Abstract

Dental caries remains a major oral health problem with high prevalence and is primarily caused by the activity of Streptococcus mutans. One of the key virulence factors of this bacterium is Sortase A protein, which plays an essential role in bacterial adhesion and biofilm formation. This study aimed to evaluate the potential of bioactive compounds from Cymbopogon citratus as inhibitors of Sortase A from Streptococcus mutans using an in silico approach. The research method involved preparation of the Sortase A protein structure (PDB ID: 4TQX) and bioactive compounds of C. citratus, followed by molecular docking analysis using AutoDock Vina integrated with UCSF Chimera. Ligand–protein interaction analysis was performed to determine binding affinity and identify key amino acid residues involved, and the results were compared with a standard ligand (1QON). In addition, ADMET prediction was conducted to evaluate the absorption, distribution, metabolism, excretion, and toxicity profiles of the major compounds. The molecular docking results showed that luteolin and rutin exhibited strong binding affinities toward Sortase A, with binding energy values of -7.3 kcal/mol, comparable to the standard ligand (-7.2 kcal/mol). Luteolin demonstrated a similar interaction pattern with the standard ligand through the key residue Phe237, while rutin formed multiple hydrogen bonds around the active site. ADMET prediction indicated that luteolin possessed better oral absorption and a more favorable safety profile than rutin. In conclusion, luteolin is the most promising bioactive compound from Cymbopogon citratus for dental caries prevention through inhibition of Sortase A of Streptococcus mutans

Abstract

Swamedikasi atau pengobatan mandiri sering kali menjadi langkah awal yang diambil masyarakat untuk mengatasi keluhan kesehatan ringan sebelum mendatangi fasilitas kesehatan. Namun, praktik ini kerap diiringi oleh rendahnya tingkat literasi terkait pemilihan obat yang tepat, ketepatan dosis, pemahaman efek samping, hingga cara pembuangan obat yang benar. Kondisi tersebut juga ditemukan di daerah Waibron, di mana akses terhadap informasi kefarmasian yang komprehensif masih terbatas. Kegiatan pengabdian kepada masyarakat ini bertujuan untuk meningkatkan kesadaran, pengetahuan, dan keterampilan masyarakat Waibron dalam melakukan swamedikasi secara aman dan rasional melalui pendekatan edukasi langsung. Metode pelaksanaan meliputi sosialisasi interaktif, penyuluhan menggunakan media visual (poster dan leaflet), serta diskusi tanya jawab interaktif serta pemahaman logo golongan obat. Hasil kegiatan menunjukkan adanya peningkatan pemahaman peserta yang diukur melalui pre-test dan post-test, di mana masyarakat menjadi lebih teliti dalam membaca informasi pada kemasan obat, mengenali tanda peringatan pada obat bebas dan obat bebas terbatas, serta menyadari pentingnya menghindari penggunaan obat keras tanpa resep dokter. Program ini terbukti efektif dalam memberdayakan masyarakat Waibron agar lebih mandiri dan bijak dalam menjaga kesehatan keluarga melalui swamedikasi yang aman dan cerdas.

Abstract

Human Immuno deficiency Virus / Acquired Immune Deficiency Syndrome (HIV / AIDS) can not be cured but this infection can be controlled with antiretroviral drugs (ARVs). Compliance of people with HIV/AIDS (PLWHA) is very important to cope with HIV/AIDS. Non-compliance with antiretroviral therapy gives the effect of drug resistance so that the drug cannot function or fail. The purpose of this study was to determine the factors that affect the compliance of antiretroviral therapy program in PLWHA in Puskesmas Harapan. The design of this study is cross sectional. The respondents of this study were PLWHA patients undergoing antiretroviral therapy program in infection health centers totaling 25 respondents determined by purposive sampling. The Data was collected with a uestionnaire heet and Pill Count. Analysis of the data using Pearson correlation test, spearman rho and multivariate logistic regression CI ③ = 0.05. The results showed that ARV treatment adherence is related to knowledge (p = 0.017), side effects (p=0.418) no relationship, and support taking medication (p = 0.278) conclusion drug side effects are the dominant factors affecting ARV therapy adherence.
