Abstract

ABSTRACT Propolis (bee glue) is a natural resinous product of honey bees which collected from exudates and plant buds, rich in biochemicals constituents including mostly flavonoids, phenols and various acids bond. These compound are believed to be responsible as immunomodulatory agents. The study aims to determine the immunomodulatory activity of ethanolic extract of propolis to the CD4+ and CD8+ T cells producing IFN-γ cytokines and analyze the differences immune responses between control and treatment group by in vivo. Stages include animal acclimation for ± 1 week, preparation of Ethanolic Extracts of Propolis / EEP, Oral Administration with doses levels of 0 mg / kg BW; 50 mg / kg BW (DI); 100 mg / kg BW( DII); 200 mg / kg BW (DIII) for 2 weeks, isolation of lymphocyte cells from spleen, flowcytometry analysis to asses cell number and surface molecule expression. Data was analyzed using Kruskal Wallis Test with α = 0,05 and followed by Mann Whitney Test by SPSS 16.0 for windows with complete randomized design. The results showed that a dose of 50 mg / kg BW was increases the relative number of CD8+ T cells producing IFN-γ cytokines significantly (p <0.05) compared with controls. However, at the same dose the relative number of CD4+ T cells producing IFN-γ cytokines was decreased significantly (p<0,05). Based on this case, its dose supposedly that the ethanolic extract of propolis play role in maintaining the balance or homeostatic of IFN-γ cytokines production by T cell subsets. Dose of 100 mg/kg BW and 200 mg / kg BW could decrease the relative number of activated CD4+ T cells producing IFN-γ cytokine significantly compared to controls. Abstract

Penelitian ini bertujuan untuk mengetahui jenis amfibi yang dapat ditemukan di kawasan OWA Cangar dan Air Terjun Watu Ondo, TAHURA R.Soerjo, mengetahui karakter habitatnya, serta mengetahui jenis yang berpotensi sebagai bioindikator. Pengambilan data mulai November 2013 hingga Maret 2014 menggunakan metode  Visual Encounter Survey (VES), total usaha 13 jam, 3 pengamat. Lokasi pengambilan data pada ketinggian 1400-1700 mdpl. Faktor abiotik yang diukur meliputi  pH air, suhu (udara & air), dan kelembapan udara. Ditemukan lima jenis amfibi yang seluruhnya tergabung dalam ordo Anura dari lima famili berbeda, yakni katak pohon emas (Philautus aurifasciatus Schlegel, 1837), katak serasah (Leptobrachium hasseltii Tschudi, 1838), bangkong kerdil (Limnonectes microdiscus Boettger, 1892), kongkang jeram (Huia masonii Boulenger, 1884), dan kodok buduk (Duttaphrynus melanostictus Schneider, 1799). Tidak ditemukan satupun spesies dari ordo Caudata maupun Gymnophiona. P. aurifasciatus kerapkali ditemukan pada tumbuhan Psychotria sp., Dypsis lutescens, dan Ficus sp.. L. microdiscus mempunyai habitat daerah hutan dari dataran rendah hingga tinggi. Habitat H. masonii selalu terkait dengan sungai berbatu yang berarus deras dan air jernih. L. hasseltii dapat ditemukan di daerah tinggi di antara serasah hutan. D. melanostictus mempunyai habitat di dekat hunian manusia atau wilayah terganggu. Spesies yang berpotensi besar sebagai bioindikator adalah P. aurifasciatus, H. masonii sebagai indikator perairan jernih, dan D. melanostictus sebagai indikator wilayah terganggu. Abstract

Biliary atresia iscondition caused by Rotavirus (RRV) infection. The aims of this study were to know the immune responses of mice model of biliary atresia treated with corticosteroid.Mice were splitinto 3 treatment groups: control (K), RRV injection (R), and RRV injection in the present of dexamethasone(R+D). In R treatment, the baby mice born in <24 hours were injected with 20 µl of phosphate buffered saline containing 1.5 x 106 fluorescence-forming units Rhesus Rotavirus (RRV). First termination was performed in the day 7 to 14, while second termination was done in the day 14 to 21. The dosage of dexamethasone which is applied in this experiment is 0.5mg/kg body weight.Immunocompetent cells were isolated from spleen,  and cell surface molecules were then analyzed by flowcytometry. The data was tested by SPSS 16.0 for Windows program. The results showed that dexamethasone given as corticosteroid for biliarry atresia theurapy couldsuppress TNF-αproduction as well as Gr-1 proliferation. In the other hand dexamethasonecan promote B220+ cell proliferation inrotavirus infected mice. Key word: Baby mice, biliary atresia, dexamethason, flowcytometry, rotavirus

Abstract

This study aimed to observe spatial variation of the Aek Sibundong red rice growth and productivity in three different areas of an organic rice field under different water logging levels, as well as to determine the interaction among rice growth with weeds and rice field’s abiotic factors on two planting seasons in the Sengguruh Village, Kepanjen District Malang Regency. Observations were conducted with a purposive sampling based on water logging in three areas. Red rice growths were observed in four growth phases: 20, 48, 82, 102 dap (days after planting) includes plant height, biomass, number of tillers and panicles, grain number and biomass. Weeds (taxa richness, % coverage and biomass) were observed using sampling plots of 0.25 m2. Abiotic factors include soil (pH, soil organic matter and soil bulk density) and rice field water (presence or absence of logging, pH, and conductivity). Data were analyzed using SPSS and PAST software. The result showed there were spatial variations of rice growth in three areas. Rice planted close by irrigation channel has taller plant height and more number of tillers and panicles but not significantly different from those planted in another two areas. However, rice planted close by irrigation channel tends to produces vegetative biomass and grain ten times more than those planted far by irrigation channel. Water logging decreased coverage and taxa richness of weeds. Some weeds after manual weeding had no effect on productivity of panicles and grains. High content of soils organic matter occurred at early vegetative phase and decreased at late reproductive phase, so the lower soils bulk density. Abstract

ABSTRAK Tujuan dari penelitian ini adalah untuk mengetahui pengaruh pemberian formalin dan benzo(α)pyrene terhadap struktur jaringan hati dan paru mencit (Mus musculus). Mencit jantan berumur dua bulan terpapar formalin dosis 2 mg/kg berat badan dengan perlakuan oral selama 60 hari dan terpapar benzo(α)pyrene dosis 250 mg/kg berat badan dengan perlakuan injeksi intraperitoneal sebanyak empat kali setelah 30 hari dengan selang waktu satu hari. Stuktur histologi hati dan paru mencit dianalisis secara deskriptif. Jumlah hepatosit yang mengalami nekrosis dianalisis dengan menggunakan uji Mann-Whitney pada software SPSS 16.0 for Windows. Hasil dari penelitian ini menunjukkan bahwa pemberian formalin, benzo(α)pyrene dan kombinasi antara formalin dan benzo(α)pyrene meningkatkan jumlah hepatosit yang mengalami nekrosis. Nekrosis terjadi di sekitar area portal triad dan di sekitar area vena sentralis pada hati mencit, sedangkan nekrosis terjadi di sekitar area vena pada paru mencit. Abstract

Ectoparasites were common found on fishes. Ectoparasites often infect parts of the fins, scales, operculum and gills in fish. This research aims to determine the types of ectoparasites on fish family Cyprinidae and the influence of environmental conditions on the number of ectoparasites. This research was conducted in three pools restaurant in Malang Raya region, then measured abiotic factors of pool water. Fish as the host of ectoparasites, taken using nets and anesthetized by using 3 phenol chloroform. Fish scales and fins were observed. Ectoparasites that attach to the parts taken and put in 70% alcohol. Ectoparasites observed and identified under a microscope. Measurement data of water quality and quantity ectoparasites were analyzed using SPSS 16 program. Argulus japonicus is a species of ectoparasite was founds in B Batu restaurant and I Dau restaurant. While ectoparasites found in G Malang restaurant was Lernaea cyprinacea. Argulus japonicus and Lernaea cyprinacea were exotic species from Japan. The entry of ectoparasites caused by introduction of Koi fish to Indonesia. The results showed that there were significant differences in the number of ectoparasites I Dau with B Batu and G Malang restaurants respectively. The correlation result between the number of ectoparasites and the physico-chemical factors were showed that the pool water DO levels were positively correlated to the number of ectoparasites.   Key word: Ectoparasites, fish family Cyprinidae, restaurant pond at Malang Area, SPSS 16

Abstract

Tujuan penelitian ini adalah mengetahui pengaruh penyapihan dini pada masa post partum terhadap siklus estrus induk kambing peranakan Boer F1 (Crossbreed Boer dan PE). Selama post partum, regulasi siklus estrus akan dihambat oleh prolaktin dan oksitosin yang dipengaruhi oleh mekanisme suckling. Penelitian ini menggunakan sampel sebanyak 9 ekor induk kambing dalam 3 kelompok yakni kelompok penyapihan selang waktu 42 hari post partum (PP), 56 hari post partum (PP), dan 91 hari post partum sebagai kontrol. Pengamatan siklus estrus dengan menggunakan vaginal smear. Data dianalisis menggunakan uji Kruskal Wallis dengan pendekatan kualitatif secara deskriptif. Hasil penelitian menunjukkan bahwa, interval siklus estrus kedua pasca sapih pada induk kambing kontrol  saat fase proestrus adalah 2 hari, fase estrus selama 2 hari, fase metestrus selama 2 - 3 hari, dan fase diestrus terjadi selama 14 - 15 hari. Selain itu siklus estrus pertama pasca sapih pada perlakuan penyapihan 42 hari (6 minggu), 56 hari (8 minggu), dan kontrol pada selang 91 hari (13 minggu) post partum adalah berkisar 16,7 ± 2,65 hari, 17,7 ± 1,53 hari , dan 10,3 ± 3,06 hari. Sedangkan siklus estrus kedua pasca sapih secara berturut – turut adalah berkisar 18 ±  1 hari, 19 ± 1,73 hari, dan 20,7 ± 1,58 hari. Hasil uji analisis menunjukkan bahwa tidak ada beda nyata (p>0,05) antar semua kelompok pada siklus estrus pertama pasca sapih. Kesimpulannya, interval siklus estrus kedua pasca sapih pada kelompok induk kambing sapih 42 hari PP adalah 18 hari dan sapih 56 hari PP adalah 19 hari dan kontrol rata – rata berkisar 20,7 hari. Abstract

ABSTRACT Biliary atresia is a neonatal obstructive cholangiopathy that progresses to end-stage liver disease. Dexamethasone is one of synthetic glucocorticoid which has function as an anti-inflammatory agent. Here, we investigated whether dexamethasone could modulate the immune activity in mice strain Balb/c with biliary atresia based on the change of quantity of IFN-γ and TNF-α as a pro-inflammatory molecules. This study consists of 2 stages. The first stage is pre-condition which is made the biliary duct become fibrosis by injecting 20 µl of phosphate buffered saline containing 1.5 x 106 fluorescence-forming units Rhesus Rotavirus (RRV) subcutaneously on first day (24 hours) after the mice was born. The second stage is injection with dexamethasone with dose 0,5 mg/kg BW subcutaneously on the 7th-14th day and 14th-21st day. The clinical effect of dexamethasone is investigated on 14th and 21st day by flow cytometry method. Data were analyzed using Kruskal-Wallis test (p<0,05) and Mann-Whitney test using SPSS 16 for Windows. Rotavirus injection subcutaneously was proven to stimulate the production of proinflammatory cytokines, especially in the third week of termination. The result indicated an increasing number of proinflammatory cytokines such as IFN-γ and TNF-α after RRV injection but after injection of dexamethasone the number of those cytokines is decreased. It can be understood that dexamethasone has a capability to reduce the effect of inflammation regard to the decrease of proinflammatory cytokines.
