Abstract

Study on biological reproduction aspects of Asian Arwanas fishes {Scleropages jardinii and S. formosus) particularly their sexual dimorphism and sexual ratio of gender is very important as a base for captivity breeding. Up to the present, it is difficult to determine the sexual identity of these Asian Arwanas, lead the difficulties in making sexual ratio description.The reliable sexual ratio in captive breeding would increase the production. The result of this research would be useful for reference in the identification of the gender of Asian Arwanas.

Abstract

Water is the main parameter to determine whether the yield potential of a plant is obtained or not. Water deficit on the tissue causes the disruption of all chemical process in the plant metabolism resulted in the plant growth impediment.In order to acquire the drought tolerance variety, various efforts have been conducted. Among others, in addition to the selection and characterization of the available germ plasma along with its cross-breeding, the development of the drought tolerance plant is conducted through somaclonal varitype induction. In the cultivation of the drought tolerance plant, beside physiological mechanism and biochemistry related to the plant tolerant to drought, the procedure of the selective and optimal selection should be managed.Hence,the mastery of the optimum selection techniques, in a relatively short time, the new drought tolerance lines could be obtained.

Abstract

Saccharomycopsis sp. produced glucoamylase (a-1,4 glucanglucohydrolase; EC 3.2.1.3) shown by the presence of clear zone on media containing 0.2% yeast extract, 0.5% pepton, 0.3% KH2PO4, 0.05% MgSO4. 7H2O, 0.01% CaClr2H2O, 2% agar and 2% soluble starch. In liquid media containing soluble starch glucoamylase production reached a maximum at 2 days fermentation, with levels of 4.2 x 10 U/ml (one unit activity is define as micromoles of glucose produce per ml per minute).Studies on the glucoamylase characterization revealed that the optimum temperature for activity was 40°C - 50°C. The enzyme was stable for lh at 45"C - 55°C, while at 60°C to 65°C, 40% of the original activities were lost. The optimum pH of the enzyme was 6.0. After incubation of crude enzyme solution for 1 h at pH 8.0, a decrease of about 40% of its original activity was observed. Saccharomycopsis sp. produced high glucoamylase when it was grown in media containing rice flour as carbon source with the glucoamylase activity at 9,28 x 10 U/ml.

Abstract

Adaptive responses of an herbivorous ladybird beetle Henosepilachna vigintioctopunctata (Epilachninae) and the host plants to dry season were studied in Pasuruan and Malang, East Java. Nineteen species of host plants of the beetle have been recorded in Java, however only fiveteen species are found as preferred host plants of the beetles in Pasuruan and Malang. During the season beetles tend to prefer the plants of Datura metel, Brugmansia spp., Solanum melongena, S. torvum, S. pseudocapsicum, and 5. nigrum as their hosts and this might course strong damage to the plants.It is obvious that the beetles tend to move to the humid places to find more suitable food. It is also found that during the days of drought, the beetles feed on eggplant fruits by tunnelling into the flash of the fruits.Some of the beetles ovaries would not develop in the drought and a part of them might enter the diapause stage.Other responses of the beetles to drought will be discussed in the paper.

Abstract

An experiment was carried out to find out the optimum nitrogen and phosphorous concentration for growth and phycocyanin production in Spirulina fusiformis culture. The cultures were grown in Zarouk medium at various nitrogen and phosphorous concentrations, which were 0.0 mM N, 7.5 mM N, 15.0 mM N, 22.5 mM N, and 30.0 mM N, as well as 0 raM P, 90 mM P, 180 mM P, 270 mM P, and 360 mM P, with four replications each. The result shows that optimal growth of the alga, which is expressed in terms of the biomass yield, was achieved at nitrogen and phosphorous concentration of 7.5 mM and 270 mM, respectively.At the same time, the highest phycocyanin content was obtained at nitrogen concentration of 22.5 mM, which was 1,2% of the biomass, and phosphorous concentration of 360 mM, which was 1.1% of the biomass. According to this result, it is suggested the optimum concentration of nitrogen and phosphorous in the media of Spirulina culture for phycocyanin production is 22.5 mM and 360 mM, respectively.

Abstract

The cucumber (Cucumis sativus L.) sex epression commonly monoceous and diaceous, however are dominated by male flowers.The percentage of female flower are only under 5% of total flowers per plant. Due to the environmental effects, the female flowers are usually fall down, thus fruitset decreased. The objective of this research is to increase the potential of female flowers to set fruits, thus impacted on fruit yield increase of cucumber by application of methyl blue and soaking seeds. The study was conducted at Kalijati Sub-District (+ 96 m asl), Subang District, West Java Province between May to June 2006 (dry season), in latosol reddish soil. The experiment was designed under a randomized block in factorial model, with twelve combination treatmeants and three replications.The the treatmeants were: A. concentration methyl blue: (Al) control (0 %), (A2) 0,03 %, (A3) 0,06 % and (A4) 0,09 %. B. the length of soaking: (Bl) 12 hours, (B2) 24 hours and (B3) 36 hours. The results shows that the stimulation of cucumber seeds in 0,03%, 0,06% and 0,09% compared to control, increasing the female flowers ability to set fruits up to 62,65%,45,61% and 13,79% respectively; and had impacted to increase yield per plant up to 40,69%, 34,48% and 14,09%. The stimulation of cucumber seeds by 0.03, 0.06 and 0.09% of methyl blue were able to increase the potential of female flowers to set fruits 62,65,45,61 and 13,79 % respectively. Meanwhile it has impacted in increasing fruits yield per plant up to 40,69%, 34,48% and 14,09%. The effect of 0,06% and 0,09% of methyl blue concentration, showed on the decrease of survival plants (28,42% and 37,73 %),seed germination (3 and 5 days) and the initiation of flowering time (1 and 2 days). The longer the seeds are soaked in methyl blue solution, there were tendencies in decreasing the potential of female flowers to set fruits thus fruits yields; also the percentage of plants to develop, longer germination period and decreasing the possibility of female flowers to set fruit.

Abstract

Talas {Colocasia esculenta (L.) Schott} and Iles-iles (Amorphophalus campanulatus Blumei) have been used as source of carbohydrate especially in tropical regions, mainly in East Indonesia. Talas and iles-iles, that contains carbohydrate and several substances for instance glucomannan would form viscous liquid and were used as emulgator, capsule cosmetics and dietary.The research was conducted to separate glucomannan from starch of talas and iles-iles fresh and dry (powder).The methods base on speed of revolution (centrifuge) in 5000 to 19000 rpm and the duration of rotation was 10 to 40 minutes and the filtrate was precipitated by using several alcohols (methanol, ethanol, propanol, isopropanol and isobutanol). The results showed that speed of rotation 17 x 1000 rpm with 10 minutes rotation resulted in good glucomannan separation from starch.Ethanol and isopropanol were the best solvent to separate glucomanan from dissolved starch in the filtrate with ratio 2 : 3 and for dry material, and the maximal agitation reached after 2 hours. Glucomannan content on fresh material was higher than as in talas (4.08 %) and iles-iles (5.64 %), compared to these in powder (dried) of talas (3.87 %) and iles-iles (5.41 %). The purerity of isolated glucomannan was higher compared to control.

Abstract

Six kinds of the endophytic fungi have been isolated from a tea plant. One of them identified as Diaporthe sp. E based on their morphological characters and rDNA sequens. By the cultivation in liquid medium, the endophytic fungus produced (+)-epiepoxydon besides gentisyl alcohol and toluquinol.
