Buletin Plasma Nutfah (BPN) is an open access scientific journal published by The Indonesian Center for Agricultural Biotechnology and Genetic Resources Research and Development (ICABIOGRAD), The Indonesian Agency for Agricultural Research and Development (IAARD), Ministry of Agriculture. This peer-refereed journal covering the area of genetic resources including exploration, characterization, evaluation, conservation, diversity, traditional knowledge, management and policy / regulation, of all kinds of genetic resources: plants, animals, fishes, insects and microbes. Manuscripts submitted to this journal are those that have never been published in other journals. This journal is published in one volume of two issues per year (June and December). We invite authors to submit the manuscripts to this journal in English or Indonesian. Abstract

Tanaman Bacopa australis dan Alternanthera reineckii mempunyai nilai ekonomi cukup tinggi sebagai tanaman hias akuatik, sehingga berpotensi untuk dikembangkan. Pertumbuhan minimal merupakan teknik penyimpanan in vitro yang banyak digunakan dengan menggunakan senyawa penghambat tumbuh seperti paklobutrazol (PBZ), sisosel (CCC), ansimidol, serta komponen osmotik seperti sorbitol dan manitol. Teknik kultur in vitro mempunyai peran penting untuk penyimpanan biakan, karena biakan dapat disimpan sampai beberapa bulan tanpa harus dilakukan subkultur. Tujuan penelitian ini adalah untuk mengetahui pengaruh zat penghambat paklobutrazol dan interaksinya dengan zat pengatur tumbuh benzil adenin (BA) untuk menghambat pertumbuhan biakan serta visual biakan selama penyimpanan. Bahan yang digunakan adalah tunas in vitro B. australis dan A. reineckii umur 4 bulan, media yang diuji adalah media dasar Murashige dan Skoog (MS) + PBZ (0,1, 0,3, 0,5, dan 0,7 mg/l) + BA (0 dan 0,1 mg/l). Penelitian dilaksanakan di Laboratorium Kultur Jaringan BB Biogen pada bulan Januari sampai Juli 2019. Hasil penelitian menunjukkan bahwa PBZ 0,1−0,7 mg/l dapat menghambat pertumbuhan tinggi tunas, jumlah tunas, jumlah akar, panjang akar, dan jumlah daun dari biakan in vitro B. australis dan A. reineckii demikian pula interaksinya dengan sitokinin BA 0,1 mg/l, yang membuat visual biakan tetap tegar sampai 6 bulan setelah tanam. Formula media yang diperoleh diharapkan dapat diaplikasikan untuk penyimpanan plasma nutfah tanaman akuatik yang sejenis.

Abstract

Javan rhino (Rhinoceros sondaicus) is one of the rare animals with critically endangered status. The Ujung Kulon National Park (UKNP) is one of javan rhino habitat in Indonesia. Every year the monitoring program for javan rhino in UKNP was performed to identify the animals. This research was aimed to analyze the population structure of javan rhino based on the records data from 2011 to 2019 in UKNP. Research showed that the natural increase (NI) and birth rate (BR) values were 17.34% (moderate) and 67.33% (high) respectively. The net return rate (NRR) value was 15.38% (male) and 14.28% (female). The NRR value in the present study was lower than 100% and caused by less number of animals in a population for 30 years of breeding length. Despite this, the inbreeding rate of javan rhino at UKNP in 2019 was 0.01 (low). It was concluded that the natural increase of javan rhino at UKNP showed a good parameter but the population number needs to be increased.

Abstract

ndonesia is well-known as a center of diversity of banana in the world. Here, wild Musa acuminata is growth widely in some areas, included Java islands. However, the genetic diversity study of wild banana, especially in Java Island is still limited. In this study, we observed the genetic diversity of wild banana in Mount Halimun-Salak National Park (TNGHS), West Java, Indonesia and studied its resistance to Fusarium oxysporum f.sp. Cubense tropical race 4 (Foc TR4) using molecular markers. The plant materials were collected and phenotyped in some different locations of conservation area TNGHS from 7 to 11 October 2019. Principal Component Analysis (PCA) was calculated to estimate the genetic diversity on eight accessions of wild M. acuminata based on eighteen morphological characters. Moreover, the study of its resistance traits to Foc TR4 was analyzed by using SCAR markers i.e. ScaU1001 and ScaS0901. The most contributed characters on principal component 2 (PC2) was showed on number of fruits with the value of 0.586 and bract apex shape with value of -0.648 on principal component 3 (PC3). For the resistance study, the eight accessions of wild M. acuminata produced specific band of ScaS0901 marker. The knowledge of the genetic diversity and the resistance study of wild M. acuminata in TNGHS would be useful for conservation strategy and bioprospecting of the wild banana in TNGHS.

Abstract

Sebanyak 25% ubi jalar yang dikoleksi di Bank Gen Pertanian Balitbangtan-BB Biogen berasal dari Papua. Informasi karakter kualitatif umbi ini sangat penting untuk diketahui. Penelitian ini bertujuan untuk mengetahui keragaman karakter umbi ubi jalar asal Papua. Materi yang digunakan adalah 349 aksesi yang berasal dari Papua yang ditanam secara terus menerus di Kebun Percobaan Cikeumeuh, Bogor. Pengamatan dilakukan setiap tahun sejak 2013 sampai dengan 2016. Karakter umbi yang diamati meliputi: bentuk umbi, tebal korteks, warna predominan kulit umbi, intensitas warna predominan kulit umbi, warna predominan daging umbi, formasi umbi, dan kadar bahan kering umbi. Terdapat keragaman pada karakter umbi lokal asal Papua yang diamati. Warna kuning dan krem mendominasi warna daging umbi, sedangkan untuk warna kulitnya cenderung tersebar merata antar variasi warna yang teramati, hanya warna kulit kuning yang tidak ada. Formasi umbi yang tersebar juga berjumlah paling banyak dibanding dengan variasi formasi umbi lainnya. Kadar bahan kering umbi asal Papua rata-rata memiliki kandungan yang tinggi (>30%).

Abstract

Tujuan penelitian untuk mengevaluasi performa sumber daya genetik babi lokal (Sus scropa domesticus) yang meliputi aspek produksi, reproduksi, manajemen, dan persepsi budi daya. Pelaksanaan penelitian selama 6 bulan, yaitu Januari–Juni 2020 di Pulau Timor, Provinsi Nusa Tenggara Timur (NTT). Penentuan lokasi dilakukan secara purposive sampling berdasarkan kriteria ketersediaan populasi dan sampel, struktur umur ternak, dan status sosial budaya. Teknik pengambilan sampling menggunakan metode conveinance sampling baik terhadap responden peternak dan ternak babi sehingga penelitian ini terdiri atas 61 responden peternak dan 91 ekor babi lokal dengan kelas umur berbeda. Teknik memperoleh data menggunakan teknik wawancara, observasi, dan dokumentasi. Jenis data terdiri atas data primer dan sekunder. Analisis data menggunakan statistik Importance-Performance Analysis dan dijelaskan secara deskriptif. Hasil penelitian menunjukkan bahwa eksistensi sumber daya genetik babi lokal di Pulau Timor masih menunjukkan nilai yang rendah, baik dari aspek performa ternak (tidak efisien dengan nilai skor 365), manajemen budi daya (tidak efisien dengan nilai skor 291), sosial budaya (tidak efisien dengan nilai skor 359), dan lingkungan (terganggu dengan nilai skor 451). Kematian anak masih tinggi mencapai 25,6 ± 3,6%/periode kelahiran (KV 14,1%) dan interval kelahiran yang panjang yaitu rata-rata 10,4 ± 2,0 bulan (KV 19,0%). Kedua hal ini merupakan penghambat pertumbuhan dan perkembangan populasi babi lokal sehingga semakin menurun dari waktu ke waktu. Adanya intervensi kebijakan, teknologi, dan manajemen budi daya yang menyangkut aspek genetik, manajemen, dan sumber daya manusia dapat meningkatkan perbaikan performa dari babi lokal di Pulau Timor.

Abstract

Termitomyces is a popular wild edible mushroom in paleotropic region including Indonesia. This mushroom is commonly harvested by people around rural area in Indonesia while foraging for food alternative. However, the information regarding Termitomyces usage in urban area is very limited. The objective of this current study is to provide preliminary information about diversity and biological characters of Termitomyces from city forest in Gunungpati, Semarang City, Central Java. Mushroom exploration is done by opportunistic sampling method and identification was carried out by macroscopic observation of morphological characteristics. A total of  7 species of Termitomyces were found  in this study. They were Termitomyces cf. medius, Termitomyces cf. globulus 1, Termitomyces cf. globulus 2 Termitomyces cf. bulborhizus, Termitomyces cf. microcarpus, Termitomyces cf. clypeatus, and Termitomyces cf. eurhizus.  This study is the first report of Termitomyces inventory in the research site and contributing to Indonesia's diversity data.

Abstract

Biodiversity is a valuable asset which should be preserved and protected. Gunung Gede Pangrango National Park (GGPNP) is one of the conservation areas with a high level of biodiversity in Indonesia. Currently, mushroom diversity in GGPNP is rarely reported. Inventory of mushroom diversity is the first step of actions to maintaining germplasm and maximizing its potential usagein the future. In this paper, we explain how to use macroscopic characters to help identification of macro mushroom. The fungi collected from GGPNP were 19 species and 14 genera, including: Armillaria sp. 1, Armillaria sp. 2, Auricularia sp., Daedalea sp. 1, Daedalea sp. 2, Fomitopsis sp., Ganoderma sp. 1, Ganoderma sp. 2, Marasmius sp., Microporus sp. 1, Microporus sp. 2,Mycena sp., Phellinus sp., Polyporus sp., Rigidoporus sp., Russula sp., Sistotrema sp., Xylaria sp. 1, and Xylaria sp. 2. These mushroom could be potentially used as food and medicinal source.
