Abstract

Testis is the male reproductive organ that performs both endocrine and exocrine functions. This study aimed to determine the distribution of carbohydrates in the testis of sumba ongole cattle. The sample consisted of six testis were collected from East Sumba Slaughter House, fixed in formalin 10 %, processed histologically, and continued with AB pH 2,5 - PAS staining. The result showed a weak alcian blue intensity (+) in the tunica albuginea and moderate intensity (++) in the interstitial connective tissue, basement membrane, and connective tissue in the lumen of the seminiferous tubules. A strong intensity (+++) to PAS staining was seen in spermatogonia, spermatocytes, and myoid cells. Spermatids, spermatozoa, and sertoli cells showed a negative reaction (-). In the intertubular area, Leydig cells and fibroblasts showed a strong PAS reaction (+++), connective tissue showed a moderate reaction (++). The intensity of the weak reaction (+) is indicated by the basement membrane of the seminiferous tubules. The variation in the distribution of acidic and neutral carbohydrates in the testis of sumba ongole cattle is correlated with the proliferation of different cell types and steroidogenic activity at various stages of testicular development.

Abstract

The aims of this study is to determine the quality of fresh cement of Landrace pigs diluted with natural diluent of green coconut water. The research was conducted at the Veterinary Reproduction Laboratory of FKKH Undana. Fresh semen of Landrace pigs was collected from male pigs that less than 3 years sexually mature and in healthy condition, then the semen quality was examined by macroscopic and microscopic evaluation. Only cement met the requirements, the proportion of spermatozoa motility not less than 70%, spermatozoa concentration not less than 200 million cells/ml and spermatozoa abnormality not more than 20%. After that, the semen was diluted by adding green coconut water and antibiotics according to the dilution formula. The results showed that semen diluted with natural green coconut was better and could be used as an alternative diluent with a higher of spermatozoa viability and motility (48.2% & 41.25%) compared to fresh semen with a shelf life of 20 hours.

Abstract

Artificial insemination with liquid semen in East Nusa Tenggara is still very rare, the Artificial insemination program is carried out using fresh semen. Liquid semen is generally stored at a temperature of 16-18 °C and lasts only a few days depending on the diluent used, for longer storage it requires the addition of antioxidants that can suppress the dangers of ROS. Therefore, this study was conducted to test the filtrate of red dragon (Hylocereus polyrhizus) in life semen diluent, which is expected to maintain the quality of spermatozoa in fresh semen. Semen was collected from 5 boars using the Glove Hand Method with Dummy Sow. Fresh semen that meets the SNI is diluted with the conditions of spermatozoa motility >70%, spermatozoa concentration >200,106 cells/ml and < abnormalities of 20%. The research was carried out in several stages: 1) examination of fresh semen 2) preparation of diluent 3) preparation of red dragon filtrate (Hylocereus polyrhizus) 4) manufacture of liquid semen. The data were analyzed descriptively and in a Complete Randomized Design with 5 treatments and 4 replicates. If there is a real difference, it can be continued with the Duncan test. Consecutive treatments: KO (fresh semen control); K1( Fresh semen control + semen life® ); P1 (fresh semen + semen life® + red dragon filtrate (Hylocereus polyhizu) 100μL); P2 (fresh semen + semen life® + red dragon filtrate (Hylocereus polyhizu) 200μL); P3 (fresh semen + semen life® + red dragon filtrate (Hylocereus polyhizu) 300μL); P4 (fresh semen + semen life® + red dragon filtrate (Hylocereus polyhizu) 400μL); P5 (fresh semen + semen life® + red dragon filtrate (Hylocereus polyhizu) 500μL). The results of the study can be concluded that the addition of red dragon phytrate (Hylocereus polyrhizus) 300μL in semen life® diluent has good effectiveness in maintaining the quality of liquid semen in storage at a temperature of 18-20 °C.
