Abstract

Pinekuhe smoked fish is a fishery product traditionally processed by fishermen of Sangihe Island Regency. Staphylococcus epidermis is an opportunistic phatogen that causes infection to humans of weak immunity. This study aims to identify S. epidermis isolated from the Pinekuhe smoked fish. Fourty isolate samples were grown on Mannitol salt Agar medium and suspect colonies were tested for Gram staining, Catalase, Motility and Coagulase test There were 13 isolates identified as S. epidermis which characteristics as gram-positive, round shapes, clustering, 0,5μm-1μm in diameter, non-motilepositive catalase, and not ferment Mannitol. So, there were 32.5% S. epidermis that contaminated Pinekuhe smoked fish.

Abstract

One of the products processed by smoked fisheries owned by fishermen regency of Sangihe Island is Pinekuhe. This research aims to determine the level of microbial contamination in the smoked fish Pinekuhe processed fishermen regency of Sangihe Islands. Sampling was taken randomly in some of the fish processing units of Pinekuhe smoke. Observation of the deterioration of the quality of fish products Pinekuhe, observed through the test of Total Plate Count (TPC), total mould, and total of Staphylococcus sp. The results showed that the TPC value of all processors meets SNI standards. TPC value of A, B, C, and D processors, respectively, 1,3 x 104 CFU/G, 2,6 x 104 CFU/G, 6,9 x 104, and 1,2 x 104 CFU/g. The total observation of Staphylococcus sp. on processor A and processor B generates a value of 0, while processor C has a total value of 1.1 x 102 TVC/G and processor D has a total value of Staphylococcus sp. 1,2 x 102 TVC/g. The total number of bacteria, the total amount of mould, and the total number of Staphylococcus sp. Still qualify SNI, except Staphylococcus sp. on processor C and processor D exceeds the amount required by the Indonesian National Standard on the limits of contamination microbes in smoked fish.

Abstract

Pinekuhe  is the local name for smoked scad fish (Decapterus sp.), a traditionally processed fish product from Sangihe Islands whose taste, aroma and form are typical and unique. In this research aims of physiological and biochemical characteristics with pathogenicity isolated S. aureus which the isolated from the product smoked scad fish (Decapterus rusellii) Pinekuhe it was produce and prepared by the local fisherman in Sangihe Island Regency. The isolated that have gathered from this researched was 111 product isolate from the smoke fish Pinekuhe which grown from media MSA. Its had isolated already through thetest of physiological comprise, Gram staining and from the test of biochemical e.g., from the test Catalase, Voges-Proskauer, and Fermentation carbohydrate tests (Glucose and Manitol). The characteristics of pathogenicity S. aureus had been done and used by making of Coagulase, Nuklease Thermostabil productionand deoksiribonuklease (DNase). The result of this research showing that had still 111 isolating strains that still ingroup which consists of 108 isolated Staphylococcus and from 108 strains to 68 strains that had been test in identifying, as of Staphylococcus aureus with characteristics of Gram positif coccus, catalase positif,Voges-Proskauer positif, and to Fermentation Glucose and Manitol. The 68 isolate S. aureus that characterize from Phatogen principles product Coagulase test, Nuklease Thermostabil and deoksiribonuklease (DNase). S. aureus is dominant (62%) contaminate smoked fish Pinekuhe processed traditional of Sangihe island.

Abstract

This community partnership empowerment activity empowers mothers of the Family Welfare Development Team (TP. PKK) in Tahuna District, Sangihe Islands Regency, North Sulawesi through education on diversifying processed fish products to prevent stunting. This activity was carried out in September-October 2024 in three villages focused on handling stunting: Soataloara 1, Mahena, and Santiago, Tahuna District. The purpose of this empowerment activity is to empower the TP. PKK in diversifying processed fish products to help prevent stunting in the Sangihe Islands Regency. The methods used include hearings, initial observations, determining priority programs, facilitation and assistance, and monitoring and evaluation. Activities consisted of counseling on fish nutrition and technical guidance on making processed fish products such as meatballs, nuggets, and kaki naga. The evaluation was conducted through pre and post-tests to measure the improvement of participants' knowledge. The results showed a significant increase in the knowledge and skills of TP. PKK mothers. The average knowledge score on fish nutrition increased from 57% to 87%. As for the technical guidance on processed fish products, the skills improvement was seen in the making of meatballs (64% to 90%), nuggets (62% to 86%), and dragon feet (66% to 94%). This community partnership empowerment activity succeeded in increasing the understanding of PKK women in the Tahuna Sub-district about the importance of fish consumption in preventing stunting and improving the skills of TP. PKK mothers in processing fish into products with high nutritional value.

Abstract

Penggunaan media kultur komersial seperti Nutrient Agar (NA) semakin terkendala oleh tingginya biaya, sehingga diperlukan alternatif berbahan alami yang lebih ekonomis namun tetap mampu mendukung pertumbuhan mikroorganisme secara optimal. Penelitian ini bertujuan mengevaluasi efektivitas media agar-agar buatan berbahan kaldu ikan tongkol (Euthynnus affinis) sebagai media kultur alternatif untuk pertumbuhan Staphylococcus sp., serta membandingkannya dengan kinerja NA instan. Penelitian dilakukan melalui rangkaian tahapan pembuatan kaldu ikan tongkol, formulasi media agar, sterilisasi, inokulasi bakteri, dan inkubasi. Evaluasi performa media dilakukan melalui pengamatan pertumbuhan koloni, uji fisiologis (pewarnaan Gram), serta uji biokimia (katalase dan oksidase), termasuk analisis Total Plate Count (TPC). Hasil penelitian menunjukkan bahwa media kaldu ikan tongkol mampu mendukung pertumbuhan koloni Staphylococcus sp. dengan karakteristik morfologi yang serupa bahkan lebih subur dibandingkan Nutrient Agar instan. Uji Gram memperlihatkan hasil konsisten berupa sel Gram-positif berbentuk kokus, sedangkan uji katalase menunjukkan reaksi positif pada kedua jenis media. Uji oksidase memberikan hasil negatif sesuai karakteristik Staphylococcus sp., mengonfirmasi bahwa media alternatif tidak memengaruhi profil biokimia bakteri. Nilai TPC pada media kaldu ikan tongkol juga berada dalam kisaran yang sebanding dengan media standar. Secara keseluruhan, media agar berbahan kaldu ikan tongkol terbukti layak digunakan sebagai alternatif ekonomis untuk kultur Staphylococcus sp., serta berpotensi diterapkan pada laboratorium dengan keterbatasan sumber daya.
