Abstract

A research about the study on some botanical aspects of an endemic orchid Coelogyne celebensis J.J.Sm from Lore Lindu National Park has been conducted from March to November 2009. The research was located at the western part of Lore Lindu National Park, Herbarium Celebense (CEB) UNTAD, Orchid house of Bogor Botanic Garden, and the Herbarium Bogoriense (BO) Cibinong West Java. The survey method in the field was used to collect the living material of Coelogyne celebensis J.J.Sm, and then the description of the orchid was done at the Herbarium Celebense (CEB), Herbarium Bogoriense and Bogor Botanic Garden. The data of some botanical aspects included morphological characteristics, ecological (bitoik and abiotik) factor and social economic of the community who living around the habitat of the orchids were presented. Key words: Endemic orchid, botanical aspect, Coelogyne celebensis J.J.Sm, Lore Lindu NP.

Abstract

The structure of plankton community have been conducted in Togean Island from September 2008 untill Februari 2009, it used descriptive method. The result of the structure of zooplankton community indicate plankton range from 0,862-0.879, with value of biodiversity range from 0.328-0.330, for the value of variety of fitoplankton range from 0.328-0.329 and found marozoobenthos range from 26-27 individual/L, with variety value of benthos range from 0.831-0.835 with value of biodiversity range from 0.339-0.341. This indicator is not indicate the ecology pressure, but is not depend on river condition. Key words: Biodeversity, zooplankton, and Togean.

Abstract

The study is devoted to investigating the therapeutic effect on severity of acute respiratory infection (ARI) and nutritional status of underfive years old. It is analytic design of evaluative by visiting the house of sample at the fourth until the ninth days after giving therapeutic. The number of sample was 50 under five years old, selected by accidental sampling. The data were analyzed with wilcoxon sign runk test and paired t-test. The study indicated that first diagnoses of acute respiratory infection severity, it was found that average of duration of ARI was 3 days (17%), severe ARI is 84% and mild ARI 16%. Based on nutritional status (weight/age = W/A), severe malnutrition (8%), undernutrion (16%), and well nutrition (76%). The fourth days of therapy was found that duration of ARI was six days (66%) and severe ARI (52%), mild ARI (32%) and healing (16%). Meanwhile, nutritional status was severe malnutrition (6%), undernutrition (20%) and well nutrition (20%). The nine days of therapy, duration of ARI was six days (56%), severe ARI was 20%, mild ARI was 34% and healing was 46%. Severe malnutrition was 6%, undernutrition was 18%, and well nutrition was 76%. Paired T-test and Wilcoxon test, shown that there was therapeutic effect on duration and severity of ARI (P = 0,000) and (P = 0,004), and no effect of nutritional status based on paired t-test (P = 0,748). The conclusion is therapeutic can reduce of duration and severe of ARI, meanwhile nutritional status dose not have effect. Key words: Therapeutic effect, severity of ARI, nutritional status, underfive years old.

Abstract

Saccharomycopsis fibuligera is yeast with high amylolitic activity. The local strain, S. fibuligera R64 has been reported to produce starch hydrolyzing enzymes, -amylase dan glucoamylase. Glucoamylase is one of the extracellular enzymes that enable to breakdown starch in starch processing, thus has potential to be developed and applied in starch processing industry, such as glucose syrup production. Wild type microorganism-producing glucoamylase normaly express low level of enzyme, while high level production and low cost enzymes are needed in many industries. To obtain strains with superior properties, a recombinant DNA is used and Pichia pastoris is a suitable expression host for producing glucoamylase at high level. Gene encoding glucoamylase S. fibuligera R64 (GLL) has been amplified and cloned into pGEM-T vector. A set of primers (Glu-F and Glu-R) was designed based on the published nucleotide sequence (Acc. No M17355). Restriction analysis of pGEM-GLL by using EcoRI and XhoI showed a ~4.500 kb DNA fragment which confirmed the size of pGEM-T vector containing GLL. This research aimed to analyze pGEM-GLL S. fibuligera R64 in silico. Gene alignment of GLL S. fibuligera R64 with GLU1 S. fibuligera showed 99 % homology. Based on in silico translation analysis, there were several nucleotide difference compared to the published sequence which result in amino acid differences, namely E27D, N46D, dan A376V. Hence it is possible that S. fibuligera R64 has different properties from other known S. fibuligera. Key words: Glucoamylase, S. fibuligera, E. coli TOP10F’, PCR, in silico analysis.

Abstract

The study is aimed to (1) identify agroforesry pattern inside and outside of forest area based on structure and species composition; (2) know carbon stocks potential of agroforestry pattern base on vertical structure; (3) estimate aboveground carbon stocks of agroforesry patterns. The study was conducted on May to September 2009 in Tinggimoncong and Parigi District, Gowa Regency and Laboratory of Silvikultur, Hasanuddin University. To know tree biomass, measurement of diameter breast height was carried out. Total number of sample plots taken was 37 with 20 x 50 m in size. Selection and placement of sample plot was conducted by Purposive Sampling based on consideration of tree species dominant on each different agroforestry patterns. Withdrawal of tree’s biomass samples was conducted by non-destructive sampling, namely without destructing samples. The study results showed that (1) agroforestry system which generally used by community at the study location was agrisilviculture consisting of nine combination’s pattern of crop species. Interval adjustment follows random and border pattern with crop interval of tree component were irregular, (2) number carbon-stock on every range of vertical structure of agroforestry patterns were varied. Agroforestry Erythrina-jackfruit-coffee, clove-annual crops, african tree-coffee, Gmelina-banana patterns tend to follow bell-shaped curve or the stored carbon was the largest on medium-diamater trees, pine-vegetable, Eucalyptus-vegetable crops, pine-coffee, fruit trees-coffee tend to follow J-shaped curve or the stored carbon was the largest on wider-diameter trees, and agroforestry Erythrina-coffee patterns tend to be irregular; (3) two agroforestry patterns which had the highest potential carbon stocks aboveground were pine-coffee and fruittree-coffeeI agroforestry pattern amounted of 199.97 ton ha-1 and 100.54, respectively, and the lowest agroforestry pattern was clove-annual crop pattern namely 18.40 ton ha-1. Key words: Carbon stocks, agroforestry pattern, vertical stratification.

Abstract

Color blindness is a condition in which certain colors can not be distinguished, and most often caused by a hereditary disease. The purpose of this study to determine the prevalence of color blindness on prospective students who enrolled in the University Tadulako. Data collection was performed by means of color-blind investigation on every applicant that comes a medical examination at the Polyclinic Bungamputi Tadulako University. From the results, the prevalence of color blindness on prospective new students Tadulako University is 1.27% consisting of male and female 0.87% 0.40%. Key words: Color blindness, prevalensi.
