Abstract

Until now Malaria remains a major health problem in Indonesia. Purworejo one of the regencies in Central Java, is a malaria endemic area and the prevalence remains the same even though some efforts to eliminate the disease have been done. This study was aimed to investigate the susceptibility of P. falciparum to chloroquine, extended in vivo test was used. Subject were enrolled using mass blood survey and mass fever survey. This study show that 45,6% P. falciparum was resistant to chloroquine, consisted of RI early recrudescence (16.22%), RI delayed recrudescence (8.11%), RII (16.22%), RIII (5.41%).Key word: invivo test, P. falciparum, chloroquine

Abstract

Finding anticancer drugs from natural resources still proceeds. Oleandrin isolated from Nerium indicum Mill. inhibited the growth of mieloma cell line in vitro better than that of vincristine sulphate. This study was aimed to determine the cytotoxic effect of oleandrin on various human cancer cell lines. Cytotoxic test of oleandrin on seven human cancer cell lines was done by SRB-method. The analysis was conducted by comparing the ID50 of oleandrin with that of doxorubicin and cisplatin as positive controls. This result indicated that oleandrin possessed the best cytotoxic effect on breast cancer (MCF7) with ID50 at 8.85 nM. Keywords : Oleandrin, cytotoxicity, human cancer cells, ID50

Abstract

Iridoid substances can be used as characteristic substances in Rubiaceae. Asperuloside and scandosidmethylesther, iridoid substances were found in Hedyotis corymbosa (L.) Lamk. (Oldenlandia corymbosa Linn). The people used several Rubiaceae plants for curing cough, rheumatic, cancer, fieber. Distribution of asperuloside and scandosidmethylesther inleaf, stem, fruit and root of Hedyotis corymbosa (L.) Lamk. has a possibility as a characteristic pattern. The aim of this research is to know the distribution profile of asperuloside, scandosidmethylesther, at the leaf, stem, fruit, and root of Hedyotis corymbosa L. (Lamk.). Trim Hill reagent was used for the qualitatve analyse of asperuloside and scandosidmethylesther and gradient system elution by high pressure chromatography was used for separation and quantitative analyses.Asperuloside content were in stem (0,146 ± 0,004 %); leaf(0,470 ± 0,020%); fruit (0,142 ± 0,005%); root(0,560 ±0,013%) and scandosidmethylester content were in stem (0,618 ± 0,22); leaf (0,156 ± 0,007); fruit(0,037 ± 0,002); root (0,136 ± 0,006).Key words: Asperuloside, scandosidmethylesther, distribution Hedyotis corymbosa (L.)Lamk.(Oldenlandia corymbosa Linn)

Abstract

Isolation of the biotransformation product artemisinine from endophitic bacteria is a new alternative for the production of the multidrugresistant antimalaria compounds. The bacteria strain, AT 12, has been proved particularly potential as an artemisinin source.This working was carried out to isolate and to identify the biotransformation product artemisinin as an antimalarial active compound from endophitic bacteria strains of Artemisia annua. The IR spectrum exhibited the characteristic bands for δ-lactone (1750-1735 cm-1, 1250-1111 cm-1) and endoperoxide function (~1115 cm-1, 890-830 cm-1). As a result, the GC-MS gave a positively identification for the majority of GC peaks. The mass spectrum of the peak (Rt 27.94 min) has a molecular ion of artemisinin with one hydrogen loss at m/z 281, representing (M+ - 1).Keywords: Artemisia spp. Artemisia annua, Endophytic, Artemisinine, sesquiterpene

Abstract

The study of the GMP implementation for the traditional medicine industries in Central Java has been performed. The GMP for traditional medicine industry is the guidance how to produce and to manage the quality of the product so that it meets to the quality of design and the quality of conformance. The aim of the study is to evaluate how far the GMP is implemented in the traditional medicine industries in Central Java.The method used of the study was a descriptive evaluative qualitative data collected from several traditional industries namely: M, N, O, P, Q, R and S by assessing the implementation level of the GMP in those industries. The evaluation instrument used was the primary data directly collected by interviewing the directors, the managers and the heads of department, and then followed by visitation to the industries to check the primary data and the actual condition.  The result showed that the evaluation of the GMP implementation for the traditional medicine industries in Central Java reached the score of 86.10% that meant of good category, although the quality control, the selfevaluation, and the documentation aspect had to be ameliorated.Key word : GMP, traditional medicine industries

Abstract

A research of cytotoxicity and antimicrobial test of bioactive compound of Stylissa flabelliformis sponge have been done. Cytotoxicity test were conducted on myeloma cells, while antimicrobial test were conducted against Staphylococus aureus,Eschericia coli and Candida albican. The research was initiated with the isolation of bioactive compound from the active fraction of sponge S. Stylissa monitored by Brine shrimp Lethality tes (BST). The active fraction was chromatograped with silica gel as its immobile phase and the mixture of hexane and ethyl acetate (1:1 ) as the mobile phase to obtain compound 1,2,3 and 4.Compound 3 has the most toxic character because it could kill A. salina equal to 100% at dose 25 mg/ml, and has LC50 of 0,9 mg/ml. Cytotoxicity test with direct staining method using tripan blue on myeloma cell at density of 4,5x10 cell / 100.ul showed that the compound 3 had a high activity against myeloma cell, having LC50 equal to 0,08 mg/ml.The result of antimicrobial effect showed that compound 3 was a fungicide against Candida albicans but having no activity on Staphylococcus aureus and Eschericia coliKey words: sponge, Stylissa flabelliformis , cytotoxicity, fungicide

Abstract

Curcumin, a plant phenol in rhizome Curcuma longa L. has been reported to be a potent inhibitor of glutathione S-transferase (GST) in rat liver with in vitro with substrate 1,2-dikloro-4-nitrobenzen (DCNB). By using DCNB, a specific substrate for GST mu, This work was aimed to know whether curcumin derivatives: 4-n-propyl curcumin can also inhibit GST activity in vitro.Glutathione activity can be measured by conjugating GSH and DCNB catalized with GST. The product can be measured spectrofotometrically. Curcumin and its derivatives decreased GST activities with IC50 values 6,86 μM for curcumin and 8,41 μM for 4-n-propyl curcumin. This means that 4-n-propyl curcumin has a weaker inhibition potency than curcumin.Curcumin (consentration 10μM) inhibited GST activity by mixed with the Ki dan Ki’ values 2,77 μM dan 7,33 μM, whereas 4-n-propyl curcumin (consentration 10μM) inhibited GST activity by competitive with the Ki values 6,28 μM.Key words : Curcumin, 4-n-propyl curcumin, glutathione S-transferase, 1,2-dikloro-4-nitrobenzen.

Abstract

Piper sarmentosum Roxb. ex Hunter (“sirih duduk”) has long been used as a traditional medicine against fungoid diseases, especially at vagina and foot-sole. The investigation of the bioactive compounds of P. sarmentosum roots has not infections been carried yet. This research was then aimed to fractinate and to examine the antifungus activity of the extract of P. sarmentosum roots.The extraction was performed by using organic solvents such as methanol, ethyl acetate, chloroform, benzene, and n-hexane. The bioactivity tests were performed against C. albicans. The active extracts were then fractionated by coloumn chromatography.The result showed that the ethyl acetate fraction of methanol extract of P. sarmentosum roots have an antifungus activity against C. albicans. Phytochemical study showed that the active fractions of the methanol extracts of P. sarmentosum roots contained alkaloid.Key words : Piper sarmentosum, activity test, Candida albicans.
