Abstract

Fermentasi mikroba endofit dari tannaman kina (Cinchona pubescens Vahl.) dalam medium Phoma telah dilakukan. Hasil fermentasi selama 8 hari diekstraksi dengan diklormetan diperoleh asam lemak yaitu asam oleat dalam bentuk cis. Hasil ini dipertegas oleh spectra RMI (proton dan karbon) dan spektramassa (GC-MS).

Abstract

Isolation of the biotransformation product artemisinine from endophitic bacteria is a new alternative for the production of the multidrugresistant antimalaria compounds. The bacteria strain, AT 12, has been proved particularly potential as an artemisinin source.This working was carried out to isolate and to identify the biotransformation product artemisinin as an antimalarial active compound from endophitic bacteria strains of Artemisia annua. The IR spectrum exhibited the characteristic bands for δ-lactone (1750-1735 cm-1, 1250-1111 cm-1) and endoperoxide function (~1115 cm-1, 890-830 cm-1). As a result, the GC-MS gave a positively identification for the majority of GC peaks. The mass spectrum of the peak (Rt 27.94 min) has a molecular ion of artemisinin with one hydrogen loss at m/z 281, representing (M+ - 1).Keywords: Artemisia spp. Artemisia annua, Endophytic, Artemisinine, sesquiterpene

Abstract

Diabetes mellitus is a degenerative disease characterized by hyperglycemia due to insulin insulin deficiency either absoluteor relative. This study was conducted to isolate endophytic fungi from plant twigs mahogany (Swietenia macrophylla King) which active as antidiabetic and antioxidant. Antidiabetic activity conducted by using the α-glucosidase inhibitory and antioxidant activity using free radical reduction method with reagent 2.2-diphenyl-1-picrylhydrazyl (DPPH). Isolation of microbes conducted in the media Corn Meal Malt Agar (CMMA) and Potato Dextrose Agar (PDA) which 7 isolates of fungus in total. Inhibitory activity against α-glucosidase to extract the filtrate and biomass of the isolates A.Sm.2F (72.59 and 92.22%), A.Sm.3F (81.87 and 79.37%), B.Sm.1F (63.40 and 98.84%), B.Sm.2F (65.60 and 62.72%), B.Sm.3F (93.91 and 51.48%), B.Sm.4F (87.48 and 74.64%) thus has potential as an antidiabetic activity. B.Sm.1F was the only isolates active as antioxidants with IC50 of 84.41.

Abstract

Endophyte fungi are microbe that living inside the plant tissue without harming the host plant. Endophyte fungi can produce secondary metabolite which can be used as antioxidant, anticancer and antimicobes compound. Endophyte fungi can be found in many plants especially herbs such as turmeric (Curcuma longa L). The aims of this study are to isolate and identify endophyte fungi from stem of C. longa L. which is potential as an antioxidant producer. The endophyte fungi isolated from turmeric stem were 12 isolates. Antioxidant activity was assayed using 1,1-Diphenyl-2-picryl-hydrazyl (DPPH) showed that isolate K.Cl.Sb.B1 produced the highest inhibition value (78,81%). Based on molecular identification, the isolate K.Cl.Sb.B1 was Colletotrichum sp.Keywords: Curcuma longa L., endophyte fungi, identification antioxidant ABSTRAKKapang endofit merupakan mikroba yang terdapat di dalam jaringan tanaman tanpa membahayakan tanaman inang. Kapang endofit mampu menghasilkan metabolit sekunder yang dapat dimanfaatkan sebagai senyawa antioksidan, antikanker dan antimikroba. Kapang endofit dapat ditemukan pada berbagai jenis tanaman terutama tanaman obat seperti kunyit (Curcuma longa L). Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi kapang endofit dari batang tanaman kunyit yang berpotensi sebagai penghasil antioksidan. Kapang endofit yang diisolasi dari batang tanaman kunyit diperoleh 12 isolat. Uji antioksidan menggunakan 1,1-Diphenyl-2-picryl-hydrazyl (DPPH) menunjukkan bahwa isolat K.Cl.Sb.B1 menghasilkan nilai inhibisi tertinggi (78,81%). Berdasarkan identifikasi molekuler, isolat K.Cl.Sb.B1 merupakan Colletotrichum sp.Kata kunci: Curcuma longa L., identifikasi antioksidan, kapang endofit

Abstract

Cassava is a staple food after rice and maize in Indonesia. In general, parts of cassava plants that consumed are the leaves and tubers. This research aimed to compare antioxidant activity of leaves and tuber from six types of cassava (Manihot utilissima Pohl). The leaves and tubers of the cassava plant from Cibinong were extracted with methanol separately. Methanol extract was measured the inhibition percentage as an antioxidant using DPPH reagent at 100 ppm. Results showed that Pucuk Biru leaves type has the highest inhibition at 88.09% and IC50 of 45.161 ppm.Keywords: antioxidant, Cibinong, DPPH, IC50, Manihot utilissima Pohl ABSTRAKKetela pohon merupakan bahan pangan pokok setelah beras dan jagung di Indonesia. Bagian tanaman singkong yang umumnya dikonsumsi adalah daun dan umbi. Penelitian ini bertujuan untuk membandingkan aktivitas antioksidan daun dan umbi dari enam jenis singkong (Manihot utilissima Pohl). Daun dan umbi tanaman singkong dari Cibinong masing-masing diekstraksi secara terpisah menggunakan pelarut metanol. Ekstrak metanol yang diperoleh diukur persentase daya inhibisinya menggunakan metode peredaman radikal bebas dengan reagen DPPH pada konsentrasi 100 bpj. Hasil pengujian antioksidan menunjukkan bahwa bagian daun singkong jenis Pucuk Biru mempunyai daya inhibisi yang paling tinggi yaitu 88,09% dan IC50 sebesar 45,16 bpj. Abstract

Microorganisms activity and oxidation process can contribute to food spoilage occurrence. Natural source that have antimicrobial activity and antioxidant are potential as a natural preservative. The aims of this study are to investigate the endophytic fungi from turmeric plant which have antibacterial toward histamine-producing bacteria and antioxidant activity. Antibacterial activity of histamine-producing bacteria was done by paper disc diffusion, whereas the antioxidant activity was performed by free radical scavenging method. In the paper disc diffusion method were obtained 3 isolates which have the highest diameter of inhibition at concentration of 20.000 ppm: from root (15 mm), flower (14 mm) and  rhizome (13 mm). Also at the antioxidant assay, the lowest IC50 values were isolates from root (39,52 ppm), flower (157,93 ppm) and rhizome (96 ppm). Endophytic fungi from root, flower and rhizome had antimicrobial and antioxidant activity thus they could potentially used as a natural food preservative.Keywords: antibacterial, antioxidant, endophyte, preservative,  turmericABSTRAKAktivitas mikroorganisme dan proses oksidasi dapat menyebabkan kerusakan pangan. Bahan alam yang mempunyai aktivitas sebagai antimikroba dan antioksidan berpotensi sebagai bahan pengawet alami. Penelitian ini bertujuan untuk mengetahui kapang endofit asal tanaman kunyit yang memiliki aktivitas antibakteri penghasil histamin dan antioksidan. Aktivitas antibakteri penghasil histamin dilakukan dengan menggunakan metode difusi cakram kertas, sedangkan aktivitas antioksidan dilakukan dengan metode peredaman radikal bebas. Pada metode difusi cakram kertas didapatkan 3 isolat yang memiliki diameter daya hambat tertinggi sampai konsentrasi 20.000 ppm yaitu berasal dari akar (15 mm), bunga (14 mm) dan rimpang (13 mm). Pada uji antioksidan didapatkan nilai IC50 terendah yaitu isolat yang berasal dari akar (39,52 ppm), bunga (157,93 ppm) dan rimpang (96 ppm). Isolat kapang endofit yang berasal dari akar, bunga dan rimpang memiliki aktivitas antimikroba dan antioksidan sehingga berpotensi digunakan sebagai bahan pengawet makanan alami.Kata kunci: antibakteri, antioksidan, endofit, kunyit, pengawet

Abstract

Tanaman ranti hitam (Solanum blumei Nees ex Blumei), famili Solanaceae, ditemukan di daerah Dairi dan Karo, secara tradisional digunakan sebagai tanaman obat. Pengujian aktivitas antikanker dari ekstrak etanol buah ranti hitam terhadap sel leukemia L1210 telah dilakukan dengan metode hemositometri setelah diinkubasi 48 jam pada suhu 37°C di dalam inkubator 5% CO2. Hasil penelitian menunjukkan ekstrak etanol buah ranti hitam berpotensi sebagai antikanker terhadap sel leukemia L1210 dengan nilai IC50 sebesar 14,88 µg/mL (aktif).

Abstract

Sambiloto (Andrographis paniculata) has been used to treat malaria. In vitro research using parasite or through heme polymerization inhibition using andrografolide and in vivo using infected animal test have been done widely, however, heme polymerization inhibition from extracts with different polarity levels has not been studied yet. The aims of this study were to investigate the heme polymerization inhibition activity and toxicity of sambiloto leaf extracts. Sambiloto leaf extracted with gradually maceration using n-hexane, ethyl acetate, and 70% ethanol respectively. Heme polymerization inhibition activity was used as in vitro antimalarial test. Brine shrimp lethality test (BSLT) was used to determine toxicity of the extracts. Phytochemically screening was done for all extracts qualitatively. The results of this study were n-hexane, ethyl acetate, and 70% ethanol extracts had heme polymerization inhibition activities with IC50 values at 2,196.57; 1,235.54; and 1,157.24 μg/mL respectively. N-hexane, ethyl acetate, and 70% ethanol have LC50 values at 1,155.79; 1,133.89; and 5,229.15 μg/mL respectively. 70% ethanol extract contains alkaloid, flavonoid, steroid/triterpenoid, saponin, and tannin. N-hexane and ethyl acetate extracts has only contains alkaloid and flavonoid. 70% ethanol, ethyl acetate, and n-hexane extracts of sambiloto leaf have ability to inhibit heme polymerization and also non toxic to Artemia salina larvae.   Abstrak Sambiloto (Andrographis paniculata) telah lama digunakan untuk mengobati malaria. Penelitian in vitro dengan parasit maupun melalui penghambatan polimerisasi hem dengan senyawa andrografolida dan in vivo dengan hewan uji terinfeksi telah dilakukan, namun demikian belum ada penelitian tentang penghambatan polimerisasi hem ekstrak dengan tingkat kepolaran yang berbeda. Penelitian ini bertujuan untuk mengetahui aktivitas penghambatan polimerisasi hem dan toksisitas ekstrak daun sambiloto. Daun sambiloto diekstraksi secara maserasi bertingkat berturut-turut menggunakan pelarut n-heksan, etil asetat, dan etanol 70%. Uji antimalaria secara in vitro menggunakan metode penghambatan polimerisasi hem. Uji toksisitas menggunakan metode uji kematian larva Artemia salina (BSLT). Skrining fitokimia dilakukan secara kualitatif untuk seluruh ekstrak. Hasil penelitian menunjukkan bahwa ekstrak n-heksan, etil asetat, dan etanol 70% memiliki aktivitas penghambatan polimerisasi hem dengan nilai IC50 masing-masing sebesar 2.196,57; 1.235,54; dan 1.157,24 μg/mL. Ekstrak n-heksan, etil asetat, dan etanol 70% masing-masing memiliki nilai LC50 sebesar 1.155,79; 1.133,89; dan 5.229,15 μg/mL. Ekstrak etanol 70% mengandung alkaloid, flavonoid, steroid/triterpenoid, saponin, dan tannin. Ekstrak n-heksan dan etil asetat hanya mengandung alkaloid dan steroid/triterpenoid. Ekstrak etanol 70%, etil asetat, dan n-heksan daun sambiloto memiliki kemampuan dalam menghambat polimerisasi hem dan tidak toksik terhadap larva Artemia salina.

Abstract

One sesquiterpene compound has been isolated and identified out of the ethylacetate extract of bawang hutan (Scorodocarpus borneensis Becc.) barks. The barks were macerated with methanol, and then partitioned with mixture of ethylacetate-water (1:1).  Fractionation of the ethylacetate phase by column chromatography gave pure compound. Based on data interpretation from of ultra-violet (UV) spectra, Fourier Transform Infra Red (FT-IR), NMR 1D (1H and13C-NMR); NMR 2D (HMQC, COSY, HMBC), and comparison with literature, the pure isolated compound was determined as sesquiterpene   compound, cadalene-β-carboxylic acid, which exhibited LC50 of 42.32 ppm. Keywords: bawang hutan, Scorodocarpus borneensis, Olaceaeae, brine shrimp lethality test.

Abstract

Malaria is still the leading cause of death worldwide with nearly half the worlds population at risk. Parasitic resistance to existing antimalarial drugs in the market makes the search for a source of new drugs from nature is very important. Therefore, the aims of this study are to determine in vitro antimalarial activity of endophytic fungi extract from turmeric root and to identify the selected isolate molecularly. Heme polymerization inhibition method was used as in vitro antimalarial assay. The selected isolate was thrn identified using ITS1, ITS2, and 5.8S sequences of rDNA. The result of this study obtained 16 isolates of endophytic fungi from root of turmeric plant with isolate code were of K.Cl.Sb.A1 - K.Cl.Sb.A16. All of the ethyl acetate extracts of isolated endophytic fungi have heme polymerization inhibition activity. K.Cl.Sb.A11 was the most active isolate on heme polymerization inhibition test with 94,31% at concentration of test material at 8 mg/mL and IC50 value at 1.84 mg/mL. Molecular analysis showed that K.Cl.Sb.A11 isolate was Penicillium sp. and potentially developed as an antimalarial drug.
