Abstract

Due to the continued presence of and impact of COVID-19 on our modern world, the benefit and necessity of alternative treatment options and effective antiviral compounds is readily apparent. This study seeks to explore the utility of chlorogenic acid as a COVID-19 antiviral. The in-silico experimental study was conducted using a laptop and publicly-available freeware. Key compounds used in this study are the comparative drugs favipiravir (CID 492405), oseltamivir (CID 65028), nafamostat (CID 4413), and spironolactone (CID 5833), to be tested against our subject, chlorogenic acid (CID 1794427). Compunds used in this study were docked with specific target proteins, such as ACE2, TMPRSS2, RdRp, dan 3Clpro, using AutoDock Vina. The visualization of molecular interactions was performed with Discovery Studio v21. Our results showed that chlorogenic acid has one Lipinski’s Rule of Five violation in H-bond Donor parameter. Chlorogenic acid had a higher affinity towards ACE2 and 3CLpro and lower affinity towards TMPRSS2 and RdRp, if compared to competing drugs, with a binding energy of -7.8, -6.9, -7.0, and -7.4 kcal/mol respectively, for ACE2, TMPRSS2, RdRp, and 3CLpro. We conclude that chlorogenic acid shows promise as oral drug candidate based on these results, and that further studies into the realm of molecular dynamics, in-vitro, or in-vivo studies are warranted in order to explore this underutalized compound and possibly bring it to human trials.

Abstract

Diabetic ulcers represent a prevalent complication among patients with poorly controlled diabetes mellitus, often resulting from inadequate blood glucose regulation, neuropathy, peripheral artery disease, and suboptimal wound care. This systematic review aims to evaluate the therapeutic efficacy of granulocyte-macrophage colony-stimulating factor (GM-CSF) for diabetic ulcers. The review utilized databases such as PubMed, ProQuest, and Google Scholar, with only six articles meeting the eligibility criteria for analysis. The findings suggest that GM-CSF exhibits therapeutic potential by enhancing neovascularization, promoting keratinocyte proliferation, facilitating granulation tissue formation, and reducing wound diameter. These results highlight the potential of GM-CSF as a therapeutic agent for diabetic ulcers. Nevertheless, the current body of research is limited, necessitating further studies with larger populations and diverse GM-CSF administration methods to substantiate these findings.

Abstract

Daun Gaharu (Aquilaria malaccensis) sejak lama telah digunakan masyarakat Kabupaten Bangka Tengah untuk menangkal keletihan akibat gangguan tidur ringan. Minuman yang diramu dari daun Gaharu yang diiris-iris lalu dijemur sebentar kemudian disedu air hangat kuku ternyata berkhasiat untuk menimbulkan rasa kantuk dan setelah bangun dari terlelap sejenak maka tubuh akan menjadi lebih segar. Penelitian ini ingin membuktikan manfaat tersebut di atas. Sejumlah 60 orang sukarelawan sehat terdiri dari 30 orang laki-laki dan 30 orang wanita dengan rentang umur 18 – 60 tahun berpartisipasi untuk membuktikan efek sedativa dan pengaruh kesehatan lainnya. Dengan disain pre-post test serta kerelaan sukarelawan sehat ini (dengan mengisi informed consent) sebagai responden, maka semuanya diberi minum teh celup Gaharu yang berisi 1 g simplisia kering/saset setiap hari selama 3 minggu berturut-turut. Setelah selesai satu sesi minum, semuanya diwawancarai pemeriksaan fisik, mental dengan menggunakan kuesioner model Pittsburgh Insomnia Rating Scale (PIRS) serta dikoleksi darah intravenanya. Hasil menunjukkan bahwa 32 % responden timbul rasa kantuk ringan, 10 % merasa lebih segar walaupun terasa sakit kepala ringan, serta berkurangnya rasa pegal dan nyeri sendi namun nafsu makan bertambah. Profil darah rutin tidak berubah namun kadar hormon seks seperti testosteron, progesteron, estradiol dan FSH meningkat ringan tanpa LH serta penurunan fibrinogen dan hs-CRP. Sedangkan transaminase hepar tidak berubah. Dari temuan ini disimpulkan bahwa konsumsi rutin teh gaharu memperbaiki profil tidur dan kesegaran jasmani namun tidak bersifat hepatotoksik. Perubahan kadar hormon seks, fibrinogen dan hs-CRP perlu dikaji lebih lanjut.
