Abstract. David D, Daniel R, Seecharran D, 2019. Antimicrobial properties of Chlorocardium rodiei on Pseudomonas aeruginosa, Bacillus spp. and Candida albicans. Nusantara Bioscience 11: 128-132. This research seeks to investigate the antimicrobial properties of Chlorocardium rodiei leaves and seeds on Pseudomonas aeruginosa, Bacillus spp. and Candida albicans. It also seeks to extract and identify the phytochemical compounds presented in leaves and seeds. The crude plant extract was obtained using the maceration method of extraction (ethanol and acetone as solvent) followed by rotary evaporation. Phytochemicals were analyzed from the crude extract using standard phytochemical procedures. 1%, 50% and 100% concentrations of each extract were prepared and used in the disk diffusion assay against C. albicans, P. aeruginosa, and Bacillus spp. and compared against Ciprofloxacin and Erythromycin for the bacteria and Fluconazole for the fungus. The results showed that the extract from the leaves possesses weak (?12 mm) to moderate (>12 ?15mm) inhibitory effect, while the extract from the seeds had weak (<12mm) inhibitory effect on the microorganisms. The phytochemical compounds tannins, essential oils, saponins, and phenols were tested positive from the crude extract. For the first time, the antimicrobial properties of C. rodiei’s leaves and seeds were evaluated and showed to possessed antimicrobial properties.

Abstract. Authors. 2019. The activity phagocytosis of isoeugenol-ester compound on Mus musculus macrophage cell. Nusantara Bioscience 11: 117-122. Chemotherapy is drugs therapy for cancer. However, chemotherapy could damage normal cells and decrease immune system. For this reason, it is necessary to find immunostimulant compound that is able to increase immune system. Isoeugenol-ester is unnatural compound that has been tested for its cytotoxicity against cancer cell and showed positive result. Nevertheless, ability of Isoeugenol-ester for inducing phagocytosis is still unknown. Therefore, bioassays study using in vitro phagocytosis were performed. Latex beads methods using antigen model in 5 isoeugenol-ester concentration series: 6, 25; 12, 5; 25; 50 and 100 ?g/mL were tested to Mus musculus peritoneal macrophage cell. The optimum concentration of Isoeugenol-ester compounds in inducing phagocytosis activity of M. musculus macrophage cell was 12.5 ?g/mL.

Abstract. Hardjo PH, Susanto DPS, Savitri WD, Purwanto MGM. 2019. Shoot multiplication of Pogostemon cablin var. Sidikalang and patchouli oil profile. Nusantara Bioscience 11: 123-127. Pogostemon cablin Benth. is a plant producing patchouli oil, which mostly consists of patchouli alcohol compound. Patchouli oil has great potentials in the world market because of its stability and high price. In this study, in vitro multiplication of Sidikalang variety of Acehnese patchouli shoots was done on solid and liquid Murashige & Skoog (MS) medium. This study aimed to determine the effect of cytokinins in various combinations of shoot multiplication and to compare the patchouli oil yield of in vitro and ex vitro culture. In vitro multiplication of Acehnese patchouli shoots by using solid MS medium with addition of 0.2 ppm benzyl aminopurine (BAP) and 0.2 ppm Kinetin resulted in shoot explants with an average growth index of 82.198 ± 0.690. Patchouli oil extraction was done on 7 weeks old in vitro shoot explants cultured on solid MS medium + 0.2 ppm BAP + 0.2 ppm Kinetin using water distillation method. In vitro shoots yielded 2.5% patchouli oil and contained ± 35% patchouli alcohol compound, whereas ex vitro shoots produced 4% patchouli oil and contained ± 25% patchouli alcohol compound. The qualitative analysis by using thin layer chromatography (TLC) showed that there were similarities in the number of spot and Rf value for each spot of ex vitro and in vitro patchouli oil.

The effect of ethanol extract of fruit peel jengkol (Archidendron pauciflorum) to spleen histological structure of streptozotocin-induced diabetic rats (Rattus norvegicus). Nusantara Bioscience 11: 157-161. Diabetes Mellitus (DM) is a chronic endocrine disorder associated with several secondary complications. The spleen is a lymph organ with the main function of filtering red blood cells, immune response and playing any kind of role in diabetes. Despite the currently available anti-diabetic drugs, there are need remedies for alternatives which are economical and safe. One alternative medicine well known by local Indonesian communities to treat DM is the fruit peel of Archidendron pauciflorum. This study aimed to evaluate the histological changes in the spleen following administration of ethanol extract of fruit peel Archidendron Pauciflorum (EEJFP) in the streptozotocin (STZ) - induced diabetic rats. Diabetic rats were used in this study consisting of six groups of four animals each. The treatments given are NC (CMC solution 0.5%), Positive Control (CMC solution 0.5%), G (glibenclamide), Treatment 1, 2, and 3 (EEJFP dose 385; 770; 1540 mg/kg bw). All of the treatment was administered orally to the experimental rats 14 days following induction. The spleen was collected and the histological study was performed using hematoxylin and eosin (H&E). The parameters observed were the number of necrosis and cell damage score, including fat degeneration, hydropic degeneration, and inflammatory cell. The result showed that EEJFP at dose 385 mg/kg bw and 770 mg/kg bw gave significant recovery (p<0.05) on spleen damage by the decreased of the number of necrotic cells and cell damage score which lower than other treatments and compared to negative control group. It was concluded that doses of 385 mg/kg bw EEJFP are the optimum dose to improve the histological structure of spleen damage in diabetic rats.

Abstract. Srimariana ES, Apituley DAN. 2019. Antioxidant activity of extracted green algae silpau (Dyctyosphaeria versluysii). Nusantara Bioscience 11: 153-156. Silpau (Dictyosphaeria versluysii) is a green algae that is widely available in Southwest Moluccas regency, living on coral reefs and not classified as seasonal plants. Silpau has long been used by local people, generally in the form of processed vegetables or colo-colo (traditional food). Except for its nutritional value, comprehensive information about silpau still unknown. Therefore, this study was carried out to determine its potential as an antioxidant. The phytochemical content of silpau was analyzed according to the standard method. The antioxidant activity of green algae silpau extract was carried out by reducing DPPH (1,1-diphenyl-2-picrylhydrazyl) free radicals. Phytochemical test of silpau revealed that silpau contain terpenoid compounds. The result of the study showed that the IC50 value of methanol extract of silpau was 547.97 ppm, indicated that silpau methanol extract categorized as a weak antioxidant.

Abstract. Purba JH, Sasmita N, Komara LL, Nesimnasi N. 2019. Comparison of seed dormancy breaking of Eusideroxylon zwageri from Bali and Kalimantan soaked with sodium nitrophenolate growth regulator. Nusantara Bioscience 11: 146-152. Eusideroxylon zwageri is a very strong and durable wood, which can be used as building materials. This wood plant needs 6-12 months to germinate, but it can be shorted with special treatment (e.g. cracking). Therefore, it is important to conduct research to stimulate E. zwageri seed germination using sodium nitrophenolate growth regulator. The purpose of this study was to analyze the comparison of the fastest time to breaking E. zwageri seed dormancy from the Bali and Kalimantan island. The research method was factorial randomized block design, using the X factor (soaking time) and Y factor (sodium nitrophenolate growth regulator). There were four soaking time treatment, i.e., immersed (soaked in a really short time), 12 and 24 hours soaking time. While the Y factor was the concentration of sodium nitrophenolate with 4 treatments: without sodium nitrophenolate solution or 0 mL (control), soaked in a 1 mL/L of water, 2 mL/L of water and 3 mL/L of water solution. The study results showed the best treatment of E. zwageri seeds germination speed was soaked for 24 hours, and by giving 0,3% sodium nitrophenolate concentration, while the interactions of both showed no significant effect. The soaked time treatment, sodium nitrophenolate concentration, and the interaction of both show no significant effect on germination percentage. The breaking of E. zwageri seed dormancy from Kalimantan has a shorter time to germinate than E. zwageri seeds from Bali.

Abstract. Lestari NKD, Deswiniyanti NW, Astarini IA, Arpiwi NLM. 2019. Callus and shoot induction of leaf culture Lilium longiflorum with NAA and BAP. Nusantara Bioscience 11: 162-165. Lilium longiflorum Thunb., an Easter lily, is a common ornamental plant used as potted plant, cut flower, cosmetic and medicine. The research on tissue culture technique to induce shoots and callus is expected to increase the yield of lilies quickly and efficiently. This study aims to determine the effect of plant growth regulator combination of Naphthalene Acetic Acid (NAA) and Benzyl Amino Purine (BAP) on the development of leaf culture and the best concentration for callus induction and shoots of lily plants. This study was conducted in a completely randomized design using leaf explants in Vacin and Went medium, with combination of NAA and BAP (0; 0.5; 1 mg..L-1 ) as the treatments. The result of the eight-week observations shows that the plant growth regulators significantly affected either in days initiation callus, days initiation of shoot, percentage of callus, percentage of shoot, diameter of callus, no of shoot and length of shoot. The combination of 1 mg.L-1 NAA and BAP had the best effect in the percentage of explants forming shoots (100%), means number of shoots (5.8), and means length of shoot (11.6 cm).

Abstract. Nahdi MS, Kurniawan AP. 2019. Ethnobotanical study of medicinal plants in karst environment in Gunung Kidul, Yogyakarta, Indonesia. Nusantara Bioscience 11: 133-141. The use of traditional medicinal plants has been a culture of the people in Yogyakarta including in Gunung Kidul District. This research aims to reveal information on medicinal plants in karst environment in Gunung Kidul, Yogyakarta, Indonesia by focusing on how medicinal plants are used, managed and inherited by the community. The research was conducted in Planjan Village and Giricahyo Village in Gunung Kidul District from June to September 2018. The method used in collecting data was exploratory survey and Participatory Rural Appraisal. Data of community knowledge and uses of medicinal plants were obtained through in-depth interview and structured interview. The identification of species was applied to every plant used. The study found that there are 46 species from 26 families used by the people in the two villages for medical treatments with the largest number of species is from Zingiberaceae family, followed by Fabaceae and Rutaceae. The medicinal plants are mostly packaged into six herbal medicinal plants called jamu beras kencur, kunir asem, paitan, kebyokan (uyup uyup), sirih and temu lawak with different compositions and efficacies. The result showed that people's knowledge about medicinal plants is obtained from elders who have a long experience in making concoction and trading the medicinal plants as traditional herbal medicine. In addition, the people are also obtained additional knowledge from any sources to make the medical plants to have economic value to improve their welfare. Most of the medicinal plants are obtained from the outside of the villages since it is impossible to cultivate in karst soils. Thus, the study is expected to help develop the local potential of medicinal plants to become more modern, can support the conservation of local wisdom and plant biodiversity, as well as be used as a source of material for developing national database on medicinal plant and its usage.

Nusantara Bioscience 11: 142-145. Bhoi cake is one of the traditional Acehnese dry snacks with basic ingredients of rice flour or wheat flour, castor sugar, eggs and vanilla. This study aims to evaluate nutritional value and consumer acceptance by organoleptic tests on bhoi cake substituted with pumpkin flour. Design of the study was a Completely Randomized Design (CRD) with 4 treatments and three replications. The treatments was the various level of pumpkin flour substitution (0%, 20%, 30%, and 40%). Consumer acceptability with the organoleptic test was carried out by 20 panelists, namely students of the Faculty of Teacher Training and Education Syiah Kuala University using a 5 points hedonic scale. The results showed that the nutritional value of bhoi cake with pumpkin flour substitution was good and have appropriate nutritional value. Sensory evaluation by organoleptic test showed that 30% pumpkin flour substitution was the best formula preferred by the panelist. Taste, aroma, color, texture and overall acceptability of formula C (30% substitution of pumpkin flour) of bhoi cake had the highest score (likes), with the characteristics of the bhoi cake were golden yellow, fragrant aroma, sweet taste, and soft texture.

Abstract. Susanto D, Auliana, Kusuma R, Amirta R. 2019. Effect of organic manure fertilizer on the growth of Macaranga gigantea. Nusantara Bioscience 11: 166-171. M. gigantea planting research has been conducted in several habitats such as natural forest gaps, drained peatland and soil with highly degraded land for rehabilitation. This study investigated the effect of organic manure fertilization on soil, tissue chemical properties and growth performance of Macaranga gigantea planted at dense space of 2,5 m x 2,5 m. The experiment used a randomized block design with treatment of fertilization using goat manure with five gradual concentrations namely P0 (0 g), P1 (100 g), P2 (200 g), P3 (300 g) and P4 (400 g) in which each treatment consisted of three groups with 20 replications for each group, totaling 300 seedlings observed. The results showed that organic manure fertilization promoted stem height, stem diameter, leaf number and nutrient concentrations in the leaves of four-month-old M. gigantea. The best growth performance was gained from the treatment of P3 (300 g), followed by P2, P1, P4, and P0. The highest nutrient contained in leave tissue was potassium, followed by calcium, magnesium, nitrogen and available phosphorus.
