Abstract

This study was aimed at investigating the effect of NAA and BAP on the development of basal slip slices in tissue culture of pineapple cv. Tangkit. The experiment was conducted at the Plant Biotechnology Laboratory Agricultural Faculty University of Jambi from July through to October 2016. Six level of NAA concentrations (0, 1, 2, 3 , 4 and 5 mgL-1) were tested in combination with six levels of BAP concentration (0, 1, 2, 3 , 4 and 5 mgL-1). A Completely Randomized Design with 5 replicates was employed in this trial. Data on the percentage of explants forming shoots, time required for shoot formation, number of shoots growing per explants, and callus proliferation were recorded. Results showed that without the involvement of plant growth regulators there had been no growth on cultured explants. However, shoot growth was found on explants cultured on medium supplemented with 1 – 3 mgL-1 NAA or 1 – 5 mgL-1 BAP. The application of NAA higher than 3 mgL-1 was found to suppress shoot growth even in combination with BAP. Shoot formation was found to be fast on medium supplemented with 1 mgL-1 NAA + 4 mgL-1 BAP, but took longer time on medium with or 1 mgL-1 NAA + 5 mgL-1 BAP as well as medium with 1 – 2 mgL-1 NAA only. The same phenomenon was also found on medium with 1 – 2 mgL-1 NAA only. This study also revealed that most of cultured explants regenerated only one shoot, but explants cultured on medium with 1 mgL-1 NAA along with 1 mgL-1 BAP produced 2.33 shoots on the average. The addition of 1 mgL-1 NAA without BAP and 1 mgL-1 BAP without NAA could increase average shoot growth by 1.78 and 1.44, respectively. Abstract

Sunda Strait has high potential of pelagic fish resources, one of which is spotted sardinella (Amblygaster sirm). High market demand of spotted sardinella leads to its high exploitation. Management of sustainable used of this species need various biological information of the target species.The aims of this research wereto analyze of therecruitment pattern, mortality, and exploitation rate of spotted sardinella. Sampling was conducted in April to August 2015 from the catchment landed at Labuan Coastal Fishing Port, Banten. Fish samples taken during the study consisted of  527 males and 245 females. The results showed thatpeak recruitment season for spotted sardinella occurs in Maret (11.64%) and July (13.67%).The natural mortality (M), fishing mortality (F), total mortality (Z) for male were 0,33; 5,30; 5,63 year-1 respectively, and for female were 0,51; 7,75;8,27 year-1 respectively. Abstract

The research aims to study the effect population density of S. oryzae against population growth and damage of rice. The population density of S. oryzae is 5, 10, 15, and 20 pairs of adult per 250 g rice. The results showed that population density S. oryzae affects pest population growth of S. oryzae, characteristics loses weight of rice, percentage powder of rice, and changes content moisture of rice. The population density S. oryzae as many as 20 pairs of adult per 250 g of rice can increase populations pest S. oryzae, characteristics loses weight of rice, percentage powder of rice, and content moisture of rice. The relationship between population density S. oryzae with a growing population, percentage loses weight, percentage rice hollow, percentage powder of rice, and changes content moisture of rice increased linearly. There is a positive correlation between population density S. oryzae with population growth S. oryzae (r = 0.997**; P < 0.01), the percentage loses weight (r = 0.987*; P < 0.05), the percentage of rice perforated (r = 0.998**; P < 0.01), the percentage powder of rice (r = 0.997**; P < 0.01), and changes content moisture of rice (r = 0.964**; P < 0.01)Keywords: Sitophilus oryzae, population density, population growth, damage rice

Abstract

Agricultural land in Indonesia was largely dominated by untapped potential land, almost of that potential land is dry land. Maize (Zea mays L.) is one of the staple crops that are known could be cultivated on dry land. Rizosphere has been known to containmicroorganisms that can improve the plant growth. In the previously study, we have screenedin vitro of rhizobacteria (6 isolates of Pseudomonas sp CRB and 7 isolates of Bacillus sp CR) as growth promoter of maize and drought tolerant. A total of 6 rhizobacterial formula were further applied in vivo in greenhouse to promote the growth of maize. In planta test on maize showed that formula CRB 19 and F3 (CR 83 + CRB 10) were the best formula that could enhance plant growth parameters under drought stress conditions. After that abundance and activities of microbe were analyzed by enumeration of total fungus, bacteria and actinomycetes, dehidrogease test, soil respiration test dan C-microbe test and those tests showed that formula CRB 19 and F3 (CR 83 + CRB 10) were the best formula.These formula could be recommended as inoculants of maize planted in dry land agriculture. Abstract

Aeromonas salmonicida are specific bacteria that can cause infections and death to the cultivation of carp (Cyprinus carpio) during larval stage. Death in carp can be prevented by a vaccine, but the vaccine can only be given on the seed over the age of 3 weeks. Maternal vaccination needs to be done to improve the immune system of the larvae by means of inactivated whole cell vaccine A. salmonicida on broodstock ready to spawn. Aims to determine the effectiveness of vaccines on breeders carp to the parent antibody titer test and larvae, as well Survival Rate (SR) and the Relative Percent Survival (RPS) larvae. This research was conducted with a completely randomized design, 4 treatments A (control); B (0.3 ml/kg); C (0.4 ml/kg); D (0.5 ml/kg) and 3 repetitions. The results show that the antibody titer of 0.3 ml dose capable of providing agglutination reaction to pitting 7th (64x dilution) in broodstock, and vaccine doses 0,4ml on broodstock able to give agglutination reaction to the larvae until all 6 wells (32x dilution). A dose of 0.4 ml/kg resulted the highest SR and RPS with 96.11% and 81.25% respectively. Clinical symptoms of redness in control larvae was spread throughout the body whereas on the vaccine treatment was only in certain body parts.Keywords: A. salmonicida, vaccines, maternal immunity, larva, specific immune respon
