Abstract

Plants of the genus Allium, such as shallots (Allium cepa var. aggregatum), garlic (Allium sativum L.), green onions (Allium fistulosum L.), and onions (Allium cepa var. cepa), are high-value commodities widely consumed in Indonesia. However, Allium production in East Kalimantan does not meet local demand, necessitating supplies from other regions. This inter-regional transport poses a risk of introducing organisms harmful to plants. Among these pests, viruses are particularly challenging to control. According to the Regulation of the Minister of Agriculture No. 25 of 2020, Onion Yellow Dwarf Virus is classified as a quarantine pest under category A2 requiring strict monitoring in Indonesia. Its distribution is currently limited to Java, Bali, West Nusa Tenggara, and Sulawesi. Therefore, accurate detection techniques are critical to prevent the spread of this pathogen to areas still free from Onion Yellow Dwarf Virus. Research results indicate that among the four types of onions, Onion Yellow Dwarf Virus (OYDV) exhibits symptoms in garlic. This finding was also confirmed through DAS-ELISA and RT-PCR testing. The detection results using DAS-ELISA on garlic plants showed an ELISA Reader absorbance value (0.6848) greater than twice the absorbance value of the negative control (0.2982). RT-PCR detection also yielded positive results with the formation of a 500 bp band.

Abstract

The existence of Hymenoptera parasitoids can be influenced by the landscape structure of agricultural land. Optimization of pest control using natural enemies can be done on a landscape scale. This study aimed to examine the characteristics of the rice cultivation landscape at each research location, examine the diversity of Hymenoptera parasitoids and their role in rice cultivation. In addition, this study aims to examine the factors that influence the diversity and abundance of Hymenoptera parasitoids, as well as examine the effect of landscape composition on the diversity and abundance of Hymenoptera parasitoids in rice cultivation in Malang Regency, East Java. The research was conducted at various designated rice planting locations. The research locations were in 4 sub-districts, namely Karangploso, Lawang, Kepanjen, and Tumpang sub-districts with three sampling points in each of these sub-districts. The research began with determining the location or ground survey. Insects were sampled using yellow pan traps,  with two traps installed in each subplot. Samples preserved in vials containing 70% alcohol were then sorted into microtubes according to sampling order, and specimens were examined morphologically to determine morphospecies. Supporting data were obtained from interviews with farmers or land owners. All data were entered into a database in Microsoft Excel and spreadsheets and analyzed using RStudio. The results indicated that the 12 study sites varied widely and were classified as complex landscapes, comprising eight land-use types dominated by natural habitats, paddy fields, and settlements. Hymenoptera parasitoids were found consisting of 8 families and 21 morphospecies. The parasitoids found act as parasitoids of eggs, larvae, eggs-larvae, and larvae-pupae. Altitude and age factors correlated with the diversity of Hymenoptera parasitoids. Landscape composition metrics, including Number of Patches (NP) and Class Area (CA) of rice fields, were correlated with the diversity and abundance of individual parasitoids.

Abstract

Weeds are plants that inhibit crop growth, development, and productivity. This leads farmers to use pesticides to control them. Herbicides, such as paraquat dichloride, are commonly used by farmers to control weeds. The U.S. Environmental Protection Agency (EPA) reported that 14 out of 41 commonly used pesticides in horticultural commodities are classified as carcinogenic compounds, and pesticide residues have been reported to contaminate 83% of the observed horticultural crops. Therefore, there is a need for technologies to address the issue of pesticide residues, such as bioremediation. The objective of this study was to determine the potential of bacteria in the cabbage rhizosphere, which is rich in diversity, as a bioremediator for maize fields contaminated with paraquat dichloride. The study was conducted at the Department of Plant Pest and Disease Laboratory, Brawijaya University, and farmers' cabbage fields from December 2019 to December 2020. The experimental design was a randomized block design (RBD) with 3 replications and 10 treatments, including a control group without bacteria or herbicide. The results showed that there were eight isolates showed bioremediation potential, and one isolate showed the best performance.

Abstract

Organic farming is a cultivation technique that applies natural materials and does not use chemical fertilizers and pesticides in cultivation techniques. Differences in cultivation techniques can affect the diversity, abundance, as well as the composition of spiders. This study was conducted to determine the diversity, abundance, as well as composition of spiders can be influenced by different cultivation techniques. The research was conducted from September 2023 to October 2023. Observations were made on organic land and conventional land. Data collection of cultivation techniques is carried out by interviews. Population and diversity data collection is carried out every 5 days within 4 weeks. Specimen collection is carried out by passive and active methods and then identification is carried out. Tabulate data in Microsoft Excel, then analyze it using Rstudio. In the Rstudio analysis, the Shapiro-Wilk test was carried out to determine the normality of the data, further Honest Significant Difference test at the level of 5%. Data analysis carried out in the form of variety analysis or and ANOSIM using the Bray-Curtis test. The results of the study, the number of spiders on organic land was more, namely 65 individuals consisting of 5 orders and 9 morphospecies. The results showed that differences in cultivation techniques had an effect on the diversity and abundance of spiders. The composition of the land shows that there is a significant difference but there are intersecting parts that indicate a similarity in the composition of spiders. The results of data analysis show that organic land has more diversity, abundance, and spider composition than conventional land.

Abstract

The study aimed to determine the effect of application Plant Growth-Promoting Rhizobacteria (PGPR). The research was carried out in the greenhouse dan laboratory of plant disease at the Brawijaya University Malang. The study was arranged in a Randomized Block Design which consisted of 8 treatments and 3 replications. The results showed that the single isolate treatment have the highest average incubation period of Helminthosporium turcicum, namely the single isolate of Bacillus subtilis and Pseudomonas fluorescens bacteria for 9 day. The treatment of the consortium of 3 bacteria, namely Bacillus subtilis+Pseudomonas fluorescens +UB19 (Pseudomonas sp). showed an excellent antagonistic activity in suppressing diseases caused by the fungus Helminthospororium turcicum. Pseudomonas sp. had the ability to increase the plants fresh weight but had no effect on the roots fresh weight.  However, the bacterial consortium (B. subtilis, P. fluorescens, and UB 19 (Pseudomonas sp.)) in sweet corn plant growth was indicated by the higher average yield of plant height, number and weight of roots compared to other treatments.
