Biodiversitas 20: 671-675. This research was conducted to assess and characterize the genetics of Taro White cattle. Genetic characterizations of this cattle are essential to conservation and breeding program. A total of 18 samples and 4 pairs of microsatellite DNA markers (BM2113, BM1824, INRA023, and ETH225) were amplified by PCR and the products were run on 8% bis-Acrylamide gels. All microsatellite markers were successfully amplified with a mean allelic number of 3.25. Means of observed and expected heterozygosity were found to be 0,25 and 0,628. The Polymorphism Information Content (PIC) values ranged from 0.448 (BM1824) to 0.627 (BM2113) and fixation index were 0.620. The deviation from Hardy-Weinberg equilibrium revealed that Taro White cattle population exhibited significant deviations from Hardy-Weinberg equilibrium (HWE) and possessed a possibility of inbreeding. Abstract. Sapari I, Perwitasari-Farajallah D, Utami Atmoko SS. 2019. The Bornean orangutan (Pongo pygmaeus wurmbii) density in a logging concession of Hulu Belantikan, Central Kalimantan, Indonesia. Biodiversitas 20: 878-883. The Bornean orangutan is currently categorized as a critically endangered species. It is found in natural forests in Borneo, where about 78% of the total population is found outside protected areas, of which 29% is in logging concessions. This study aimed to analyze the density of the orangutan population and the abundance of fruiting plants in a logging concession and Protection Forest (Hutan Lindung) in the Hulu Belantikan forests in Lamandau District, Central Kalimantan, Indonesia. Research and data collection was conducted between December 2013 to October 2014. Orangutan population density was calculated using line transect methodology based on nest counts. Abundance of fruit plants was calculated using the fruit trail method. The highest orangutan density, 4.8 individuals/km2, was found in Protection Forest; and the lowest density, 0.4 individuals/km2, was in [Sopanan] the 2013 logging block. Observations in the logging area indicated that selective logging could alter the structure and gap of the canopy and reduce the proportion of large trees. Changes in forest structure resulted in negative effects on the density of the orangutan population. The remaining degraded forests can still be a valuable resource for the orangutan. As long as the disturbance is not intensive, orangutans will retain access to the less disturbed forest nearby and to forests that are still connected to primary forests. The existence of Protection Forest and old logged-over forests around logging blocks are very important for conservation, providing refugia for orangutans and other species when logging occurs.

Abstract

Source, harvesting, conservation status, threats and management of indigenous plant used for respiratory infections and related symptoms in the Limpopo Province, South Africa. Biodiversitas 20: 789-810. This survey explored Bapedi traditional healer’s (THs) practices pertinent to native plants used to treat respiratory infections (RIs) and related symptoms (RSs). Semi-structured questionnaires and participatory observations were used to gather information from 240 THs in the Limpopo Province, South Africa. 186 plants from 75 families were harvested by these THs, mainly from the communal lands (81.2%), throughout the year. Plant parts used for RIs and RSs remedies was destructively harvested in wilderness compared to homegardens. Most (n=174) species from which these parts are obtained appears on the South African National Red Data List of plants, with 88.5% having a list concern status. This included Adansonia digitata, Boscia albitrunca, Catha edulis, Securidaca longepedunculata and Sclerocarya birrea which are also protected under the National Forest Act of 1998 (Act no. 84 of 1998). A further, 8.6% (n=15) of Red Data Listed plants are of conservation concern, with various status namely near threatened (38.3%), declining (20%), data deficient (13.3%), critically endangered and vulnerable (3.3%, for each), as well as endangered (6.6%). There were both consensus and disjunction amongst THs and Red Data List regarding the status of plants in the wild. This study provides valuable data for the conservation of medicinal plants in Limpopo Province.

Abstract. Triandiza T, Zamani NP, Madduppa H, Hernawan UE. 2019. Distribution and abundance of the giant clams (Cardiidae: Bivalvia) on Kei Islands, Maluku, Indonesia. Biodiversitas 20: 884-892. Giant clams, ecologically important bivalves in coral reefs, are under anthropogenic pressures in most parts of their range, necessitating the study of population status for conservation management. Here, we assessed species composition, distribution, size density, and habitat condition of giant clams in Kei Islands. A total of 9 reefs around the islands (Dar 1, Dar 2, Pulau Kur, Pulau Tanimbar Kei, Pulau Adranan, Pulau Dullah Laut, Difur, Labetawi, dan Ohoidertoom), were surveyed using quadrat-transect line in September-December 2017. We found five species of giant clams, i.e., Tridacna crocea, T. maxima, T. squamosa, T. noae, and Hippopus hippopus. Of these species, we report a new record for T. noae which was previously not known to be present in the region. The overall density was recorded at 0.0428 individual/m2. Juveniles clams were fewer than the adults clam, this may indicate that the survival rate of recruit is low. Most individuals were found living on dead coral algae substrate (75 %). Based on the findings, this study suggests that giant clam population in Kei Islands are imperiled, indicated by low population density and local extinction for species T. gigas and T. derasa. It is recommended to establish a protected area and restocking of giant clams by through implementing sasi laut (traditional law) with a minimum period of 5 years.

Species confirmation of juvenile cloudy grouper, Epinephelus erythrurus (Valenciennes, 1828), based on a morphologic analysis and partial CO1 gene sequencing. Biodiversitas 20: 914-921. The genus Epinephelus is the most species among the genera within the subfamily Epinephelinae. Species identification techniques for groupers, especially in the Epinephelus, are commonly based on color patterns and a suite of morphological characters, including body shape and the size and number of fins, scales and gill rakers. In some species, juveniles are morphologically distinct from adults of the same species, making morphological identification highly problematic. This present work will provide some morphological description or variations of juveniles that have been identified as Epinephelus erythrurus based on CO1 sequences. Further, the present study demonstrates that a molecular genetic technique, based on partial sequencing of the mitochondrial CO1 gene, may be used for the rapid species confirmation of every developmental stage and phase of an organism (juvenile E. erythrurus). Two DNA sequences of E. erythrurus from the Pangandaran District (7o43’8.31”S 108o30’11.59”E) have been submitted to GenBank under accession numbers KP998441 and KP998442.

Impact of Bukit Dua Belas rainforest transformation to oil palm plantation on phylogenetic of soil bacterial communities in Sarolangun, Jambi, Sumatra, Indonesia. Biodiversitas 20: 811-818. Land use change from forest to oil palm plantation at Sumatra could decrease biodiversity, including bacterial diversity. The case of Bukit Dua Belas transformation from forest to oil palm plantation was gotten for measuring shift community of soil bacterial in both areas. The diversity of bacterial communities from rainforest and oil palm plantation topsoil in Sumatra was studied using pyrosequencing of 16S rRNA gene and common biodiversity indices. Phylogenetic approach was used to reveal the community shift of bacterial phyla and genera in both areas. Ecological approach was carried out by measuring soil pH, TC (total carbon), TN (total nitrogen), AP (available phosphorous), bacterial diversity with Shannon and Simpson indices, and bacterial richness with Chao1-ACE index and OTUs. Bacterial diversity and richness on lowland forest topsoil and oil palm plantation soil were not different, as soil pH, TC, and TN as substrate factors were not different significantly. The majority of sequences related to Acidobacteria (56.33%), Proteobacteria (27.43%), Actinobacteria (7.11%), and Cyanobacteria (5.55%) were from forest; whereas those related to Acidobacteria (50.11%), Proteobacteria (31.63%), Actinobacteria (7.58%), Chloroflexi (2.60%), and Gemmatimonadetes (2.71%) invented from oil palm plantation. Acidobacteria was the most dominant phyla in both habitats, because soil pH in both areas was acidic (3.77 - 4.80 pHH2O). The genera of alpha-proteobacteria dominated in genera phylotype of bacterial 16S rRNA phylogenetic revealed in both forest and oil palm plantation topsoil. The most genera in phylogenetic tree are Burkholderia from Beta-proteobacteria. The bacterial community shift occurred in forest transformation, even though the oil palm plantation showed more bacterial phyla and genera than the lowland rainforest.

Regardless of the several attempts of the early and recent studies to separate the wild species of Cucumis from the cultivated ones, there is still taxonomic confusion brought about by the similarities in morphology of the genus. In a gene bank with so many species and accessions of Cucumis stored, it is appropriate to delineate these numerous accessions to save time and resources as well. This study aims to delineate fifty selected Cucumis accessions based on leaf architecture. Using Unweighted Pair Group Method using Averages (UPGMA) and Euclidean distance coefficient, a cluster analysis for the fifty Cucumis accessions was done. A dendogram with cophenetic coefficient of 0.9606 supported the clustering of the Cucumis species and accessions. At Eucledian distance of 1.5 two major clusters were formed on the basis of secondary vein spacing. Cucumis melo accessions separated from all the remaining accessions of C. myriocarpus, C. metuliferus, C. anguria and C. anguria var longaculeatus for having an increasing towards the base secondary vein spacing while the rest have irregular pattern of secondary vein spacing. Further sub-clustering of the remaining accessions comprising four species were delineated on the basis of tertiary vein (C. myriocarpus), tertiary vein angle to primary (C. metuliferus), and blade class (C. anguria and C. anguria var longaculeatus). Laminar shape delineated C. myriocarpus accessions from each other, apex angle for C. metuliferus accessions, and primary vein size for C. melo accessions. Results implied that leaf architecture is a good tool to classify the numerous accessions of Cucumis.

Abstract. Pandiangan D, Silalahi M, Dapas F, Kandou F. 2019. Diversity of medicinal plants and their uses by the Sanger tribe of Sangihe Islands, North Sulawesi, Indonesia. Biodiversitas 20: 621-631. The Sanger tribe is an indigenous tribe in Sangihe Islands, North Sulawesi, who still continue to use traditional medicinal plants. The objectives of this study were to document the diversity of medicinal plants and their general uses by the Sanger tribe in the selected villages of Sangihe Islands and to identify plants that have anti-cancer potential. The study was conducted with ethnobotanical approach using survey and interviews among 90 respondents selected from 9 study villages located in 3 sub-districts of Sangihe Islands. The study showed that 118 species of medicinal plants belonging to 47 families had been used by the Sanger tribe to treat 39 different types of diseases. Thirteen (13) species each were used for treating fever and ulcers, 12 species in the treatment of poisoning, 10 species each for cough and kidney diseases, 8 species for each of the diabetes mellitus and cancer or tumors, and 7 species each for itching, hypertension, physical injury and blood circulation disorders. For other diseases, the number of plant species used as medicines varied from 1 to 6. Leaves were the most predominantly used part for medicinal purposes as this was used in case of 73 species, followed by stems (37 species), roots (29 species), flowers (25 species), fruits (20 species) and seeds (17 species). Twenty percent (20% of the recorded medicinal plants were consumed directly and others after different methods of processing and preparation, such as boiling (51%), crushing (15%), roasting (2%), soaking (16%) and pasting (7%), and a species can be used more once. A utilization of Dischidia nythesiana as medicines for cancer/tumor by the Sanger tribe has not been reported before.

Biodiversitas 20: 819-823.   Conservation management in the buffer and transition zones of Biosphere Reserves play an important role in ensuring enhanced core zone protection. Tambolongan and Polassi are two islands located in the administrative region of South Sulawesi, Indonesia, and part of the transition zone area of UNESCO’s Taka Bonerate-Kepulauan Selayar (TBKS) Biosphere Reserve (BR). These two islands can be considered as some of the most populated islands in the region, containing a variety of human activities. These human activities, such as the ever expanding agriculture and the building of human habitation have put pressures toward ecosystem and species conservation in the BR. Bird are often the focus of conservation efforts, due to their roles in supporting the ecological functioning of their habitats. This study aims to provide baseline data of bird diversity in Tambolongan and Polassi that can be used as a proxy indicator of ecosystem health and input to conservation strategies within the BR area. A Visual Encounter Survey (VES) method is used to examine bird diversity index. Results from the observation recorded a total of 29 species were found in both islands, with 5 listed as protected species and 3 categorized as migratory species. The comparison between the two islands showed that the bird diversity index (H’) differs only slightly (H’Tambolongan=2.98; H’Polassi=2.71). This study provided the initial attempt in understanding the role of habitats or land cover types, guild types and human interferences in determining the diversity of bird population - and related ecosystem health - in the area. Such knowledge is essential in providing the information necessary for sustainable management practices. This work represents the first study to inventorize avian species diversity in the transition zone of TBKS BR.

Abstract. Rahmawati D, Khumaida N, Siregar UJ. 2019. Morphological and phytochemical characterization of susceptible and resistant sengon (Falcataria moluccana) tree to gall rust disease. Biodiversitas 20: 907-913. Gall rust disease has severely attacked sengon tree (Falcataria moluccana Miq. Barneby & J.W. Grimes)) and it rapidly spread in tree plantations causing the decrease of wood productivity. Although sengon is widely cultivated in several areas in Indonesia, there are limited studies on secondary metabolite composition of its wood. This research was aimed to study differences on morphology, wood anatomy and secondary metabolites content of resistant and susceptible sengon tree trunk to gall rust disease. Both resistant and susceptible tree samples to gall rust were selected from the same plantation plots to minimize environmental effects. Structure of wood samples was observed using Scanning Electron Microscope (SEM) JSM-5310LV. Seven types of secondary metabolites, i.e., alkaloids, flavonoids, saponins, phenolic hydroquinones, tannins, triterpenoids, and steroids were qualitatively analyzed from powdered wood samples. Based on field observations, the gall rust fungi changed the color, texture, pores of the tree surface and decreased wood density. In microscopic observations, sengon trunk, which is resistant to gall rust disease, showed organized and regularly structured wood anatomy and did not show hyphae of the infecting fungi inside the wood. The fungi damaged susceptible sengon trunk as indicated by the dispersed and irregular structure of wood tissue, while the presence of fungal hyphae among wood cells was detected. Phytochemical analyses indicated that trunk of sengon containing flavonoids, saponins, phenolic hydroquinones, tannins, triterpenoids, and steroids, except alkaloids. Sengon trunk, which is resistant to gall rust disease, had stronger reactions of flavonoids, saponins, triterpenoids, and steroids compared to that susceptible one.
