Abstract

A member of the cytochrome P450 superfamily enzyme that play a role in microsomal fatty acids oxidation process encoded by cytochrome P450 2A6 (CYP2A6). This study aimed to examined the CYP2A6 gene polymorphisms and their association on fatty acid traits in Indonesian native sheep. A total of 299 rams aged 10-12 months with body weights between 20-30 kg used to identify the CYP2A6 gene polymorphism are consisted of 36 samples of barbados cross sheep (BCS), 35 samples of compass agrinak sheep (CAS), 20 samples of javanese fat tailed sheep (JFT), 36 javanese thin tailed sheep (JTT), 20 samples of garut sheep (GS), 45 samples of garut composite sheep (GCS), meanwhile for the fatty acid analysis was carried out by using 107 of loin samples from the total sheep. Identification of the CYP2A6 gene polymorphisms were performed using PCR-RFLP (Polymorphism Chain Reaction-Restriction Fragment Length Polymorphism) with the BSmAI restriction enzymes. The amplification product was 286 bp. Polymorphism were found in JFT, JTT, GCS and JS with GG and GT genotypes, while BCS, CAS, and GS were monomorphic with TT genotype. The CYP2A6 BSmAI polymorphism was in Hardy-Weinberg Equilibrium for JFT, JTT and JS, while BCS, CAS, GS, GCS, and combined was deviated based on chi square. A SNP g.49170107 G>T of the CYP2A6 gene polymorphism was significantly associated (P <0.05) with only erucic acid (C22:1n9). The GT genotypes had a higher value than GG genotypes The CYP2A6 gene could be used as a selection marker to improve fatty acid traits in Indonesian native sheep.

Abstract

The aim of this research was to identify the effect of feeding fermented Sargassum sp. as complete feed ingredient up to 20% of total feed on nutrient consumption and digestibility, and growth performance of thin-tailed sheep. Twelve female thin-tailed sheep aged 6 to 8 months with an average initial body weight of 14.13 ± 2.08 kg were divided into three groups. The treatments were control without fermented Sargassum sp. (S0), 10% fermented Sargassum sp. (S1), and 20% fermented Sargassum sp. (S2). Complete feed was formulated by isoprotein and isoenergy formulation, containing 16% of crude protein and 61% of total digestible nutrients. Complete feed was given at 3.5% of body weight (% dry matter) and drinking water was provided ad libitum. This research was done in 6 weeks with 14 days adaptation period and seven days of collection period. Growth performance was measured for 4 weeks after the adaptation period. The variables observed were dry matter (DM), organic matter (OM), crude protein (CP), ether extract (EE), crude fiber (CF), nitrogen-free extract (NFE), and total digestible nutrients (TDN) consumption. The data obtained were subjected to one-way ANOVA (analysis of variance) and the difference existed (P value <0.05) between means were analyzed with Duncan’s multiple range test. The results showed that there was no significant effect of feeding fermented Sargassum sp. on nutrient intake, and digestibility of nutrients (P>0.05), however, S0 treatment appeared to have CP, CF, and EE intake, as well as digestibility of DM, CP, CF, and NFE higher than S1 and S2 treatment. The results showed that there was no significant effect of feeding fermented Sargassum sp. on nutrient intake, digested and digestibility of nutrient, absolute and relative weight gain as well as feed conversion.

Abstract

The objective of this research was to characterized morphology and estimated genetic distance between intra population of Kuantan cattle. A Total of 213 cattle (44 male and 169 female with age ranging from 2-3 years) were used in this study and collected from extensive ranging systems in Three sub-population (Cerenti, Inuman, and Kuantan Hilir regions) Kuantan Singingi Regency, Riau Province. Five variables were measured that is Body Length (BL)(cm), Wither Height (WH)(cm), Hip Height (HH)(cm), Chest Girth (CG)(cm), and Chest Depth (CD)(cm). Data obtained were descriptive analysis, Principal Components Analysis (PCA) and Hierarchichal Clustering Analysis (HCA)  using XLSTAT program. All variables of body measurement in the Kuantan Hilir region were higher than Cerenti dan Inuman, Kuantan Singingi Regency. The first factor in PCA described body measurement contributed 32.77%, and the second factor described body shape contribute 25.83% of total variability. The dendrogram showed there is Three clusters of Kuantan Cattle based on this research.

Abstract

Increasing knowledge of farming is essential in increasing goat production in the rural areas of Banjarnegara Regency. Increased knowledge come from many sources, such as extension and non-extension services (other farmers, books, and other government officials). Systematic efforts continue to increase agricultural extension workers' role in providing information and knowledge to goat farmers. This study aims to identify farmers' preference for the agricultural extension as a source of knowledge and analyze the factors that influence the selection of sources of information and knowledge. One hundred sixty goat farmers in Banjarnegara Regency were selected as respondents using a multistage sampling method. The data collected were analyzed using descriptive statistics and logistic regression. The analysis results showed that most of the goat farmers in Banjarnegara Regency (74.37 percent) preferred obtaining knowledge from non-extension sources. In other words, only a small proportion of goat farmers (25.63 percent) prefer to get information and knowledge from agricultural extension workers. Based on the logistic regression analysis, it was found that goat farmers' education and age of farmers were essential factors (P<0.05) in making decisions related to source in obtaining knowledge on raising goats from the agricultural extension or non-agricultural extension workers. Goat farmers in Banjarnegara Regency who have lower education prefer to get information related to goat farming from non-agricultural extension workers. Meanwhile, younger goat farmers tend to get information from non-agricultural extension workers. Therefore, agricultural extension workers have a more strategic role for goat farmers who have higher education and are more mature with adequate goat farming knowledge.

Abstract

This study aims to determine the increase of the growth of Deutch Democratic Yokohama (DDY) strain mice through a selection program from generation 0 (G0, n = 323 ),  generation 1 (G1, n = 367) to generation 2 (G2, n = 386). The variables used for the selected program were weaning weight (WW) and adult weight (AW). For WW, the selection was made on the best 50% WW, and for the AW, the selection was carried out by choosing the best eight males and the best 32 females, using the value of genetic parameters and breeding values.  Based on the study, WW increased from G0 to G1 and G1 to G2 of 12,63% and 12,05%, respectively. The increase in AW from G0 to G1 and G1 to G2 was 7,85 % and 17,78%, respectively. The conclusion was that it was possible to increase weaning weight and adult weight by selecting the program.

Abstract

The present study aimed to determine the effect of cherry leaf extract (Muntingia calabura L) on hatchability of Balitnak Superior Free Range Chickens (KUB) through artificial hatching. The doses of cherry leaf extract used in this study were 0%, 10%, 15% and 20%. The hatching eggs used came from KUB hens that were intensively reared in the poultry house at UPBS BPTP, North Sumatra. The procedures applied in this study were collection of hatching eggs, fumigation of machines and equipment, setting of machines and hatching eggs, egg washing with cherry leaf extract and hatching of eggs for 21 days. During the hatching process, observations were made on predetermined variables. The observed variables were egg shape index, egg weight loss, shell temperature and hatchability. The method used was completely randomized design (CRD) with 4 treatments and 100 replications. The results showed that the hatching egg shape index in this study was 75,77 – 77,05%. The lowest egg weight loss was at P0 (0%), while the largest weight loss was at P2 (15%). The highest hatchability was observed in P1 (10%) at 84%. The most effective P1 treatment resulted in the highest egg hatchability in the present study.

Abstract

 One of the main characteristics of chamois leather is its high ability to absorb water; thus, chamois leather is generally used for cleaning and drying. The usual tanning material for chamois leather is fish oil. However, it makes the chamois leather product have a strong odor. This study aims to evaluate the quality of chamois leather from rabbit skin tanned with corn oil. The treatments included the addition of several corn oil concentrations consisting of P0 (30 % fish oil), P1 (10 % corn oil), P2 (20% corn oil), and P3 (30% corn oil). The results showed that corn oil as a tanning material was able to increase the ash content, elongation, tear strength, and water absorption of chamois skin and was able to reduce oil content, and did not change the tensile strength, color and odor of the chamois leather produced. The quality of chamois leather with 30% corn oil indicated the ash content as much as 3.19%, oil content as much as 7.87%, elongation as much as 94.51%, tear strength as much as 46.47 N/mm, water absorption (24 hours) as much as 279.03%; and tensile strength as much as 9.9 N/mm2. It also produced yellowish-white color with no odor. This quality has met the requirements of the Indonesian National Standard (SNI): 0617521990. The study concludes that 30% corn oil can substitute fish oil to produce chamois leather.

Abstract

This study aimed to compare the productivity of Sentul chickens and first-generation Kampung chickens (G1) until the age of 3 months. The research material were Sentul chicken and Kampung chicken. The method used was an experiment method or direct observation with a sample of 174 chickens from each strain. The data collected were egg weight, doc weight, body weight, body weight gain, body measurements, and selection response. Data on egg weight, body weight, and body measurements were analyzed using the average difference test (t-test), while the average value vector of body measurements Sentul chicken and Kampung chicken were analyzed using the T2-Hotelling statistical test. To identify the body size and body shape characteristics of Sentul chickens and Kampung chickens were analyzed using principal component analysis. Data processing used the statistical software Minitab version 18. The results of this study showed that egg weight, body weight at the age of DOC-3 months and body measurements of Sentul chickens were significantly different (P<0.05) higher than the Kampung chickens. This study concludes that the egg weight, body weight, body weight gain, and body sizes of Sentul chickens are higher than Kampung chickens. The size characteristic of Sentul and Kampung chickens is chest circumference. The characteristic of the shape of the Sentul chicken is the length of the wings, while the shape of the native chicken is the width of the chest. The selection response and the heritability value of the Sentul chickens was higher than Kampung chikens.

Abstract

The GDF9 gene is a gene that affects the maturation of oocytes. GDF9 is expressed in oocytes and granulosa cell, it can stimulate granulose cell proliferation and regulate cumulus cell function from pre-ovulation to ovulation. The GDF9 gene is associated with an increase in the ovulation rate and litter size in animals. This study aims to determine the kinship relationship of local goats compared to goats in Asia on prolific traits and to determine the restriction mapping of the GDF9 gene in goats based on the different SNP locations. The local goat comes from the Bligon goat, Kacang goat and Kejobong goats which is compared to the GenBank data (EF446168, EU883989 and KY780296). GDF9 sequences were analyzed using BioEdit and sequencing results to identify Single Nucleotide Polymorphism (SNP) and using NEBCutter V2 to determine the restriction enzyme which recognized the sequence around SNP. The result shows that three variations of SNP were found in exon 2 (g.3615T>C, g.3760T>C and g.3855A>C). Identification of SNP position found 1 SNP position identified by restriction enzyme at g.3855A> C. The identified restriction enzyme is HpaII and MspI. The results of this study are expected to provide genetic information that will be used for further research on the relationship between GDF9 gene polymorphisms to animal prolific.

Abstract

Lactating ewes require high nutrients for basic life requirements and milk production. If not fulfilled, it can have an impact on Negative Energy Balance (NEB) that reduces body weight, Body Condition Score (BCS), and extend the appearance of Postpartum Estrus (PPE). Premating flushing feed is an effort to improve ewe nutrients by adding high nutrition for preparation before mating so that after lambing and suckling, the ewe immediately estrus. This research aimed to evaluate the performance of ewes through premating flushing feeding. The research was conducted at Mendo Galak Farm, Sleman, Yogyakarta. Twenty ewes 2-3 years old with BCS 2-3 divided into two groups; the group with flushing treatment consisted of dried water spinach (Ipomoea reptans poir), concentrate feed with Spirulina sp. (14,92% crude protein, 60,28% total digestible nutrients), and the control group (PS) without Spirulina sp. (crude protein 11,82%, total digestible nutrients 53,20%). Flushing feed was given after a month postpartum as much as 3% dry matter of body weight. The recorded parameter was daily feed consumption, monthly body weight, BCS, and postpartum estrus. The data obtained were tested by an independent T-test with Statistical Product and Service Solution (SPPS ver. 22). The results showed the consumption and digestibility of CP, TDN, and ewe's ADG had a significant difference (P<0.05).  Postpartum estrus (PPE) of flushed ewes had no significant difference (P>0.05), 73.90±11.55 vs. 77.60±14.65 days, respectively. The conclusion was that flushing premating treatment with the addition of Spirulina platensis increased the nutrient intake and digestibility of CP, TDN, and ADG but had not shortened on postpartum estrus of lactating ewes.
