Abstract

Antibiotic are secondary metabolites yielded by microbe especially fungus. Previous research succesfull screened four kinds of fungus that live in termites queen’s nest, one of them was Aspergillus flavus. Furthermore, Alen et al (2016g) states that this fungus last to produce metabolite compounds on SDA media which only exist in first and second  subculture extract, omit gradually for the next culture. It was presumed happen caused a different habitat to grow. So it is necessary to enrich the media with queen termite nest to get back the initial metabolites. The enrichment was done using four different media concentrations (0.25; 0,50; 0.75 and 1 grams of nest/mL media). The results show that enrichment of 1 gram of nest/mL media provides the most optimum fungus growht. The third subculture of Aspergillus flavus is cultured on enriched media which will become the fourth subculture, this fungus cultured until tenth subculture, do extraction and   fracination to each culture. Based on TLC profile analysis, the initial metabolite not yet formed until tenth subculture, but forms six new stain terpenoid compounds.  The result of  columns chromatography obtained 10 sub-faction. Activity test was done by diffusion method to 12  test bacteria and 3 test fungus. Spot 2,5,6 (Rf 0.84; 0,36; 0,26) inhibit the growth of Pseudomonas aeruginosa ATCC 27853, spot 3,4 (Rf 0.74; 0,52) inhibit the growth of Micrococcus luteus ATCC 10240. The use of enriched media affect formation of  secondary metabolites Aspergillus flavus.  Keyword : Secondary metabolites, Aspergillus flavus, Macrotermes gilvus Hagen., Enriched media, TLC Profiles, Activity Assay

Abstract

Vitamin E (Tocopherol acetate) is used in both oral and topical dosage.  The aim of  the research was to study  different concentration of Carrageenan, as gelling agent in Tocopherol acetate Emulgels.  This experimental study was initiated by emulgel formulation using different concentration of carrageenan as gelling agent, as much as 0,5% (F1), 0,75% (F2), 0,85% (F3), 0,95 % (F4), 1% (F5), 1,125% (F6), 1,25% (F7), 1,5% (F8) and 2% (F9).  Tween 20 and Span 20 were used as emulsifiers (1 and 1,5 %),  Liquid paraffin as oily phase (7,5%), Propylen glycol as humectant (10%), propyl and methyl paraben as antimicrobial preservative (0,01% and 0,03%). The physical investigation of emulgel were included pH, spreading test, viscosity and freeze thawtest. The results for promising formula F5 showed that physical parameters of emulgels were in the range of requirement for topical dosage form. Keywords: Tocopherol acetate (Vitamin E), emulgels, carrageenan

Abstract

Bawang Tiwai (Eleutherine americana (L.) Merr bulbus. is a typical plant from Kutai tribes which commonly used as an anti-bacterial and food coloring. The stability test is performed to determine the stability of the extract. Stability test was conducted in two solvents are distilled water and ethanol include the effects of temperature (25°C, 50°C, 80°C), pH (3, 5, 7), an oxidant for 6 hours, conditions of storage at room temperature and the temperature of the refrigerator for 24 hours and storage for 2 weeks. The absorbance of extract in ethanol and aquadest was decreasing due to the change of temperature, time, the presence of oxidator but due to pH changes, the absorbance of extract was increasing. Meanwhile, on the storage condition, showed the highest absorbance on dark, and room temperature storage compared to other condition.Key words: Eleutherine americana (L.) Abstract

An UPLC method has been developed for the simultaneous determination of ketorolac Tromethamine and tiemonium methylsulphate in pharmaceutical and biosamples. Calibration curves were linear with correlation coefficient 0.9997 over a studied concentration range of (01-10μg m ) for both drugs. Method was found to be reproducible with relative standard deviation (RSD) for intra to be 0.32-0.99% for KTR and 0.40-1.01% for TMS and inter day precision to be 0.51-2.77% for KTR and 0.41-2.26% for TMS over the said concentration range. The mean recovery of the drug obtained from the intraday assay was 99.18-103.34% for KTR, 99.14-101.76% for TMS and inter-day assay at six different days was 99.39-102.04% for KTR and 98.08-103.78% for TMS. The LOD and LOQ were found to be 0.125 and 0.41μg mL-1, respectively for KTR  and 0.150 and 0.50 μg mL-1, respectively for TMS. Method was successfully validated as per ICH guidelines can be conveniently employed for routine quality control analysis of ketorolac tromithamine and tiemonium methylsulphate in pharmaceutical formulations and biological samples without any interference.

Abstract

Anthocyanin is active compound of purple sweet potato, has effect as anti-oxidative, anti-inflammatory, and anti-itching, make them interesting ingredients for skin treatment. The body scrub is one of popular cosmetic product in SPA industry. Their stability property is one crucial factor to develop a purple sweet potato body scrub cream. The steam and oven heating of tuber were involved on the scrub powder preparation. The oven and non-heating preparation scrub powder provided fine-sized powder more than 50%, but the steaming produced granular particle distribution. Total anthocyanin content in scrub grains was 326.8 mg/100 g for steam preparation, 103.3 mg/ 100 g for oven, and 34.4 mg/100g for non-heating, respectively. The steamed scrub powder preparation method produced stabile anthocyanin content in body scrub cream for 25 days observation.
