal the potential and the conservation challenge of this plant in Papua. Key words: phytoremediation, phytostructure, open green space area, X. chrysanthus, Potential.

Abstract

Kayu secang (Caesalpinia sappan) and rimpang temu mangga (Curcuma mangga) are medicinal plants that are efficacious for diarrhea due to bacterial infection. Kayu secang and Rimpang temu mangga contain secondary metabolite compounds capable of providing antibacterial activity, ie flavonoids. This study aimed to obtain the granule as supplies, which contain a combination of kayu secang and rimpang temu mangga with good quality, it is favored by consumers and has high antibacterial activity. Both powdered simplisia powder was kinetic with ethanol 70 % and dried by freeze drying method. Both powder extract tested the quality of raw material and test of antibacterial activity against bacteria Escherichia coli, Salmonella sp and Shigella sp. Both powder extracts were made granular preparations with the addition of excipients (Handbook of Pharmaceutical Excipients). The granule preparations were tested for granule quality standards and preferred levels and antibacterial activity. Data were analyzed statistically with one way ANOVA method (p= 0.05). Rimpang temu mangga extract powder added maltodextrin did not provide antibacterial activity. The granule extract of Kayu secang extract of formula 1 (F1) meets the requirements of granule quality standard, preferably consumer and has high antibacterial activity, i.e 18.7 mm against E. coli bacteria, 28.3 against Salmonella sp bacteria and 30.5 against Shigella sp bacteria.  Key words: Secang, C. sappan, temu mangga, extract, granule, antibacterial

Abstract

Daun afrika (Vernonia amygdalina) contains flavonoids, tannins, saponins, and terpenoids that can be used for pharmacology activity: annihilate the parasites that causing schistosomiasis, malaria leishmaniasis, antiamoeba, antitumor and antimicrobials. The purposes of this research were to screen the phytochemistry of african leaf, formulate and test the stability of vanishing cream (VC) base. The methods used were sample collection, extract preparation, formulation, and stability evaluation. The results showed that extract afrika leaves had alkaloids, flavonoids, tannins, saponins, steroids, and triterpenoids. Extract of daun afrika can be made to good VC base formula including organoleptic test, homogeneity test, pH test, spreading test, sticky test, and VC safety test by distributing questionnaires. The cream was stabil for 21 days at room temperature. Observation of physical properties good performed every week to organoleptis, pH, adhesion, and spreading power.  Key words: stability, V. amygdalina, kill parasites, Vanishing Cream

Abstract

Beauveria bassiana is one type of entomopathogenic fungi that can be used as a biopesticide. Papua province is rich in natural contents, as well as B. bassiana fungus isolated from soil rhizofir banana plants at the location Housing Cenderawasih University Lecturer Jayapura, Perumnas III Waena, Heram District, Jayapura, Papua Province. Local isolates of B. bassiana fungus reproduced in the Laboratory of Microbiology, Cenderawasih University Jayapura. This study aimed to get to get a concentration of B. bassiana fungus that effectively kill pests Spodoptera litura mustard greens (Brassica juncea), the time interval applications (morning, afternoon, and evening) B. bassiana fungus that effectively kill pests S. litura the mustard greens (B. juncea) and knowing the difference effectiveness of B. bassiana fungus concentration and application time (morning, afternoon, and evening) to pests S. litura green mustard (B. juncea) as well as the interaction fungus B. bassiana and application time (morning, afternoon and evening) to pests S. litura green mustard (B. juncea) with Randomized Complete Design Factorial (RAK-F) which consists of two factors: the concentration of fungi B. bassiana and time of application. The results obtained that the concentration of the fungus B. bassiana were effectively kill pests S. litura is a concentration of 1 % and 1.5 %. The time interval B. bassiana fungus that effectively kill pests S. litura is morning, afternoon and evening. Differences in the effectiveness of the fungus B. bassiana and applications in the time interval kill pests S. litura is a concentration of 0.5 % and 1.5 % in the morning, afternoon, and evening. Interval time of application of the interactions of fungi   B. bassiana in killing pests S. litura is an afternoon with a concentration of 1.5 % on day 5 by 5 %, the 8th day amounted to 17.5 %, the 11th day amounted to 32.5 % and on day-14 amounted to 42.5 %.  Key words: B. bassiana, B. juncea, effectiveness, time applications.

Abstract

The aim of this research was to measure the differences amount of erythrocytes in the blood of the rat (Rattus norvegicus) strain wistar before and after been given of the extract of the red fruid (Pandanus conoideus).  This was a pre experiment with The One Group Pre Test – Post Test only design, the extract of red fruit (P. conoideus) was the  independent variable and the dependent variable was the amount erythrocyte in rat blood. After the acclimatisation program in 7 days, the blood of this 20 rats were taken to measure the erythrocytes. For the next 7 days, all of the rats were given the extract of red fruit per oral by the sonde 5m/bb.  The blood were taken again to measure the erythrocytes again, in the day after the last given of the extract of red fruit. The data were tabulated for normality test with Shapiro-wilk (SPSS v.24), homogeneity test, Levene test, and followed by Paired T-Test. The results were show that the mean of the erythrocyte before given the extract of red fruit was 5.54 x 106/mm3 and after that was 7.65 x 106/mm3.  Key words: R. novergicus, P. conoideus, erythrocyte.
