Abstract

The objective of the investigation was to study the relationship between ratio prooxidant and total antioxidant (peroxidation index) and cell damage in male rats. Fourty male Sprague dawley rats, 3 month of age, 150-200 g of body weight were alloted randomly to 2 groups of 20 each. The first group was fed normal diet (control), the second group was fed high fat diet (20% fat). After 2 weeks, the blood samples were taken out from 5 rats of each group for measur­ing prooxidants and antioxidants. The prooxidants and anti-oxidants st-atus in this study was done by measuring thio barbituric add subs tance I TEARS) and total antloxldant status (TAS) concentration. The heart, liver and kidney tissues were collected for histopatological analysis, Perox­idation index (PI) were determined by dividing TBARS value with TAS value. The same procedures were repeated after 4, 8, and 16 weeks. Analysis of the data showed that after 16 weeks on experimental diet, the mean concentration of TBARS in blood of group I was 1.45Q+G.158 umol/L and the mean concentration of TBASS in blood of group 11 was 4,75 + 2,17 nmol/L. The highest ratio between prooxidant and total antioxidant was 17.47 and the Lowest ratio between oxidant and total antioxidant was 0.849. Analysis of histopatology showed that there were hyalinitation and oardiomyopathy in the heart tissu, fatty liver degeneration and necrosis in the liver and necrose and atrophy of the kidney. From the results can be concluded that (1) feeding high fat diet induced high ratio between proojiidant and antioxidant con^ centration (peroxidation Index). (2) Cell damage can be caused by high peroxidation index. (3) Individual variation might be caused by genetic factor.

Abstract

Estrogen is an inhibitor of bone resorption. Although there is some disagreement, most studies indicated that bone loss is accelarated at the time of menopause wether it is induced surgically or naturally. When the condition men­tioned above is supported by imbalance diet, especially calcium to phosphor ratio, the bone loss will be acce­larated. The obyectlves of the research were to study the effect of panhisterectomized and high phosphorous intake on calcium and phosphate anorganic concentration in the blood, and bone changes in rats.Sixteen female of rats ± 30 days of age were randomly divided into four groups (A, B. C, and DJ of four each. The group A and C were unpanhisterectomized. Whereas the group B and D were panhisterectomized. The group A and B were fed diet contain Ca:P ratio = 1.5:1, whereas the ratio of calci­um and phosphorus 1.5:6 was given to group C and D. At the end of the experiment, the blood samples were taken directly from eye (plexus retroorbitalis) for calcium and phosphate analysis. The left femur were taken for histopathological analysis.From the results can be concluded that the diet con­taining the Ca:P in variation ratio, panhistcrectomized. and interaction between two of them had significant effect on the blood inorganic phosphate (P < 0,01). The diet contain­ing CaiP = 1.5:6 for unpahisterectomized caused earliest lesion of osteodistrphia fibrosa, whereas for panhisterec-tomized caused mild osteodistrophia fibrosa.

Abstract

All vertebrata genomes studied so far contain SINEs, that consti tute up to 5% of the total genome. Most of the SINE sequences studied so far are mammalian genomes. There Is very limited information about SINE sequences from rep­tiles, especially of Varanus family.Isolation of SINE sequences were carried out by con­structing the komodo genomic library in E. coli. Suspected positive clones were detected by genome to genome hybridiza­tion. The SINE sequences identification were carried out by enzymatic method, and the DNA sequences were analysed with the GENETYX-SV program-Up to now there are 2 SINE sequences from komodo genome have been isolated and identified. Their sequences homo logy is 85-2% From Database search analysis with the GenBank indicated that those SlNEs were not evolved from any spe­cific tRNA gene.

Abstract

A research has been conducted to evaluate the effect of certain desinfectants on the speculation of the Eimerla tenclla oocysts.In this research in vitro methode has been used. This research used oocysts Eimeria tenella which have not yet Sporulated. 6 kinds of disinfectants and water as control. These desinfectants were used in the research namely : Lysol, Creolin. Kalium Jodida (KJ). Formalin, Bromoquod and Ca (OH)2.The oocysts which have not yet sporulation were imerted in these desinfectants. The duration of liners ion at each treatment was succescively 10 minutes. 20 minutes and 30 minutes. The desinfectants concentration used is of three degree of strength, in the following proportion = 1:2:4.The data colected was the percentage of oocystys which have sporulated 24 hours, 48 hours and 72 hours, after treatment. The data was analyzed with the Split Plot method (Gill. 1978).The result of research showed that each desinfectant had a different effect on the percentage of the oocyst sporulation (P < 0,01) . Bromoquod was found to be the best desinfectant, and the others were in this following order = Creolin. KJ, Water, Lysol. CafQHJn and formalin. The Concen­tration was also found significantly effective (P < 0,6) in suppressing the sporulation of the oocysts. Creolin concen­tration of the 4tri strength was found to be the best desin­fectant and the others were in this following order : water. KJ. Bromoquod, Lysol, Ca(OH)£ and formalin. The duration of imersion had no significant influence

Abstract

Coccidiosis is one of the problems in poultry husbandry since it causes a lot of loss. Nowadays, several ionophore coccidiostats have been used to prevent coccidiosis. However, because of some complaints for ionphore coccidios­tats usage, especially Monensin which effect to the weight of digestive tract, therefore the use of coccidiostats should be taken with care. This investigation is intended to undestand the Increase of weight and some changes in diges­tive tract because of the use of 4 ionophore coccidiosis.This investigation used 52 Hubbard broiler of 1 days age. After 2 weeks of adaption 4 broiler were taken randomly as a control group (without coccidiostat). and the remaining 46 broilers were divided randomly into A group, that is. Monensin (Mo). Karacin (Na). Salinomicin (Sal), and Madura­micin (Mad)- The Mo group was divided futher into 3 sub­groups, those are Mo 70, Mo 110, and mo 130 that were fed containing Monensin 70, 110. and 130 mg/kg feed. The other groups were also divided similarly into 3 subgroups, those are, Na 50, Na 70. Na 90; Sal 40, Sal 60, Sal 80; and Mad 4, Mad 5, Mad 6. Number after the abbreviation of coccidiostats indicated the amount of mg/kg reed. The coccidiostats were used to the broilers since 14 until 45 days old. and finally all of the broilers were Killed 4 days later.Using variance analysis with split plot pattern showed that all of the coccidiostats did not effect to the weight growth of broilers (the weighting of broilers was carried out every week since 14 until 49 days old). The use of orthogonal polynomial contrast test indicated that the Monensin coccidiostat effected to the index (the related organ weight/the total weight x 100%) of praventriculus to Cloaca by an equation of VA = 26,9736 - 0,4602 X + G,0024X^P therefore the suggested dosage is 96,53 mg/kg feed. For the Maduramicin coccidiostat. besides it effected to the index of proventriculus-to cloaca by an equation of VR = -12.790 + 7,461 X - 0.696 X*. it also effected to the inffex of duode­num to cloaca by an equation of yc = -5,910+3,585 X - 0.315 X * therefore the suggested dosage is 4 mg/kg feed. The use of Naracin and Salinomicin did not influence to the Index of tractdigestive. All of the coccidiostats did not effect on the thickness of duodenum, jejunum, and ileum. covering diameter (by macroscopic measurement), also the indexes of epithelium/villi weight, mucosa layer thickness, and mus-culer layer thickness that measured by microscopic method-From this investigation can conlcuded that the use of Monensin in 96,53 mg/kg feed. Naracin In 50-90 mg/kg feed, Salinomicin in 40-80 mg/kg feed, and Maduramicin in 4 mg/kg feed will be safe, which means that they willnot increase the weight of tractusgastrointestine.

Abstract

The purpose of the investigation is to find out the serologically prevalence oT toxoplasrnosis in animal husban­dry primarily in sheep, goats, cattle and pigs-Serum samples were taken from 100 sheep. 50 goats, 50 cattle and 100 pigs in a Yogyakarta abattoir. The sera were examined for Toxoplasma gonddi antibody by using direct agglutination test with Toxo-Scrven DA. It was considered that titers of 1:40 or greater were positive.The obtained results showed that prevalence of anti­bodies to T. gondii were : Sheep 50%, goats 15&. cattle 2% and pigs 44X. The sex was no err 1.1 on the prevalence of toxoplasmosis.
